Cancer treatment by radiotherapy and chemotherapy, and chemotherapy treatments is more mostly used all over the world. Generally, chemotherapy drugs cusses the damage by free radicals and produce cytotoxicity. Natural antioxidant compounds of daily diet are more effective to scavenge or neutralize the free radicals which create by chemotherapy drugs and finally, improvement histopathological change

    Evaluation of Isolated Compounds Activity from Convolvulus Arvensis Against Algae

    Abstract This study includes isolation, purification, and identification of algae from different aquatic environments in Baghdad city. Nine unialgal cultures were obtained. These algal cultures included 6 species of blue-green algae (Microcystis aeruginosa, Microcystis flos-aquae, Oscillatoria limnetica, Nostoc carneum, Westillopesis prolifica, Mastigocluds lamiosus), and 3 species of green algae (Mougeotia sclaris, Scenedesmus dimorphus and Chlorella vulgaris). In addition that aerial parts from Convolvulus arvensis were collected. Terpens, alkaloids and phenols were extracted of mentioned plant, and the antialgal activity of extracts types were evaluated in 3 concentrations (5, 10, and 20 mg/ml) by wells and diffused in the agar media. Results showed that phenols extracts from C. arvensis were the most effective against isolated algae than other extracts, and terpens extracts were less effective. O. limentica was the most sensitive to attack by extracts than other algae, while N. carneum was lower sensitive, also W. prolifica, M. lamiosus were the most resistance to attack by C. arvensis extracts. Chemical composition of phenols and alkaloids were analyzed by High Performance Liquid Chromatography (HPLC) and showed that present 10 phenolic compounds, 8 alkaloid compounds in C. arvensis.     Assessment of the Genotoxicity, Antigenotoxicity on Alloxan-Induce Diabetic in Mice by Micronuclus(MN) Assay

    Fenugreek (Trigonellafoenum-graecum ) plant has become interest subject because of its beneficial effects on human health, good nutritive value in addition it has antigenotoxic  effects therefore, the purpose of the present study was to investigate antigenotoxic  effects of alcoholic extract of Fenugreek seed at 200 mg /kg b.w  on mice bone marrow which treatmed with alloxan, 6-mercaptopurine anticancer at 200 mg /kg b.w by using micronuclus test at 5 day for all treatments. Cytogenetic study showed significant increasing at (p< 0.05) in Micronuclus number  MN in diabetic mice, anticancergroup or both when compared to the negative control. After 5day of alloxan treated mice with marked hyperglycemia (blood glucose),results recorded significant increase at (p< 0.05) in diabetic group ( group treated with alloxan) than anticancer in same time compared with control. Finally, alcoholic extracts of  Fenugreek seeds found significant reduced MN, blood glucose in all treated groups. These results indicate that fenugreek plant were one of the primary supplements used to support type I diabetics or insulin-dependent diabetes mellitus (NIDDM), in addition  it has active compounds which have important role on antigenotoxic, hyperglycemia  activity.     Free radical may be responsible of several pathophysiology disease for threatening human life, and they are produced from endogenous and exogenous sources. Using the balanced amount  of nutritious diet lead to a good health, may be neutralizing or scavenging  free radicals by antioxidants compounds. daily diet have large number of vitamin A, E and C, carotenoids, polyphenols, etc. as natural antioxidants, the main bases of them are fruits, cereals, vegetables, and beverages. Enzymatic and non-enzymatic antioxidant substances are two different kinds that lessen the reactions of free radicals. In order to protect itself from reactive oxygen species, the human body uses an enzyme antioxidant. The two kinds of non-enzymatic antioxidants are natural antioxidants and synthetic antioxidants. The aim of this review is to knowledge a reasons which causes the free radical and balance them by natural antioxidant constituents, and replacement synthetic antioxidant by natural antioxidant, due to daily diet have large amounts from natural antioxidants, and natural antioxidants more effective than synthetic antioxidants, in additional they occurrence in nature and more inexpensive

    Immunodetectable cyclin D 1 is associated with oestrogen receptor but not Ki67 in normal, cancerous and precancerous breast lesions

    Cyclin D1 is associated with cell cycle regulation and has more recently been shown to stimulate the transcriptional functions of the oestrogen receptor (ER). Furthermore, in normal breast there is a negative association between expression of ER and the proliferation marker Ki67 indicating that either ER positive cells are non-dividing or that the receptor is down-regulated as cells enter cycle. This important relationship breaks down in many ER-positive cancers and precancerous breast lesions where the receptor is often detected on proliferating cells. The aims of the present study were to determine the interplay between ER, Ki67 and cyclin D 1 in individual cells within the spectrum of human breast lesions ranging from normal to invasive carcinoma by using dual staining immunofluorescence. We found that in normal breast there was a strong positive association between ER and cyclin D 1 expression. In contrast there was a strong negative association between cyclin D 1 and Ki67 expression. Similar findings were seen for the other precancerous and cancerous breast lesions. Thus immunodetectable cyclin D 1 within individual cells does not appear to be associated with cell cycle progression in the benign or malignant breast but instead may have important interactions with ER. © 2001 Cancer Research Campaign http://www.bjcancer.co

    Compliance with guidelines is related to better local recurrence-free survival in ductal carcinoma in situ

    The aim was to study the effect of compliance with guidelines on local recurrence (LR)-free survival in patients treated for ductal carcinoma in situ (DCIS). From January 1992 to December 2003, 251 consecutive patients had been treated for DCIS in two hospitals in the North Netherlands. Every case in this two-hospital sample was reviewed in retrospect for its clinical and pathological parameters. It was determined whether treatment had been carried out according to clinical guidelines, and outcomes in follow-up were assessed. In addition, all patients treated for DCIS in this region (n=1389) were studied regarding clinical parameters, in order to determine whether the two-hospital sample was representative of the entire region. In the two-hospital sample, 31.4% (n=79) of the patients had not been treated according to the guidelines. Positive margins were associated with LR (hazard ratio (HR)=4.790, 95% confidence interval (CI) 1.696–13.531). Breast-conserving surgery and deviation from the guidelines were independent predictors of LR (HR=7.842, 95% CI 2.126–28.926; HR=2.778, 95% CI 0.982–6.781, respectively). Although the guidelines changed over time, time was not a significant factor in predicting LRs (HR=1.254, 95% CI 0.272–5.776 for time period 1992–1995 and HR=1.976, 95% CI 0.526–7.421 for time period 1996–1999). Clinical guidelines for the treatment of patients with DCIS have been developed and updated from existing literature and best evidence. Compliance with the guidelines was an independent predictor of disease-free survival. These findings support the application of guidelines in the treatment of DCIS

    Keratin 6 is not essential for mammary gland development

    INTRODUCTION: Keratin 6 (K6) has previously been identified as a marker of early mammary gland development and has also been proposed to be a marker of mammary gland progenitor cells. However, the function of K6 in the mammary gland was not known, so we examined the expression pattern of the protein during both embryonic and postnatal mammary development, as well as the mammary gland phenotype of mice that were null for both K6a and K6b isoforms. METHOD: Immunostaining was performed to determine the expression pattern of K6a throughout mammary gland development, from the embryonic mammary bud to lactation. Double immunofluorescence was used to co-localize K6 with known markers of mammary gland development. Wild-type and K6ab-null mammary tissues were transplanted into the cleared fat pads of nude mice and the outgrowths were analyzed for morphology by whole-mount staining and for markers of mammary epithelium by immunostaining. Finally, progesterone receptor (PR) and bromodeoxyuridine co-localization was quantified by double immunofluorescence in wild-type and K6ab-null mammary outgrowths. RESULTS: Here we report that K6 is expressed earlier than described previously, by embryonic day 16.5. K6a is the predominant isoform expressed in the mammary gland, localized in the body cells and luminal epithelial cells but not in the cap cells or myoepithelial cells. Co-localization studies showed that most K6a-positive cells express steroid receptors but do not proliferate. When both the K6a and K6b genes are deleted, mammary gland development appears normal, with similar expression of most molecular markers examined in both the pubertal gland and the mature gland. Loss of K6a and K6b, however, leads to an increase in the number of steroid-receptor-positive cells, and increased co-localization of steroid receptor expression and proliferation was observed. CONCLUSION: Although K6a was not essential for mammary gland development, loss of both K6a and K6b resulted in an increase in PR-positive mammary epithelial cells and decreased proliferation after exposure to steroid hormones. There was also increased co-localization of PR and bromodeoxyuridine, suggesting alterations in patterning events important for normal lobuloalveolar development

    DAX-1 expression in human breast cancer: comparison with estrogen receptors ER-α, ER-β and androgen receptor status

    BACKGROUND: So far there have been no reports on the expression pattern of DAX-1 (dosage-sensitive sex reversal, adrenal hypoplasia critical region, on chromosome X, gene 1) in human breast cells and its relationship to the estrogen receptors, ER-α and ER-β, and the androgen receptor (AR). METHODS: In this study we evaluated, by immunohistochemistry and Western blot analysis, the presence and distribution of DAX-1 in benign breast disease (BBD), in situ carcinoma (CIS), and ductal and lobular breast carcinomas. RESULTS: In BBD and breast carcinomas, DAX-1 was present in both the nuclei and the cytoplasm of epithelial cells, although in infiltrative carcinomas the percentage of nuclear immunoreaction was higher than in CIS. An important relation was observed between DAX-1 and AR expression and between this orphan receptor and nodal status. CONCLUSION: DAX-1 might modify the AR and ER-β intracellular location, and because a direct positive relation between the expression of these three receptors was found it could be assumed that the presence of DAX-1 in neoplastic cells might indicate a possible failure of endocrine therapies
