Silver as antibacterial toward Listeria monocytogenes

    Get PDF
Listeria monocytogenes is a serious foodborne pathogen that can contaminate food during processing and can grow during food shelf-life. New types of safe and effective food contact materials embedding antimicrobial agents, like silver, can play an important role in the food industry. The present work aimed at evaluating the in vitro growth kinetics of different strains of L. monocytogenes in the presence of silver, both in its ionic and nano form. The antimicrobial effect was determined by assaying the number of culturable bacterial cells, which formed colonies after incubation in the presence of silver nanoparticles (AgNPs) or silver nitrate (AgNO(3)). Ionic release experiments were performed in parallel. A different reduction of bacterial viability between silver ionic and nano forms was observed, with a time delayed effect exerted by AgNPs. An association between antimicrobial activity and ions concentration was shown by both silver chemical forms, suggesting the major role of ions in the antimicrobial mode of action

In Patagonia, sheep farming constitutes one of the major economic activities, but the quality of lamb meat is seriously affected by a disagreeable taste found in its tissues, derived from the ovine intake of neneo (Mulinum spinosum (Cav.) Persoon, Apiaceae), predominant in grazing lands easily accessed by sheep during the warm seasons. With the aim of detecting the affected animals before their slaughter, phytochemical testing techniques were applied on different organs of the plant and animal accordingly, in order to identify matches. We studied two groups (one had neneo in their diet while the other did not) of six animals each, of which were extracted samples of meat, fat, liver, urine and blood. The compounds responsible for the effect of neneo belong to the terpene/sterol group, the only present in the flowers of neneo as in the urine of lambs. Various animal tissues were inadequate to detect the presence of neneo in lambs, only urine has been identified as the most suitable to examine the metabolites. A protocol, to experiment with it, was proposed, with the aim of generate a useful tool for the development of low-cost technologies. This protocol is available to researchers and producers to put to the test, with the aim of detecting a disagreeable taste in the animals before their slaughter and avoiding the consequent economic loss.Fil: Seoane, Nicolás Fernando. The apolipoprotein A-I Milano (apoA-I M ) is a mo- lecular variant of apoA-I characterized by the Arg 173 → Cys substitution, leading to the formation of homodimers A-I M / A-I M. Upon interaction with palmitoyloleoylphosphatidyl- choline, A-I M /A-I M forms only two species of reconstituted HDL (rHDL) particles, with diameters of 7.8 and 12.5 nm. We used limited proteolysis to analyze the conformation of A-I M /A-I M in the two rHDL particles, in comparison with that of apoA-I in rHDL of similar size. ApoA-I in the small, 7.8-nm rHDL is degraded to a greater extent (50% after 6 h) than in the large rHDL ( 10% degraded after 6 h). The pro- tease susceptibility of A-I M /A-I M in small and large rHDL is instead remarkably the same, with A-I M /A-I M being much more sensitive to proteolytic digestion (50% degraded after 10 min) than apoA-I. The identification of the proteolytic fragments by immunoblotting, N-terminal sequencing, and molecular mass determination, shows that the N-terminus of both proteins is resistant to proteolysis, with six cleavage sites located in the central and carboxy-terminal portions of the molecules. Cleavage in the middle of apoA-I occurs at dis- tinct sites in 7.8-nm (Lys 118 ) and 12.7-nm (Arg 123 ) rHDL, in- dicating a different conformation in small and large rHDL particles. The A-I M /A-I M instead adopts a unique and identi- cal conformation in small and large rHDL, with the carboxy- terminal portion of the molecule being remarkably more ac- cessible to the proteases than in apoA-I. This suggests the presence of a novel carboxy-terminal domain in A-I M /A-I M , not organized in a compact structure and not shared by wild-type apoA-I, which may account for the unique functional proper- ties of A-I M /A-I M. —Calabresi, L., G. Neuroendocrine tumors (NETs) can be sporadic or they can arise in complex hereditary syndromes. Patients with hereditary NETs can be identified before the development of tumors by performing genetic screenings. The aim of the study was to evaluate the clinical and prognostic impact of a preclinical genetic screening in subjects with hereditary NET syndromes. 46 subjects referred for hereditary NET syndrome [22 MEN1, 12 MEN2, 12 Familial Paragangliomatosis (FPGL)] were enrolled and divided in 2 groups (group A, 20 subjects with clinical appearance of NET before the genetic diagnosis; group B, 26 subjects with genetic diagnosis of hereditary NET syndromes before the clinical appearance of NETs). The main outcome measures were severity of disease, prognosis, and survival. The rate of surgery for MEN1-, MEN2-, FPGL4-related tumors was 90% in group A and 35% in group B (p<0.01). Both symptoms related to tumors and symptoms related to therapies were significantly less frequent in group B than in group A (p<0.05). Tumor stage was locally advanced or metastatic in 50% of group A and in no one of group B (p<0.01). The mortality rate was 25% in group A and 0% in group B (p<0.05). An early genetic screening for hereditary NET syndromes results in an improvement in clinical presentation and morbidity. A potential impact of the genetic screening on the mortality rate of these subjects is suggested and needs to be investigated in further and more appropriate studies

Revisión de Programas hospitalarios para tratar la ansiedad quirúrgica infantil: ¿Es evidente todo lo que reluce?

    Get PDF
There are still few hospitals that consider it important to include in their services a program that helps to decrease the level of anxiety in the patients. However, recently it is possible to find different initiatives and programs in the pediatric area for this purpose. Initiatives born under the current "humanization of hospitals" trying to achieve, with greater or lesser fortune, a closeness and better assimilation of the surgical protocols present in a hospital. Personally we believe that there is nothing more human than to exercise our office. Exercising medicine and curing a disease implies humanity in itself, even if it is applied under scientific evidence and seeking an adequate effectiveness. There is nothing more human than the fact of healing others, although in that way science is prioritized over comfort and closeness. Therefore, the authors are in favor of the new, and poorly called, humanization of health but, first ensuring, proper care. We are therefore witnessing the birth of proposals and initiatives that try to reduce the anxiety level of our smaller patients, who try to bring our medical activity closer to the daily life and try to transform the instrumentalization and modernization of medicine into something understandable to all. Something positive and desirable. However we can not forget that medicine is based on the scientific method, and that without it, these initiatives can remain in a mere superficial marketing. They may even endanger the performance and health outcomes of the health care activity itself. This does not mean in any way that the aspects that concern the psyche and the emotional part of the patient are not important. In fact they are a fundamental part of the medical activity but always from a scientific and consensual point of view. That is why it is always desirable to quantify results and evaluate aspects that aim to improve our work. In the case of childhood preoperative anxiety, initiatives and protocols have been developed that try to alleviate the causes and their consequences. The development of stories, toys, the presence of parents in anesthetic induction and other actions seem to be a current trend developed by some hospitals concerned about this issue. But what is the truth about it? How are these initiatives properly developed? And above all ...

Immunoanalytical Approach for Detecting and Identifying Ancestral Peptide Biomarkers in Early Earth Analogue Environments

Several mass spectrometry and spectroscopic techniques have been used in the search for molecular biomarkers on Mars. A major constraint is their capability to detect and identify large and complex compounds such as peptides or other biopolymers. Multiplex immunoassays can detect these com-pounds, but antibodies must be produced for a large number of sequence-dependent molecular targets. Ancestral Sequence Re-construction (ASR) followed by protein "resurrection" in the lab can help to narrow the selection of targets. Herein, we propose an immunoanalytical method to identify ancient and universally conserved protein/peptide sequences as targets for identifying ancestral biomarkers in nature. We have developed, tested, and validated this approach by producing antibodies to eight previously described ancestral resurrected proteins (three beta-lactamases, three thioredoxins, one Elongation Factor Tu, and one RuBisCO, all of them theoretically dated as Precambrian), and used them as a proxy to search for any potential feature of them that could be present in current natural environments. By fluorescent sandwich microarray immunoassays (FSMI), we have detected positive immunoreactions with antibodies to the oldest beta-lactamase and thioredoxin proteins (ca. 4 Ga) in samples from a hydrothermal environment. Fine epitope mapping and inhibitory immunoassays allowed the identification of well-conserved epitope peptide sequences that resulted from ASR and were present in the sample. We corroborated these results by metagenomic sequencing and found several genes encoding analogue proteins with significant matches to the peptide epitopes identified with the antibodies. The results demonstrated that peptides inferred from ASR studies have true counterpart analogues in Nature, which validates and strengthens the well-known ASR/protein resurrection technique and our immunoanalytical approach for investigating ancient environments and metabolisms on Earth and elsewhere
