IR Bismuth active centers in optical fibers: Combined excitation-emission spectroscopy

3D excitation-emission luminescence spectra of Bi-doped optical fibers of various compositions were measured in a wide wavelength range 450-1700 nm. Such luminescence spectra were obtained for Bi-doped pure silica and germania fibers, and for Bi-doped Al- or P-codoped silica fibers (at room and liquid nitrogen temperatures). The energy level schemes of IR bismuth active centers in pure silica and germania core fibers were derived from spectra obtained. The energy level schemes similarity of bismuth active centers in these two types of fibers was revealed.Comment: 12pages, 7 figures, 5 table

Optimization of thermal design and geometrical parameters of a flat tube-fin adsorbent bed for automobile air-conditioning

Adsorbent bed design and performance strongly affect the overall performance of adsorption systems. In the present study, an analytical model was developed to determine the optimum geometrical and thermal parameters of a flat tube-fin adsorbent bed to reach the maximum system performance. This types of heat exchangers offer substantial reduced in weight, cost, volume and thermal conductivity, which can make them a good choice for adsorbent beds in automobile applications. Results showed that the overall thermal conductance of the bed and the maximum practical specific cooling capacity increased when reducing in flat tube thickness and fin pitch as well as by increasing in fin thickness and water channel wall thickness. The specific thermal conductance increased by 2.5% when reducing the channel pitch from its design value to a minimum permissible (0.004 m). From thermal parameters that have been studied, the adsorbent thermal conductivity is the most sensitive parameter to the specific thermal conductance in beds. The system performance also significantly enhanced by reducing the mass of the metal bed and the heat transfer fluids as well as the desorption heat of the selecting working pair. Bright ligand-activatable fluorescent protein for high-quality multicolor live-cell super-resolution microscopy

    Get PDF
We introduce UnaG as a green-to-dark photoswitching fluorescent protein capable of high-quality super-resolution imaging with photon numbers equivalent to the brightest photoswitchable red protein. UnaG only fluoresces upon binding of a fluorogenic metabolite, bilirubin, enabling UV-free reversible photoswitching with easily controllable kinetics and low background under Epi illumination. The on- and off-switching rates are controlled by the concentration of the ligand and the excitation light intensity, respectively, where the dissolved oxygen also promotes the off-switching. The photo-oxidation reaction mechanism of bilirubin in UnaG suggests that the lack of ligand-protein covalent bond allows the oxidized ligand to detach from the protein, emptying the binding cavity for rebinding to a fresh ligand molecule. We demonstrate super-resolution single-molecule localization imaging of various subcellular structures genetically encoded with UnaG, which enables facile labeling and simultaneous multicolor imaging of live cells. UnaG has the promise of becoming a default protein for high-performance super-resolution imaging. Photoconvertible proteins occupy two color channels thereby limiting multicolour localisation microscopy applications. Here the authors present UnaG, a new green-to-dark photoswitching fluorescent protein for super-resolution imaging, whose activation is based on a noncovalent binding with bilirubin

Age-related changes can significantly alter the state of adaptive immune system and often lead to attenuated response to novel pathogens and vaccination. In present study we employed 5′RACE UMI-based full length and nearly error-free immunoglobulin profiling to compare plasma cell antibody repertoires in young (19–26 years) and middle-age (45–58 years) individuals vaccinated with a live yellow fever vaccine, modeling a newly encountered pathogen. Our analysis has revealed age-related differences in the responding antibody repertoire ranging from distinct IGH CDR3 repertoire properties to differences in somatic hypermutation intensity and efficiency and antibody lineage tree structure. Overall, our findings suggest that younger individuals respond with a more diverse antibody repertoire and employ a more efficient somatic hypermutation process than elder individuals in response to a newly encountered pathogen
