    International chicken trade and increased risk for introducing or reintroducing highly pathogenic avian influenza A (H5N1) to uninfected countries.

    Every year billions of chickens are shipped thousands of miles around the globe in order to meet the ever increasing demands for this cheap and nutritious protein source. Unfortunately, transporting chickens internationally can also increase the chance for introducing zoonotic viruses, such as highly pathogenic avian influenza A (H5N1) to new countries. Our study used a retrospective analysis of poultry trading data from 2003 through 2011 to assess the risk of H5N1 poultry infection in an importing country. We found that the risk of infection in an importing country increased by a factor of 1.3 (95% CI: 1.1-1.5) for every 10-fold increase in live chickens imported from countries experiencing at least one H5N1 poultry case during that year. These results suggest that the risk in a particular country can be significantly reduced if imports from countries experiencing an outbreak are decreased during the year of infection or if biosecurity measures such as screening, vaccination, and infection control practices are increased. These findings show that limiting trade of live chickens or increasing infection control practices during contagious periods may be an important step in reducing the spread of H5N1 and other emerging avian influenza viruses

    Hidden Markov models identify major movement modes in accelerometer and magnetometer data from four albatross species

Thorne under award number 79804, and by a Minghua Zhang Early Career Faculty Innovation award to L Thorne.Background : Inertial measurement units (IMUs) with high-resolution sensors such as accelerometers are now used extensively to study fine-scale behavior in a wide range of marine and terrestrial animals. Robust and practical methods are required for the computationally-demanding analysis of the resulting large datasets, particularly for automating classification routines that construct behavioral time series and time-activity budgets. Magnetometers are used increasingly to study behavior, but it is not clear how these sensors contribute to the accuracy of behavioral classification methods. Development of effective classification methodology is key to understanding energetic and life-history implications of foraging and other behaviors.  Methods : We deployed accelerometers and magnetometers on four species of free-ranging albatrosses and evaluated the ability of unsupervised hidden Markov models (HMMs) to identify three major modalities in their behavior: ‘flapping flight’, ‘soaring flight’, and ‘on-water’. The relative contribution of each sensor to classification accuracy was measured by comparing HMM-inferred states with expert classifications identified from stereotypic patterns observed in sensor data.  Results : HMMs provided a flexible and easily interpretable means of classifying behavior from sensor data. Model accuracy was high overall (92%), but varied across behavioral states (87.6, 93.1 and 91.7% for ‘flapping flight’, ‘soaring flight’ and ‘on-water’, respectively). Models built on accelerometer data alone were as accurate as those that also included magnetometer data; however, the latter were useful for investigating slow and periodic behaviors such as dynamic soaring at a fine scale.  Conclusions : The use of IMUs in behavioral studies produces large data sets, necessitating the development of computationally-efficient methods to automate behavioral classification in order to synthesize and interpret underlying patterns. HMMs provide an accessible and robust framework for analyzing complex IMU datasets and comparing behavioral variation among taxa across habitats, time and space.Publisher PDFPeer reviewe

    Human IL-12 p40 as a reporter gene for high-throughput screening of engineered mouse embryonic stem cells

    <p>Abstract</p> <p>Background</p> <p>Establishing a suitable level of exogenous gene expression in mammalian cells in general, and embryonic stem (ES) cells in particular, is an important aspect of understanding pathways of cell differentiation, signal transduction and cell physiology. Despite its importance, this process remains challenging because of the poor correlation between the presence of introduced exogenous DNA and its transcription. Consequently, many transfected cells must be screened to identify those with an appropriate level of expression. To improve the screening process, we investigated the utility of the human interleukin 12 (IL-12) p40 cDNA as a reporter gene for studies of mammalian gene expression and for high-throughput screening of engineered mouse embryonic stem cells.</p> <p>Results</p> <p>A series of expression plasmids were used to study the utility of IL-12 p40 as an accurate reporter of gene activity. These studies included a characterization of the IL-12 p40 expression system in terms of: (i) a time course of IL-12 p40 accumulation in the medium of transfected cells; (ii) the dose-response relationship between the input DNA and IL-12 p40 mRNA levels and IL-12 p40 protein secretion; (iii) the utility of IL-12 p40 as a reporter gene for analyzing the activity of <it>cis</it>-acting genetic elements; (iv) expression of the IL-12 p40 reporter protein driven by an IRES element in a bicistronic mRNA; (v) utility of IL-12 p40 as a reporter gene in a high-throughput screening strategy to identify successful transformed mouse embryonic stem cells; (vi) demonstration of pluripotency of IL-12 p40 expressing ES cells <it>in vitro </it>and <it>in vivo</it>; and (vii) germline transmission of the IL-12 p40 reporter gene.</p> <p>Conclusion</p> <p>IL-12 p40 showed several advantages as a reporter gene in terms of sensitivity and ease of the detection procedure. The IL-12 p40 assay was rapid and simple, in as much as the reporter protein secreted from the transfected cells was accurately measured by ELISA using a small aliquot of the culture medium. Remarkably, expression of Il-12 p40 does not affect the pluripotency of mouse ES cells. To our knowledge, human IL-12 p40 is the first secreted reporter protein suitable for high-throughput screening of mouse ES cells. In comparison to other secreted reporters, such as the widely used alkaline phosphatase (SEAP) reporter, the IL-12 p40 reporter system offers other real advantages.</p

    Trans-equatorial migration of Short-tailed Shearwaters revealed by geolocators

    Until recent decades, details of the migratory movements of seabirds remained largely unknown owing to the difficulties in following individuals at sea. Subsequent advances in biologging technology have greatly increased our knowledge of seabird migration and distribution, particularly of highly pelagic species. Short-tailed Shearwaters (Ardenna tenuirostris) (~500 g) have been studied extensively during their breeding season but our understanding of their movements outside this period remains poor. Here, we present the first tracks of the trans-equatorial migration of Short-tailed Shearwaters from a colony on Great Dog Island, Tasmania, Australia. Data were obtained from global location sensors (GLS loggers or geolocators), which enable the estimation of location twice per day based on ambient light levels. After breeding, tracked Shearwaters flew south of the Antarctic Polar Front to a previously unknown stopover site, where they remained for several weeks, before travelling rapidly northward through the western Pacific Ocean to coastal waters off Japan. Short-tailed Shearwaters spent the bulk of the northern hemisphere summer, either in this region or further north in the Bering Sea, before returning south through the central Pacific to their breeding sites. Our results, for the first time, describe in detail the complete migration of this long-lived seabird, reveal individual variation in timing and distribution, and describe the environmental characteristics of their key non-breeding habitats

    Water sensitive urban design in the city of the future

    With timeframes for addressing the issues of the City of the Future (CotF) rapidly approaching (e.g. 2020, 2025, 2050), this paper integrates international research knowledge and expertise from four continents. It critically evaluates the role of water sensitive urban design (WSUD) in the CotF in terms of overlapping theory and practice. The aspirations of water sensitive cities are reviewed and multiple drivers for applying WSUD are described from developing and developed country perspectives In addition, the potential for WSUD to support cities in ‘leap-frogging’ towards their visions are explored. The role of WSUD within the wider context of achieving sustainable living objectives (e.g. greater resilience, low carbon living, sustainable transportation, local food supply and social stability) is debated and the concept of the ‘multi-objective city’ introduced. Conclusions are drawn regarding opportunities for the WSUD process to provide a framework within which professionals from many disciplines can support landscape architects and urban planners in achieving multiobjective liveable cities are identified

    The interdependence of behavioral and somatic health: implications for conceptualizing health and measuring treatment outcomes

    <b>Purpose:</b> The interdependence of behavioral and somatic aspects of various health conditions warrants greater emphasis on an integrated care approach.<br><br> <b>Theory:</b> We propose that integrated approaches to health and wellness require comprehensive and empirically-valid outcome measures to assess quality of care.<br><br> <b>Method:</b> We discuss the transition from independent to integrated treatment approaches and provide examples of new systems for integrated assessment of treatment outcome.<br><br> <b>Results:</b> Evidence suggests that support for an independent treatment approach is waning and momentum is building towards more integrated care. In addition, research evidence suggests integrated care improves health outcomes, and both physicians and patients have favorable impressions of integrated care.<br><br> <b>Conclusions:</b> As treatment goals in the integrated perspective expand to take into account the intimate relationships among mental illness, overall health, and quality of life, clinicians need to develop outcome measures that are similarly comprehensive. Discussion: Increased recognition, by researchers, providers, and insurers, of the interdependence between behavioral and physical health holds great promise for innovative treatments that could significantly improve patients' lives
