    Epidemiological and virological characteristics of pandemic influenza A (H1N1) 2009 in school outbreaks in China

    Background: During the 2009 pandemic influenza H1N1 (2009) virus (pH1N1) outbreak, school students were at an increased risk of infection by the pH1N1 virus. However, the estimation of the attack rate showed significant variability. Methods: Two school outbreaks were investigated in this study. A questionnaire was designed to collect information by interview. Throat samples were collected from all the subjects in this study 6 times and sero samples 3 times to confirm the infection and to determine viral shedding. Data analysis was performed using the software STATA 9.0. Findings: The attack rate of the pH1N1 outbreak was 58.3% for the primary school, and 52.9% for the middle school. The asymptomatic infection rates of the two schools were 35.8% and 37.6% respectively. Peak virus shedding occurred on the day of ARI symptoms onset, followed by a steady decrease over subsequent days (p = 0.026). No difference was found either in viral shedding or HI titer between the symptomatic and the asymptomatic infectious groups. Conclusions: School children were found to be at a high risk of infection by the novel virus. This may be because of a heightened risk of transmission owing to increased mixing at boarding school, or a lack of immunity owing to socioeconomic status. We conclude that asymptomatically infectious cases may play an important role in transmission of the pH1N1 virus

    Phylogenetic distribution and predominant genotype of the avian infectious bronchitis virus in China during 2008-2009

    <p>Abstract</p> <p>Background</p> <p>The nephropathogenic avian infectious bronchitis (IB) caused unprecedented economic losses to the commercial chicken industry of China in 2008-2009. To investigate the prevalence of nephropathogenic IB in China, eighty IBV isolates from different provinces during 2008-2009 were identified by dwarf embryo test and RT-PCR.</p> <p>Results</p> <p>The strains were mostly isolated in winter and spring with a wide age range of IB outbreaks, from 4 to 69 days. By the virus recovery trials, 70/80 of the strains resulted in the deaths or distresses of birds from nephritis. To learn more about the molecular evolutionary characteristics of the circulating field strains, the coding region of major spike 1 (S1) protein gene of these strains was RT-PCR amplified and sequenced. Compared to the published representative strains, nucleotides and amino acids sequence analysis indicated that the S1 genes of these strains and the reference strains displayed homologies ranging from 75.1% to 99.8% and from 73.1% to 99.8% respectively. S1 protein of the major pandemic strains contained 540 or 542 amino acids with the cleavage site of HRRRR or RRFRR. Phylogenetic analysis revealed that recent field isolates of IBV in China were mostly belonged to A2-branch (QXIBV-branch) and HN08-branch, only one isolate was belonged to Gray-branch and M41-branch respectively. Most of the 80 strains showed evolutionarily distant from vaccine strains.</p> <p>Conclusions</p> <p>The results of this study suggested that nephropathogenic IBVs were mainly A2-like strains in China during 2008-2009.</p

    Silky-feather has been selected and fixed in some breeds due to its unique appearance. This phenotype is caused by a single recessive gene (hookless, h). Here we map the silky-feather locus to chromosome 3 by linkage analysis and subsequently fine-map it to an 18.9 kb interval using the identical by descent (IBD) method. Further analysis reveals that a C to G transversion located upstream of the prenyl (decaprenyl) diphosphate synthase, subunit 2 (PDSS2) gene is causing silky-feather. All silky-feather birds are homozygous for the G allele. The silky-feather mutation significantly decreases the expression of PDSS2 during feather development in vivo. Consistent with the regulatory effect, the C to G transversion is shown to remarkably reduce PDSS2 promoter activity in vitro. We report a new example of feather structure variation associated with a spontaneous mutation and provide new insight into the PDSS2 function

This study aims to determine the effect ofcompetency, career development and training on employee performance of the DirectorateGeneral of Fiscal Balance, Ministry of Finance. The method used is descriptive quantitative.The data used are primary data collected by questionnaire and also secondary data. Thepopulation in this study were all employees as many as 387 people. The samples are taken byrandom sampling technique, using the Slovin‟s formula to take 80 people for samples. Thevariables in this study consisted of independent variables that are competency, careerdevelopment and training, while the dependent variable is the performance of employees. Thevariable measured with Likert Scale and hypothesis testing using multiple linear regression byusing t test and F test.Results of this study concluded that: 1) in partially competency hadpositive and significant impact on employee performance; 2) in partially career developmentprovide uneffect on employee performance; 3) in partially training had positive andsignificant impact on employee performance; and 4) in simultaneously competency, careerdevelopment and training have positive and significant impact on employee performance.Competency is the most influence variable on employee performance

    An Analysis of Culling Patterns during the Breeding Cycle and Lifetime Production from the Aspect of Culling Reasons for Gilts and Sows in Southwest China

    To investigate culling patterns during the breeding cycle and lifetime production associated with culling reasons, 19,471 culling records were collected in southwest China. Lifetime pigs born alive (LPBA) and parity for culling reasons, and reason distribution at different parities and breeding cycle were analyzed. Sows culled for stress and death (SD), lameness (LA), common disease (CD), not being pregnant, return to estrus, and abortion (NP) had fewer than 20 LPBA (p &lt; 0.05). Gilts were mainly culled for anestrus beyond nine months (AB9), CD, and LA, while weaned sows were culled for reproductive system disease (RS), CD, and anestrus beyond seven days (p &lt; 0.0033). Gestating sows were mainly culled for NP, CD, and SD, while lactating sows were mainly culled for low or no milk production (NM), poor litter size, and CD (p &lt; 0.0033). Moreover, sows were mainly culled at parity 0, 1, and 2 (p &lt; 0.0024). Besides CD and RS, LA and NP were the primary reasons for parity 1 and 2 culls, respectively. In conclusion, SD, LA, CD, and NP sharply decrease sow lifetime production. AB9, RS, NP, and NM mainly occurred in gilts, weaned, gestating, and lactating sows, respectively. Low parity sows had a higher risk of CD, RS, LA, and NP

    Genomic selection for the improvement of antibody response to Newcastle disease and avian influenza virus in chickens.

    Newcastle disease (ND) and avian influenza (AI) are the most feared diseases in the poultry industry worldwide. They can cause flock mortality up to 100%, resulting in a catastrophic economic loss. This is the first study to investigate the feasibility of genomic selection for antibody response to Newcastle disease virus (Ab-NDV) and antibody response to Avian Influenza virus (Ab-AIV) in chickens. The data were collected from a crossbred population. Breeding values for Ab-NDV and Ab-AIV were estimated using a pedigree-based best linear unbiased prediction model (BLUP) and a genomic best linear unbiased prediction model (GBLUP). Single-trait and multiple-trait analyses were implemented. According to the analysis using the pedigree-based model, the heritability for Ab-NDV estimated from the single-trait and multiple-trait models was 0.478 and 0.487, respectively. The heritability for Ab-AIV estimated from the two models was 0.301 and 0.291, respectively. The estimated genetic correlation between the two traits was 0.438. A four-fold cross-validation was used to assess the accuracy of the estimated breeding values (EBV) in the two validation scenarios. In the family sample scenario each half-sib family is randomly allocated to one of four subsets and in the random sample scenario the individuals are randomly divided into four subsets. In the family sample scenario, compared with the pedigree-based model, the accuracy of the genomic prediction increased from 0.086 to 0.237 for Ab-NDV and from 0.080 to 0.347 for Ab-AIV. In the random sample scenario, the accuracy was improved from 0.389 to 0.427 for Ab-NDV and from 0.281 to 0.367 for Ab-AIV. The multiple-trait GBLUP model led to a slightly higher accuracy of genomic prediction for both traits. These results indicate that genomic selection for antibody response to ND and AI in chickens is promising

    Avian leukosis is a neoplastic disease caused in part by subgroup J avian leukosis virus J (ALV-J). Micro ribonucleic acids (miRNAs) play pivotal oncogenic and tumour-suppressor roles in tumour development and progression. However, little is known about the potential role of miRNAs in avian leukosis tumours. We have found a novel tumour-suppressor miRNA, gga-miR-375, associated with avian leukosis tumorigenesis by miRNA microarray in a previous report. We have also previously studied the biological function of gga-miR-375; Overexpression of gga-miR-375 significantly inhibited DF-1 cell proliferation, and significantly reduced the expression of yes-associated protein 1 (YAP1) by repressing the activity of a luciferase reporter carrying the 3'-untranslated region of YAP1. This indicates that gga-miR-375 is frequently downregulated in avian leukosis by inhibiting cell proliferation through YAP1 oncogene targeting. Overexpression of gga-miR-375 markedly promoted serum starvation induced apoptosis, and there may be the reason why the tumour cycle is so long in the infected chickens. In vivo assays, gga-miR-375 was significantly downregulated in chicken livers 20 days after infection with ALV-J, and YAP1 was significantly upregulated 20 days after ALV-J infection (P<0.05). We also found that expression of cyclin E, an important regulator of cell cycle progression, was significantly upregulated (P<0.05). Drosophila inhibitor of apoptosis protein 1 (DIAP1), which is related to caspase-dependent apoptosis, was also significantly upregulated after infection. Our data suggests that gga-miR-375 may function as a tumour suppressor thereby regulating cancer cell proliferation and it plays a key role in avian leukosis tumorigenesis
