    Revision of Madagascar's Dwarf Lemurs (Cheirogaleidae:Cheirogaleus): Designation of Species, Candidate Species Status and Geographic Boundaries Based on Molecular and Morphological Data

    The genus Cheirogaleus, the dwarf lemurs, is a radiation of strepsirrhine primates endemic to the island of Madagascar. The dwarf lemurs are taxonomically grouped in the family Cheirogaleidae (Infraorder: Lemuriformes) along with the genera Microcebus, Mirza, Allocebus, and Phaner. The taxonomic history of the genus Cheirogaleus has been controversial since its inception due to a paucity of evidence in support of some proposed species. In this study, we addressed this issue by expanding the geographic breadth of samples by 91 individuals and built upon existing mitochondrial (cytb and COII) and nuclear (FIBA and vWF) DNA datasets to better resolve the phylogeny of Cheirogaleus. The mitochondrial gene fragments D-loop and PAST as well as the CFTR-PAIRB nuclear loci were also sequenced. In agreement with previous genetic studies, numerous deep divergences were resolved in the C. major, C. minor and C. medius lineages. Four of these lineages were segregated as new species, seven were identified as confirmed candidate species, and four were designated as unconfirmed candidate species based on comparative mitochondrial DNA sequence data gleaned from the literature or this study. Additionally, C. thomasi was resurrected. Given the widespread distribution of the genus Cheirogaleus throughout Madagascar, the methodology employed in this study combined all available lines of evidence to standardize investigative procedures in a genus with limited access to type material and a lack of comprehensive sampling across its total distribution. Our results highlighted lineages that likely represent new species and identified localities that may harbor an as-yet undescribed cryptic species diversity pending further field and laboratory work.We are most grateful to the Ahmanson Foundation, the Theodore F. and Claire M.     An Analytic Needs Assessment for Extension Education: Views from Extension Professionals and Faculty

    This study describes professional development for Extension educators and curricula expectations for those preparing for Extension careers – as perceived by members of the American Association for Agricultural Education (AAAE) and Joint Council of Extension Professionals (JCEP). We relate and differentiate findings from two surveys with open-ended questions. Regarding professional development, JCEP respondents identified program planning and evaluation, but AAAE respondents prioritized research methods. JCEP respondents prioritized practical experiences in Extension Education college courses, and AAAE members reported some practical Extension experiences in the college curriculum. College faculty may apply the results for impactful academic, professional development, and research programs

    An investigation of the mechanical and physical properties of copper-silver alloys and the use of these alloys in Pre-Columbian America

Cataloged from student-submitted PDF version of thesis.Includes bibliographical references (p. 94-96).In both the Andean zone of South America and in Mesoamerica, copper-silver alloys were important in the production of thin, silver-colored sheet metal artifacts. This thesis examines the mechanical and physical properties of the copper-silver alloy system that are important to understanding why copper-silver alloys became central to the metallurgies that developed among prehistoric societies of the Andean zone and Western Mexico. These properties include their range of malleability, the microstructures behind their toughness, and the recrystallization and annealing behaviors that led to their development of silver-enriched surfaces. To determine these properties, a series of cold rolling, cold hammering, and annealing experiments were performed on five Cu-Ag alloys and pure copper. Results of the cold rolling and cold hammering experiments reported here indicate that over the copper-silver alloy compositional range studied, the alloys can be cold rolled without annealing to over 90% reduction in thickness. Similar reductions in thickness were also achieved in two alloys (95 wt% Cu - 5 wt% Ag and 30 wt% Cu - 70 wt% Ag) by cold hammering. The rate of work hardening and the Vickers Hardness Number, as functions of the percent reduction in thickness, are similar for alloy compositions containing between 30 wt% and 80 wt% Cu. This suggests that ancient metalsmiths likely annealed the copper-silver alloy artifacts intentionally to produce the desired silver surface color rather than for any improvement in malleability. The silver surface colors were important for their cultural associations. The recrystallization temperature for the copper-silver alloys tested (70 wt% Cu - 30 wt% Ag and 30 wt% Cu - 70 wt% Ag) is determined to be 500°C given a 30 minute anneal time.by Shannon L.     Development and Testing of a Green Monopropellant Ignition System

    This paper will detail the development and testing of a "green" monopropellant booster ignition system. The proposed booster ignition technology eliminates the need for a pre-heated catalyst bed, a high wattage power source, toxic pyrophoric ignition fluids, or a bi-propellant spark ignitor. The design offers the simplicity of a monopropellant feed system features non-hazardous gaseous oxygen (GOX) as the working fluid. The approach is fundamentally different from all other "green propellant" solutions in the aerospace in the industry. Although the proposed system is more correctly a "hybrid" rocket technology, since only a single propellant feed path is required, it retains all the simple features of a monopropellant system. The technology is based on the principle of seeding an oxidizing flow with a small amount of hydrocarbon.1 The ignition is initiated electrostatically with a low-wattage inductive spark. Combustion gas byproducts from the hydrocarbon-seeding ignition process can exceed 2400 C and the high exhaust temperature ensures reliable main propellant ignition.     Genome-Wide Gene Expression Profiling of Nucleus Accumbens Neurons Projecting to Ventral Pallidum Using both Microarray and Transcriptome Sequencing

    The cellular heterogeneity of brain poses a particularly thorny issue in genome-wide gene expression studies. Because laser capture microdissection (LCM) enables the precise extraction of a small area of tissue, we combined LCM with neuronal track tracing to collect nucleus accumbens shell neurons that project to ventral pallidum, which are of particular interest in the study of reward and addiction. Four independent biological samples of accumbens projection neurons were obtained. Approximately 500 pg of total RNA from each sample was then amplified linearly and subjected to Affymetrix microarray and Applied Biosystems sequencing by oligonucleotide ligation and detection (SOLiD) transcriptome sequencing (RNA-seq). A total of 375 million 50-bp reads were obtained from RNA-seq. Approximately 57% of these reads were mapped to the rat reference genome (Baylor 3.4/rn4). Approximately 11,000 unique RefSeq genes and 100,000 unique exons were identified from each sample. Of the unmapped reads, the quality scores were 4.74 ± 0.42 lower than the mapped reads. When RNA-seq and microarray data from the same samples were compared, Pearson correlations were between 0.764 and 0.798. The variances in data obtained for the four samples by microarray and RNA-seq were similar for medium to high abundance genes, but less among low abundance genes detected by microarray. Analysis of 34 genes by real-time polymerase chain reaction showed higher correlation with RNA-seq (0.66) than with microarray (0.46). Further analysis showed 20–30 million 50-bp reads are sufficient to provide estimates of gene expression levels comparable to those produced by microarray. In summary, this study showed that picogram quantities of total RNA obtained by LCM of ∼700 individual neurons is sufficient to take advantage of the benefits provided by the transcriptome sequencing technology, such as low background noise, high dynamic range, and high precision

    Perspectives of people in Mali toward genetically-modified mosquitoes for malaria control

    Background: Genetically-modified (GM) mosquitoes have been proposed as part of an integrated vector control strategy for malaria control. Public acceptance is essential prior to field trials, particularly since mosquitoes are a vector of human disease and genetically modified organisms (GMOs) face strong scepticism in developed and developing nations. Despite this, in sub-Saharan Africa, where the GM mosquito effort is primarily directed, very little data is available on perspectives to GMOs. Here, results are presented of a qualitative survey of public attitudes to GM mosquitoes for malaria control in rural and urban areas of Mali, West Africa between the months of October 2008 and June 2009. Methods: The sample consisted of 80 individuals - 30 living in rural communities, 30 living in urban suburbs of Bamako, and 20 Western-trained and traditional health professionals working in Bamako and Bandiagara. Questions were asked about the cause of malaria, heredity and selective breeding. This led to questions about genetic alterations, and acceptable conditions for a release of pest-resistant GM corn and malaria-refractory GM mosquitoes. Finally, participants were asked about the decision-making process in their community. Interviews were transcribed and responses were categorized according to general themes. Results: Most participants cited mosquitoes as one of several causes of malaria. The concept of the gene was not widely understood; however selective breeding was understood, allowing limited communication of the concept of genetic modification. Participants were open to a release of pest-resistant GM corn, often wanting to conduct a trial themselves. The concept of a trial was reapplied to GM mosquitoes, although less frequently. Participants wanted to see evidence that GM mosquitoes can reduce malaria prevalence without negative consequences for human health and the environment. For several participants, a mosquito control programme was preferred; however a transgenic release that satisfied certain requirements was usually acceptable. Conclusions: Although there were some dissenters, the majority of participants were pragmatic towards a release of GM mosquitoes. An array of social and cultural issues associated with malaria, mosquitoes and genetic engineering became apparent. If these can be successfully addressed, then social acceptance among the populations surveyed seems promising

    Mixed valency in cerium oxide crystallographic phases: Determination of valence of the different cerium sites by the bond valence method

    We have applied the bond valence method to cerium oxides to determine the oxidation states of the Ce ion at the various site symmetries of the crystals. The crystals studied include cerium dioxide and the two sesquioxides along with some selected intermediate phases which are crystallographically well characterized. Our results indicate that cerium dioxide has a mixed-valence ground state with an f-electron population on the Ce site of 0.27 while both the A- and C-sesquioxides have a nearly pure f^1 configuration. The Ce sites in most of the intermediate oxides have non-integral valences. Furthermore, many of these valences are different from the values predicted from a naive consideration of the stoichiometric valence of the compound

    Sprint cycling rate of torque development associates with strength measurement in trained cyclists

    Purpose: A cyclist’s rate of force/torque development (RFD/RTD) and peak force/torque can be measured during single-joint or whole-body isometric tests, or during cycling. However, there is limited understanding of the relationship between these measures, and of the mechanisms that contribute to each measure. Therefore, we examined the: (i) relationship between quadriceps central and peripheral neuromuscular function with RFD/RTD in isometric knee extension, isometric mid-thigh pull (IMTP), and sprint cycling; and (ii) relationship among RFD/RTD and peak force/torque between protocols. Methods: Eighteen trained cyclists completed two familiarisation and two experimental sessions. Each session involved an isometric knee extension, IMTP, and sprint cycling protocol, where peak force/torque, average and peak RFD/RTD, and early (0 – 100 ms) and late (0–200 ms) RFD/RTD were measured. Additionally, measures of quadriceps central and peripheral neuromuscular function were assessed during the knee extension. Results: Strong relationships were observed between quadriceps early EMG activity (EMG50/M) and knee extension RTD (r or ρ = 0.51 – 0.65) and IMTP late RFD (r = 0.51), and between cycling early or late RTD and peak twitch torque (r or ρ = 0.70 – 0.75). Strong-to-very strong relationships were observed between knee extension, IMTP, and sprint cycling for peak force/torque, early and late RFD/RTD, and peak RFD/RTD (r or ρ = 0.59 – 0.80). Conclusion: In trained cyclists, knee extension RTD or IMTP late RFD are related to measures of quadriceps central neuromuscular function, while cycling RTD is related to measures of quadriceps peripheral neuromuscular function. Further, the strong associations among force/torque measures between tasks indicate a level of transferability across tasks
