    This study was supported by the Bureau of Land Management through interagency agreement with the National Oceanic and Atmospheric Administration under which a multi-year program responding to need of petroleum development of the Alaskan continental shelf is managed by the Outer Continental Shelf Environmental Assessment Program (OCSEAP) Office. Drawings were done by Debbie Cocci a and Jim Burton. The idea of this project arose during a conversation between George Edwardson, Jr., and one of the Principal Investigators. Rachael Craig and Edna MacLean helped in initiating the project and Mrs. Molly Pederson of the North Slope Borough Inupiaq Language Commission obtained some of the narratives and interviews and provided translations. Finally, the hospitality of the family of Kenneth Toovak during the translation process is gratefully acknowledged.The objective of this program was to extend the data base on ice hazards along the Beaufort Sea coast of Alaska backward in time by using the knowledge and understanding of ice and weather conditions of the local residents. Information for this pilot project was obtained through direct interviews with residents, or from narratives supplied by them. The results of these procedures were evaluated to provide a basis for improving similar efforts in future. Observations of particular interest obtained from these interviews and narratives include (1 ) a description of a major motion of the landfast ice off Harrison Bay in late February, (2) a description of the formation of ice push ridges and ride-up at Cape Halkett during break up, (3) reports of whales traveling inshore of Cross Island during the fall migration and of whales being taken by crews from the Prudhoe Bay area at that time of year, and 4) descriptions of conditions in the nearshore area during summer.     Reciprocity-Enhanced Optical Communication Through Atmospheric Turbulence—Part II: Communication Architectures and Performance

    Free-space optical (FSO) communication provides rapidly deployable, dynamic communication links that are capable of very high data rates compared with those of radio-frequency systems. As such, FSO communication is ideal for mobile platforms, for platforms that require the additional security afforded by the narrow divergence of a laser beam, and for systems that must be deployed in a relatively short time frame. In clear-weather conditions the data rate and utility of FSO communication links are primarily limited by fading caused by microscale atmospheric temperature variations that create parts-per-million refractive-index fluctuations known as atmospheric turbulence. Typical communication techniques to overcome turbulence-induced fading, such as interleavers with sophisticated codes, lose viability as the data rate is driven higher or the delay tolerance is driven lower. This paper, along with its companion [J. Opt. Commun. Netw. 4, 947 (2012)], present communication systems and techniques that exploit atmospheric reciprocity to overcome turbulence that are viable for high data rate and low delay tolerance systems. Part I proves that reciprocity is exhibited under rather general conditions and derives the optimal power-transfer phase compensation for far-field operation. Part II presents capacity-achieving architectures that exploit reciprocity to overcome the complexity and delay issues that limit state-of-the-art FSO communications.United States.     Irradiated Human Endothelial Progenitor Cells Induce Bystander Killing in Human Non-Small Cell Lung and Pancreatic Cancer Cells

    Purpose To investigate whether irradiated human endothelial progenitor cells (hEPCs) could induce bystander killing in the A549 non-small cell lung cancer (NSCLC) cells and help explain the improved radiation-induced tumor cures observed in A549 tumor xenografts co-injected with hEPCs. Materials and Methods We investigated whether co-injection of CBM3 hEPCs with A549 NSCLC cells would alter tumor xenograft growth rate or tumor cure after a single dose of 0 or 5 Gy of X-rays. We then utilized dual chamber Transwell dishes, to test whether medium from irradiated CBM3 and CBM4 hEPCs would induce bystander cell killing in A549 cells, and as an additional control, in human pancreatic cancer MIA PaCa-2 cells. The CBM3 and CBM4 hEPCs were plated into the upper Transwell chamber and the A549 or MIA PaCa-2 cells were plated in the lower Transwell chamber. The top inserts with the CBM3 or CBM4 hEPCs cells were subsequently removed, irradiated, and then placed back into the Transwell dish for 3 h to allow for diffusion of any potential bystander factors from the irradiated hEPCs in the upper chamber through the permeable membrane to the unirradiated cancer cells in the lower chamber. After the 3 h incubation, the cancer cells were re-plated for clonogenic survival. Results We found that co-injection of CBM3 hEPCs with A549 NSCLC cells significantly increased the tumor growth rate compared to A549 cells alone, but paradoxically also increased A549 tumor cure after a single dose of 5 Gy of X-rays (P < 0.05). We hypothesized that irradiated hEPCs may be inducing bystander killing in the A549 NSCLC cells in tumor xenografts, thus improving tumor cure. Bystander studies clearly showed that exposure to the medium from irradiated CBM3 and CBM4 hEPCs induced significant bystander killing and decreased the surviving fraction of A549 and MIA PaCa-2 cells to 0.46 (46%) ± 0.22 and 0.74 ± 0.07 (74%) respectively (P < 0.005, P < 0.0001). In addition, antibody depletion studies demonstrated that the bystander killing induced in both A549 and MIA PaCa-2 cells was mediated by the cytokines TNF-α and TGF-β (P < 0.05). Conclusions These data provide evidence that irradiated hEPCs can induce strong bystander killing in A549 and MIA PaCa-2 human cancer cells and that this bystander killing is mediated by the cytokines TNF-α and TGF-β

    DMAPT inhibits NF-κB activity and increases sensitivity of prostate cancer cells to X-rays in vitro and in tumor xenografts in vivo

    Constitutive activation of the pro-survival transcription factor NF-κB has been associated with resistance to both chemotherapy and radiation therapy in many human cancers, including prostate cancer. Our lab and others have demonstrated that the natural product parthenolide can inhibit NF-κB activity and sensitize PC-3 prostate cancers cells to X-rays in vitro; however, parthenolide has poor bioavailability in vivo and therefore has little clinical utility in this regard. We show here that treatment of PC-3 and DU145 human prostate cancer cells with dimethylaminoparthenolide (DMAPT), a parthenolide derivative with increased bioavailability, inhibits constitutive and radiation-induced NF-κB binding activity and slows prostate cancer cell growth. We also show that DMAPT increases single and fractionated X-ray-induced killing of prostate cancer cells through inhibition of DNA double strand break repair and also that DMAPT-induced radiosensitization is, at least partially, dependent upon the alteration of intracellular thiol reduction-oxidation chemistry. Finally, we demonstrate that the treatment of PC-3 prostate tumor xenografts with oral DMAPT in addition to radiation therapy significantly decreases tumor growth and results in significantly smaller tumor volumes compared to xenografts treated with either DMAPT or radiation therapy alone, suggesting that DMAPT might have a potential clinical role as a radiosensitizing agent in the treatment of prostate cancer

    On the Deformation of a Hyperelastic Tube Due to Steady Viscous Flow Within

    In this chapter, we analyze the steady-state microscale fluid--structure interaction (FSI) between a generalized Newtonian fluid and a hyperelastic tube. Physiological flows, especially in hemodynamics, serve as primary examples of such FSI phenomena. The small scale of the physical system renders the flow field, under the power-law rheological model, amenable to a closed-form solution using the lubrication approximation. On the other hand, negligible shear stresses on the walls of a long vessel allow the structure to be treated as a pressure vessel. The constitutive equation for the microtube is prescribed via the strain energy functional for an incompressible, isotropic Mooney--Rivlin material. We employ both the thin- and thick-walled formulations of the pressure vessel theory, and derive the static relation between the pressure load and the deformation of the structure. We harness the latter to determine the flow rate--pressure drop relationship for non-Newtonian flow in thin- and thick-walled soft hyperelastic microtubes. Through illustrative examples, we discuss how a hyperelastic tube supports the same pressure load as a linearly elastic tube with smaller deformation, thus requiring a higher pressure drop across itself to maintain a fixed flow rate.Comment: 19 pages, 3 figures, Springer book class; v2: minor revisions, final form of invited contribution to the Springer volume entitled "Dynamical Processes in Generalized Continua and Structures" (in honour of Academician D.I.     Protecting eyewitness evidence: Examining the efficacy of a self-administered interview tool

    Given the crucial role of eyewitness evidence, statements should be obtained as soon as possible after an incident. This is not always achieved due to demands on police resources. Two studies trace the development of a new tool, the Self-Administered Interview (SAI), designed to elicit a comprehensive initial statement. In Study 1, SAI participants reported more correct details than participants who provided a free recall account, and performed at the same level as participants given a Cognitive Interview. In Study 2, participants viewed a simulated crime and half recorded their statement using the SAI. After a delay of 1 week, all participants completed a free recall test. SAI participants recalled more correct details in the delayed recall task than control participants

    Electrically Guiding Migration of Human Induced Pluripotent Stem Cells

    A major road-block in stem cell therapy is the poor homing and integration of transplanted stem cells with the targeted host tissue. Human induced pluripotent stem (hiPS) cells are considered an excellent alternative to embryonic stem (ES) cells and we tested the feasibility of using small, physiological electric fields (EFs) to guide hiPS cells to their target. Applied EFs stimulated and guided migration of cultured hiPS cells toward the anode, with a stimulation threshold of <30 mV/mm; in three-dimensional (3D) culture hiPS cells remained stationary, whereas in an applied EF they migrated directionally. This is of significance as the therapeutic use of hiPS cells occurs in a 3D environment. EF exposure did not alter expression of the pluripotency markers SSEA-4 and Oct-4 in hiPS cells. We compared EF-directed migration (galvanotaxis) of hiPS cells and hES cells and found that hiPS cells showed greater sensitivity and directedness than those of hES cells in an EF, while hES cells migrated toward cathode. Rho-kinase (ROCK) inhibition, a method to aid expansion and survival of stem cells, significantly increased the motility, but reduced directionality of iPS cells in an EF by 70–80%. Thus, our study has revealed that physiological EF is an effective guidance cue for the migration of hiPS cells in either 2D or 3D environments and that will occur in a ROCK-dependent manner. Our current finding may lead to techniques for applying EFs in vivo to guide migration of transplanted stem cells
