    The effects of aqueous root extract of Senna italica subsp. arachoides on the feeding performance of Hyalomma marginatum rufipes adults

    The effects of aqueous root extracts of Senna italica subsp. arachoides on the feeding performance of adults of Hyalomma marginatum rufipes in three consecutive infestations of rabbits were studied under laboratory conditions. Rabbits were divided into treatment group (n = 3), fed aqueous root extracts and rabbit pellets while the control group (n = 3) was fed with rabbit pellets and given distilled water only. Ticks that infested the treatment rabbits fed poorly, taking significantly longer feeding periods (P < 0.05) to acquire the bloodmeal, and having significantly reduced engorgement weights (P < 0.05) compared to those that fed on the control rabbits during the first and second infestations. These results suggest that aqueous root extracts of S. italica subsp. arachoides reduced the feeding performance of H. marginatumrufipes adults

    Aloe arborescens aqueous gel extract alters the activities of key hepatic enzymes and blood concentration of triglycerides, glucose and insulin in alloxan-induced diabetic rats

    The present study investigated the antidiabetic activity and the possible mechanisms of action of aqueous extract of Aloe arborescens leaf gel (AALGEt) on normal and alloxan-induced diabetic rats. Diabetes was induced in 12 h fasted rats by intraperitoneal injection of 140 mg/kg body weight of alloxan. Blood glucose levels, body weight and water intake were determined on day 7, 14 and 21 of AALGEt treatment. Plasma insulin and triglycerides levels, as well as activities of hepatic glucokinase and glucose 6-phosphatase (G6Pase) were determined at the end of the study. Blood glucose levels, plasma triglyceride and insulin levels, as well as the activity of hepatic G6Pase were significantly increased in diabetic rats. With the exception of hepatic glucokinase activity, daily oral administration of AALGEt to diabetic rats significantly reversed the effects induced by alloxan. The activities of glucokinase and glucose-6-phosphatase as well as plasma insulin levels in AALGEt-treated normal rats were comparable with those observed in untreated normal rats. The results suggest that AALGEt ameliorates physiological parameters altered by the diabetic state. These effects may be mediated in part, through the protection of pancreatic beta cells from further damage by alloxan.Keywords: Aloe arborescens, alloxan, diabetic rats, antidiabetic activity, hepatic enzymes

    Radical scavenging activity of selected medicinal plants from Limpopo Province of South Africa

    Plants collected from Limpopo province of South Africa were investigated for their antioxidative potential using the DPPH radical scavenging assay. Acetone extracts of Flueggea virosa had the highest antioxidant activity with an IC50 value of 30 ìg/ml, closely matching the ascorbic acid with an IC50 value of 25 ìg/ml. The lowest antioxidant readings were observed with extracts of Rhynchosia venulosa (root extract) and Ficus ingens (leaf extract). Acetone extract of Bridelia virosa leaves had the highest phenolic content (156 mg GAE/g extract), while the lowest content was recorded for R. venulosa root extract and leaf extract of F. ingens (8.3 and 17.7 mg GAE/g extract, respectively). There was a linear correlation between antioxidant activityand total phenolic content. Extracts with high phenolic content had low IC50 values, while extracts with low phenolic concentrations had high IC50 values

    The phytochemical, antibacterial and antioxidant activity of five medicinal plants against the wound infecting bacteria

    Leaf extracts of Senna italica, Ricinus communis, Lantana camara, Lippia javanica and Ziziphus  mucronata were screened for biological activity against bacteria which infect wounds. The leaves were extracted using different solvents of varying polarity (hexane, dichloromethane, acetone and methanol). Phytochemical analyses of the extracts were performed using thin layer chromatography (TLC). The extracts were loaded on TLC plates and developed in three solvent systems that is benzene/ethanol /ammonium solution (BEA), chloroform/ethyl acetate/formic acid (CEF) and ethyl acetate/methanol /water (EMW). Antibacterial activity of the plants was evaluated using micro-dilution and bioautography methods. The test organisms used were Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus and Enterococcus faecalis. Acetone extracts were chosen for antioxidant activity. Methanol was the best extractant, followed by acetone and dichloromethane (DCM). In the phytochemical analysis,  more compounds were observed on BEA, followed by EMW and CEF plates. Lantana camara had no  activity against any of the bacteria used. P. aeruginosa was the most resistant bacterium with only two plants active against it. E. faecalis and E. coli were sensitive to the extracts. More antibacterial  compounds were observed on BEA plates against all the test bacteria in bioautographic method. The Rf  values calculated from bioautography indicated that the selected plants have different active compounds. The most active compounds were from S. italica and Z. mucronata. BEA and EMW plates had good  antioxidant activity. No antioxidant activity was observed on the CEF plate. Most extracts were active against wound pathogens; their application on the wound area may prevent infection. Further studies are required to identify the active compounds in the plant extracts which showed significant anti-bacterial activities.Key words: Thin layer chromatography (TLC), plant extract, bacteria

    Medicinal plant use of villagers in the Mopani district, Limpopo province, South Africa

    Background: An ethno-medical survey of plants was conducted at Mashishimale village near Phalaborwa, in Limpopo Province (South Africa). Owing to high level of poverty and lack of proper daily operational healthcare facilities in certain parts of Mashishimale village, the use of plants as medicine persists. The aim of the study was to compile a list of medicinal plants used by traditional health practitioners for treatment of diabetes mellitus and other ailments.Materials and methods: Traditional health practitioners were identified using the snowballing sampling technique. Structured interviews were conducted and a list of medicinal plants compiled. Data collection included local names of the plants, the plant parts used in the traditional medicine, the disease treated by the plant-based remedies and the method of preparation of the medicines.Results and discussion: A total of 49 plants species belonging to 20 different families we identified. Plant parts used in preparation of remedies by practitioners are roots (51%), stem barks (30%) and leaves (19%). The most preferred methods of administration were boiling (74%) and infusion (12%). About 14 plants species were indicated for treatment of diabetes mellitus, while most plant species were indicated for sexually transmitted diseases, fertility and erectile dysfunction.Conclusion: The results of the survey indicate that plants play a significant role in primary healthcare of the community of Mashishimale Village. It would seem that a number of major diseases/ailments are treated by traditional methods. Scientific investigation is therefore needed to isolate active compounds and to determine safe dosages for treatment.     Unlocking the in vitroanti- inflammatory and antidiabetic potential of Polygonum maritimum

    Context: Several Polygonum species (Polygonaceae) are used in traditional medicine in Asia, Europe and Africa to treat inflammation and diabetes. Objective: Evaluate the in vitro antioxidant, anti-inflammatory and antidiabetic potential of methanol and dichloromethane extracts of leaves and roots of the halophyte Polygonum maritimum L. Material and methods: Antioxidant activity was determined (up to 1mg/mL) as radical-scavenging activity (RSA) of 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), copper (CCA) and iron (ICA) chelating activities and iron reducing power (FRAP). NO production was measured in lipopolysaccharide (LPS)-stimulated macrophages for 24 h at concentrations up to 100 mu g/mL and antidiabetic potential was assessed by alpha-amylase and alpha-glucosidase inhibition (up to 10 g/mL) assays. The phytochemical composition of the extracts was determined by gas chromatography-mass spectrometry (GC-MS). Results: The methanol leaf extract had the highest activity against DPPH center dot (IC50 = 26 mu g/mL) and ABTS1(+)center dot (IC50 = 140 mu g FRAP (IC50 = 48 mu g/mL) and CCA (IC50 = 770 mu g/mL). Only the dichloromethane leaf extract (LDCM) showed anti-inflammatory activity (IC50 = 48 mu g/mL). The methanol root (IC50 = 19 mu g/mL) and leaf (IC50 = 29 mu g/mL) extracts strongly inhibited baker's yeast alpha-glucosidase, but LDCM had higher rat's alpha-glucosidase inhibition (IC50 = 2527 mu g/mL) than acarbose (IC50 = 4638 mu g/mL). GC-MS analysis identified beta-sitosterol, stigmasterol, 1-octacosanol and linolenic acid as possible molecules responsible for the observed bioactivities.     A latent class analysis of young women’s co-occurring health risks in urban informal settlements in Durban, South Africa

Few studies have explicitly measured the co-occurrence of these health risks or the impact on mental health and wellbeing for this population. To this aim, we use a person-centred approach to explore the clustering of health risks among young Black women from urban informal settlements in Durban, South Africa, enrolled in an intervention trial. Latent class analysis identified three health risk subgroups with increasing levels of health risk co-occurrence: while all three subgroups had high rates of emotional/economic intimate partner violence, they differed in their levels of the other health risks, with one (“lower-risk”) subgroup defined by experiencing violence against women (VAW), another by the co-occurrence of VAW with problematic alcohol use (i.e. “mid-risk”), and the last (“high-risk”) subgroup by the co-occurrence of VAW, problematic alcohol use and sexual risk behaviour. Descriptive analyses showed that lower education and food insecurity were associated with greater health risk co-occurrence and that this in turn was associated with increased chances of depression and suicidal ideation. Between subgroup differences persisted over time - after two years, the chances of experiencing violence, problematic alcohol use, transactional sex and depression remained elevated for the women who initially experienced more health risks. Persistent yet differing levels of risk suggest the need for urgent structural interventions that address these health risks synergistically while taking account of individual differing primary and secondary prevention needs. Our analyses highlight that social epidemics such as poverty, racism and gender inequality play into the production of poor health outcomes, including poor mental health. These are the underlying structural issues that need to be addressed in order to protect women’s health and reduce harm.UKR

    Challenges and opportunities in coproduction: reflections on working with young people to develop an intervention to prevent violence in informal settlements in South Africa

Not applicable.Coproduction is widely recognised as essential to the development of effective and sustainable complex health interventions. Through involving potential end users in the design of interventions, coproduction provides a means of challenging power relations and ensuring the intervention being implemented accurately reflects lived experiences. Yet, how do we ensure that coproduction delivers on this promise? What methods or techniques can we use to challenge power relations and ensure interventions are both more effective and sustainable in the longer term? To answer these questions, we openly reflect on the coproduction process used as part of Siyaphambili Youth (‘Youth Moving Forward’), a 3-year project to create an intervention to address the social contextual factors that create syndemics of health risks for young people living in informal settlements in KwaZulu-Natal province in South Africa. We identify four methods or techniques that may help improve the methodological practice of coproduction: (1) building trust through small group work with similar individuals, opportunities for distance from the research topic and mutual exchanges about lived experiences; (2) strengthening research capacity by involving end users in the interpretation of data and explaining research concepts in a way that is meaningful to them; (3) embracing conflicts that arise between researchers’ perspectives and those of people with lived experiences; and (4) challenging research epistemologies through creating spaces for constant reflection by the research team. These methods are not a magic chalice of codeveloping complex health interventions, but rather an invitation for a wider conversation that moves beyond a set of principles to interrogate what works in coproduction practice. In order to move the conversation forward, we suggest that coproduction needs to be seen as its own complex intervention, with research teams as potential beneficiaries.Medical Research Council (MRC

    Activity of the multikinase inhibitor dasatinib against ovarian cancer cells

    BackgroundHere, we explore the therapeutic potential of dasatinib, a small-molecule inhibitor that targets multiple cytosolic and membrane-bound tyrosine kinases, including members of the Src kinase family, EphA2, and focal adhesion kinase for the treatment of ovarian cancer.MethodsWe examined the effects of dasatinib on proliferation, invasion, apoptosis, cell-cycle arrest, and kinase activity using a panel of 34 established human ovarian cancer cell lines. Molecular markers for response prediction were studied using gene expression profiling. Multiple drug effect/combination index (CI) isobologram analysis was used to study the interactions with chemotherapeutic drugs.ResultsConcentration-dependent anti-proliferative effects of dasatinib were seen in all ovarian cancer cell lines tested, but varied significantly between individual cell lines with up to a 3 log-fold difference in the IC(50) values (IC(50) range: 0.001-11.3 micromol l(-1)). Dasatinib significantly inhibited invasion, and induced cell apoptosis, but less cell-cycle arrest. At a wide range of clinically achievable drug concentrations, additive and synergistic interactions were observed for dasatinib plus carboplatin (mean CI values, range: 0.73-1.11) or paclitaxel (mean CI values, range: 0.76-1.05). In this study, 24 out of 34 (71%) representative ovarian cancer cell lines were highly sensitive to dasatinib, compared with only 8 out of 39 (21%) representative breast cancer cell lines previously reported. Cell lines with high expression of Yes, Lyn, Eph2A, caveolin-1 and 2, moesin, annexin-1, and uPA were particularly sensitive to dasatinib.ConclusionsThese data provide a clear biological rationale to test dasatinib as a single agent or in combination with chemotherapy in patients with ovarian cancer
