Transcriptional activation of CBFβ by CDK11p110 is necessary to promote osteosarcoma cell proliferation.
BACKGROUND:Aberrant expression of cyclin-dependent protein kinases (CDK) is a hallmark of cancer. CDK11 plays a crucial role in cancer cell growth and proliferation. However, the molecular mechanisms of CDK11 and CDK11 transcriptionally regulated genes are largely unknown. METHODS:In this study, we performed a global transcriptional analysis using gene array technology to investigate the transcriptional role of CDK11 in osteosarcoma. The promoter luciferase assay, chromatin immunoprecipitation assay, and Gel Shift assay were used to identify direct transcriptional targets of CDK11. Clinical relevance and function of core-binding factor subunit beta (CBFβ) were further accessed in osteosarcoma. RESULTS:We identified a transcriptional role of protein-DNA interaction for CDK11p110, but not CDK11p58, in the regulation of CBFβ expression in osteosarcoma cells. The CBFβ promoter luciferase assay, chromatin immunoprecipitation assay, and Gel Shift assay confirmed that CBFβ is a direct transcriptional target of CDK11. High expression of CBFβ is associated with poor outcome in osteosarcoma patients. Expression of CBFβ contributes to the proliferation and metastatic behavior of osteosarcoma cells. CONCLUSIONS:These data establish CBFβ as a mediator of CDK11p110 dependent oncogenesis and suggest that targeting the CDK11- CBFβ pathway may be a promising therapeutic strategy for osteosarcoma treatment

The Antecedents Of Process Innovation And Smes Growth: Empirical Evidence From Shoe Manufacturing Sector In The Ashanti Region Of Ghana
This article contributes to the study of process innovation as a growth strategy for SMEs, enriching and complementing the widely held view that innovation in general impacts positively on SMEs productivity and growth. In furtherance, this study looks at the process innovation from three main angles as proposed by the Oslo Manuel, which are new process, improved process, and new and improved distribution. Firm level data were purposively collected from shoes manufacturing companies located in the Ashanti Region of Ghana, where Structural Equation Model were employed in coming out with the path estimates in determining the relationship between the variables. The results indicated that, adoption of new and improved distribution strategy impact positively on the growth of the SMEs, by way of reducing the cost of operations and increases customers’ satisfaction. This was followed by the adoption of improved process strategy, where the productivity saw a massive improvement in the areas of quality and quantity, but there was no significance in the reduction of cost of production, whereas the adoption of new process strategy saw an increase in production, but its associated high cost of production negated the growth, to impact negatively on the growth of the SMEs. Base on the results from the study, our results suggest that adoption of process innovation have significant effect on the performance of the shoes manufacturing, the positive impact is not a wholesale, therefore entrepreneurs should not adopt all process innovation blanketly
Study of New Wall Materials Design Based on TRIZ Integrated Innovation Method
New wall materials alleviate the problems of high energy consumption and heavy pollution in the production process through the forms of new natural raw materials, energy conservation, land conservation, waste utilization, etc.. In this paper, design of new building wall materials is achieved through the integrated innovation method of Theory of Inventive Problem Solving (TRIZ), Technology Acceptance Model (TAM), and Quality Function Deployment (QFD). Technical contradictions and physical contradictions in various stages of product design and production are resolved from the perspectives of user survey, R & D design, manufacturing, marketing. Cloning and expression analysis of the FaWRKY70 gene in strawberry
Abstract [Objective] WRKY is one of the plant specific transcription factor families, involved in various
plant life activities. However, currently there are few reports related to the WRKY70 gene in strawberry.
Analyzing the role of WRKY70 homologous genes in strawberry in response to stress will help accelerate
the application of molecular breeding technology and cultivate new strawberry germplasm resources.
[Methods] The FaWRKY70 gene was cloned from ‘Benihoppe’ strawberry fruits using homologous cloning
method. Its conserved domain, physicochemical properties, protein structure, and evolutionary relationship
were analyzed using bioinformatics. Expression pattern analysis was performed using qRT-PCR.
[Results] The FaWRKY70 gene had a length of 1 020 bp and encodes 339 amino acids. Homologous gene
alignment revealed that FaWRKY70 had a high degree of amino acid sequence similarity with species in the
same family such as apple and peony. The homologous genes were mostly related to plant responses to biotic
and abiotic stress, suggesting that FaWRKY70 may be involved in resistance to stress. FaWRKY70
was expressed in different organs of strawberry, with significant differences. The expression level was
highest in flowers and lowest in fruits. Under salicylic acid treatment, the FaWRKY70 gene was quickly
responded, reaching its highest expression level after 3 hours, and gradually decreased thereafter. Under
the treatment with methyl jasmonate, the FaWRKY70 gene showed a slight degree of induction and an
overall downward trend. [Conclusion] FaWRKY70 is involved in growth and hormone signal transduction
through different response modes in strawberry
