    Crack Growth Rate Model under Constant Cyclic Loading and Effect of Different Singularity Fields

    AbstractThe effect of two singularity fields: the Hutchinson-Rice-Rosengren (HRR) fields and the Rice-Kujawski-Ellyin (RKE) fields, is investigated in the present study to discuss a former fatigue crack growth (FCG) model based on energy balance during growth of the crack. A parameter which can demonstrates the effect of different types of singularity fields is included in the crack propagation model which can predict stage-II FCG behavior independently from the basic low cycle fatigue properties. Good agreement between experimental and theoretical results is obtained

    Generation of Inducible CRISPRi and CRISPRa Human Stromal/Stem Cell Lines for Controlled Target Gene Transcription during Lineage Differentiation

    Background. Human bone marrow stromal/stem cells (hMSCs, also known as the skeletal stem cells or mesenchymal stem cells) are being employed to study lineage fate determination to osteoblasts, adipocytes, and chondrocytes. However, mechanistic studies employing hMSC have been hampered by the difficulty of deriving genetically modified cell lines due to the low and unstable transfection efficiency. Methods. We infected hMSC with a CRISPR/Cas9 lentivirus system, with specific inducible dCas9-coupled transcription activator or repressor: dCas9-KRAB or dCas9-VP64, respectively, and established two hMSC lines (hMSC-CRISPRi and hMSC-CRISPRa) that can inhibit or activate gene expression, respectively. The two cell lines showed similar cell morphology, cell growth kinetics, and similar lineage differentiation potentials as the parental hMSC line. The expression of KRAB-dCas9 or VP64-dCas9 was controlled by the presence or absence of doxycycline (Dox) in the cell culturing medium. To demonstrate the functionality of the dCas9-effector hMSC system, we tested controlled expression of alkaline phosphatase (ALP) gene through transfection with the same single ALP sgRNA. Results. In the presence of Dox, the expression of ALP showed 60-90% inhibition in hMSC-CRISPRi while ALP showed more than 20-fold increased expression in hMSC-CRISPRa. As expected, the ALP was functionally active and the cells showed evidence for inhibition or enhancement of in vitro osteoblast differentiation, respectively. Conclusion. hMSC-CRISPRi and hMSC-CRISPRa are useful resources to study genes and genetic pathways regulating lineage-specific differentiation of hMSC

    Actin depolymerization enhances adipogenic differentiation in human stromal stem cells

    Human stromal stem cells (hMSCs) differentiate into adipocytes that play a role in skeletal tissue homeostasis and whole body energy metabolism. During adipocyte differentiation, hMSCs exhibit significant changes in cell morphology suggesting changes in cytoskeletal organization. Here, we examined the effect of direct modulation of actin microfilament dynamics on adipocyte differentiation. Stabilizing actin filaments in hMSCs by siRNA-mediated knock down of the two main actin depolymerizing factors (ADFs): Cofilin 1 (CFL1) and Destrin (DSTN) or treating the cells by Phalloidin reduced adipocyte differentiation as evidenced by decreased number of mature adipocytes and decreased adipocyte specific gene expression (ADIPOQ, LPL, PPARG, FABP4). In contrast, disruption of actin cytoskeleton by Cytochalasin D enhanced adipocyte differentiation. Follow up studies revealed that the effects of CFL1 on adipocyte differentiation depended on the activity of LIM domain kinase 1 (LIMK1) which is the major upstream kinase of CFL1. Inhibiting LIMK by its specific chemical inhibitor LIMKi inhibited the phosphorylation of CFL1 and actin polymerization, and enhanced the adipocyte differentiation. Moreover, treating hMSCs by Cytochalasin D inhibited ERK and Smad2 signaling and this was associated with enhanced adipocyte differentiation. On the other hand, Phalloidin enhanced ERK and Smad2 signaling, but inhibited adipocyte differentiation which was rescued by ERK specific chemical inhibitor U0126. Our data provide a link between restructuring of hMSCs cytoskeleton and hMSCs lineage commitment and differentiation.     Stressing state characteristics of reinforcement concrete box-girders strengthened with carbon fiber reinforced plastic

    This paper investigates structural performance of five reinforcement concrete (RC) box-girders under a combination loading of bending, shear and torsion, applying the structural stressing state theory. The measured strain data is modeled as generalized strain energy density (GSED) to characterize the structural stressing state mode. Then the Mann-Kendall (M-K) criterion is innovatively applied to detect the leap characteristics of RC box-girders’ stressing state from the E’-T curves, deriving the new definition of structural failure load. Furthermore, the reinforcement effects of different Carbon Fiber Reinforced Plastic (CFRP) wrapping schemes on the behaviors of experimental RC box-girders are revealed through comparing strain modes of stirrup and longitudinal reinforcement. Finally, the method of numerical shape function is applied to reasonably expand the limited strain data for further exploring the strain distribution of cross section and analyzing the stressing state characteristics of the RC box-girders. The research results provide a new angle of view to conduct structural analysis and a reference to the improvement of reinforcement scheme. First published online 29 November 201

    An investigation into working behavior characteristics of parabolic CFST arches applying structural stressing state theory

    This paper conducts the experimental and simulative analysis of stressing state characteristics for parabolic concretefilled steel tubular (CFST) arches undergoing vertical loads. The measured stain data is firstly modeled as the generalized strain energy density (GSED) to describe structural stressing state mode. Then, the normalized GSED sum Ej,norm at each load Fj derives the Ej,norm-Fj curve reflecting the stressing state characteristics of CFST arches. Furthermore, the Mann-Kendall criterion is adopted to detect the stressing state change of the CFST arch during its load-bearing process, leading to the revelation of a vital stressing state leap characteristic according to the natural law from quantitative change to qualitative change of a system. The revealed qualitative leap characteristic updates the existing definition of the CFST arch’s failure load. Finally, the accurate formula is derived to predict the failure/ultimate loads of CFST arches. Besides, a method of numerical shape function is proposed to expand the limited strain data for further analysis of the stressing state submodes. The GSED-based analysis of structural stressing state opens a new way to recognize the unseen working behavior characteristics of arch structures and the updated failure load could contribute to the improvement on the structural design codes

    Effect of lateral stiffness of secondary suspensions on heavy-haul locomotives stability during braking based on simulation and experiment

    This paper aimed to investigate the effect of the lateral stiffness of secondary suspensions on the stability capacity and running safety of heavy-haul locomotives during braking based on the dynamic model and the field braking tests. The dynamic model of heavy-haul locomotives included two double-unit locomotives and five coupler systems. Simulation results indicate that the increasing of the lateral stiffness of secondary suspensions can improve the stability capacity and running safety of heavy-haul locomotives. Then, the field braking experiments were conducted to validate the dynamic model. Comparing the experiment results of different locomotives, the coupler and carbody yaw angles are respectively decreased by 31.8 and 29.5%, which is consistent with the simulation results. It is worthy to be noted that lateral vibration behaviour of the carbody increases with the increasing of the lateral stiffness of secondary suspensions. For the improved locomotive, the main frequency of lateral acceleration is 1…2 Hz. However, the main frequency of lateral acceleration is 0.5…1 Hz in the original locomotive tests. Moreover, the high-frequency vibration is increased, especially in 10…12.5 Hz. According to the simulation and experiment results, the reasonable lateral stiffness of secondary suspensions is 400 kN/m for the test locomotive

    Evaluation and Empirical Research on Eco-Efficiency of Financial Industry Based on Carbon Footprint in China

    Since finance is the core of economic development, the green development of the financial industry is an essential driving force not only for achieving the dual “carbon” goal of China but also for economic and social sustainable development. An accurate understanding of the ecological efficiency of the financial industry is of great importance for guiding sustainable economic development. In this paper, we first calculate the carbon footprint of China’s financial industry in 2012 and 2017 based on the life cycle theory and the input–output analysis method. Second, we analyze the primary sources and final flows of the carbon footprint of the financial industry in each province from the perspectives of the industrial chain and final demand. Finally, we estimate the ecological efficiency, emission reduction, and value-added potential of the financial industry by using the radially adjusted slack variable DEA model (SRAM-DEA) under two assumptions, natural disposability and managerial disposability. The results show that (1) the ecological efficiency of the financial industry in most provinces is low, and the regional differences are significant; (2) the overall ecological efficiency of the financial industry in 2017 was better than that in 2012; (3) technological innovation of financial products and the upgrading of capital supervision play an essential role in promoting the improvement of ecological efficiency. Especially, under managerial disposability, the ecological efficiency of the financial industry in each province has a greater potential for emission reduction and added value
