    Low-Income Mothers\u27 Perceptions of Barriers to Using Farmers Markets: A SNAP-Ed Initiative to Understand Access Points to Local Foods

    The study reported here describes a Virginia Family Nutrition Program\u27s target population\u27s perceptions of barriers to using a farmers market to access local foods. Mothers from a rural county photographed their shopping experience. Using a photo elicitation process to develop themes related to food access, the mothers identified barriers to shopping at farmers markets. The results can provide guidance to agents, program assistants, and farmers market coordinators on promoting use of farmers markets by low-income populations. The development of experiential learning opportunities to overcome barriers is critical if farmers markets are encouraged as an avenue for promoting healthy eating

    Online Advertising for Cancer Prevention: Google Ads and Tanning Beds

    Google receives more than 3.5 billion Internet searches daily, and the advertisements on their results pages may provide a unique opportunity to transmit targeted public health information to a large audience. Skin cancer is more common than all other cancers combined, and indoor tanning is a preventable risk factor that accounts for more than 450 000 new malignant neoplasms each year. Tanning bed use remains common, with 1 in 5 adolescents and more than half of college students exposed. Awareness of the dangers of tanning beds is one of the factors that can lead to behavior change.     Engagement of patients with scleroderma to revise an internet self-management program

    Systemic sclerosis (SSc) or scleroderma is a rare connective tissue disease. Many people do not have access to education programs. A self-management program was developed several years ago based on the literature and input from people with SSc. However, new therapies and treatment options have been developed since the program was developed. The purpose of this qualitative study was to identify and remedy gaps in an internet SSc self-management program to improve the quality of critical information relevant to effective management of the disease. Six focus groups with 30 participants with SSc were conducted: 2 telephone groups and 4 face-to-face groups. Prior to the focus group meetings, participants reviewed the existing website. A semi-structured interview guide elicited participants’ responses. Gaps were expressed in affect and positive affirmation; disease and symptom management; self-advocacy; information for caregivers, families, coworkers and strangers; tracking systems; information about local support groups; pictures and information on underrepresented groups; and general format. Discussants were positive regarding the audio voice over, exercise module, current content, health logs and checklists. People with SSc identified additional content to improve the internet self-management program. Many of the suggestions were incorporated into the existing program as modifications and additions to existing modules, patient testimonials, worksheets, resources sheets, and/or links to additional websites. People with rare, chronic conditions such as SSc need education and reliable sources of information and self-management skills.     IL-13 is a driver of COVID-19 severity

    Immune dysregulation is characteristic of the more severe stages of SARS-CoV-2 infection. Understanding the mechanisms by which the immune system contributes to COVID-19 severity may open new avenues to treatment. Here, we report that elevated IL-13 was associated with the need for mechanical ventilation in 2 independent patient cohorts. In addition, patients who acquired COVID-19 while prescribed Dupilumab, a mAb that blocks IL-13 and IL-4 signaling, had less severe disease. In SARS-CoV-2–infected mice, IL-13 neutralization reduced death and disease severity without affecting viral load, demonstrating an immunopathogenic role for this cytokine. Following anti–IL-13 treatment in infected mice, hyaluronan synthase 1 (Has1) was the most downregulated gene, and accumulation of the hyaluronan (HA) polysaccharide was decreased in the lung. In patients with COVID-19, HA was increased in the lungs and plasma. Blockade of the HA receptor, CD44, reduced mortality in infected mice, supporting the importance of HA as a pathogenic mediator. Finally, HA was directly induced in the lungs of mice by administration of IL-13, indicating a new role for IL-13 in lung disease. Understanding the role of IL-13 and HA has important implications for therapy of COVID-19 and, potentially, other pulmonary diseases. IL-13 levels were elevated in patients with severe COVID-19. In a mouse model of the disease, IL-13 neutralization reduced the disease and decreased lung HA deposition. Administration of IL-13–induced HA in the lung.     Factors Determining Seagrass Blue Carbon Across Bioregions and Geomorphologies

We examined the variability in seagrass soil Corg stocks and composition across Australia and identified the main drivers of variability, applying a spatially hierarchical approach that incorporates bioregions and geomorphic settings. Top 30 cm soil Corg stocks were similar across bioregions and geomorphic settings (min-max: 20–26 Mg Corg ha−1), but meadows formed by large species (i.e., Amphibolis spp. and Posidonia spp.) showed higher stocks (24–29 Mg Corg ha−1) than those formed by smaller species (e.g., Halodule, Halophila, Ruppia, Zostera, Cymodocea, and Syringodium; 12–21 Mg Corg ha−1). In temperate coastal meadows dominated by large species, soil Corg stocks mainly derived from seagrass Corg (72 ± 2%), while allochthonous Corg dominated soil Corg stocks in meadows formed by small species in temperate and tropical estuarine meadows (64 ± 5%). In temperate coastal meadows, soil Corg stocks were enhanced by low hydrodynamic exposure associated with high mud and seagrass Corg contents. In temperate estuarine meadows, soil Corg stocks were enhanced by high contributions of seagrass Corg, low to moderate solar radiation, and low human pressure. In tropical estuarine meadows formed by small species, large soil Corg stocks were mainly associated with low hydrodynamic energy, low rainfall, and high solar radiation.     Apolipoprotein E, especially apolipoprotein E4, increases the oligomerization of amyloid β peptide

    Alzheimer’s disease (AD) is the most common progressive neurodegenerative disorder causing dementia. Massive deposition of amyloid β peptide (Aβ) as senile plaques in the brain is the pathological hallmark of AD, but oligomeric, soluble forms of Aβ have been implicated as the synaptotoxic component. The apolipoprotein E epsilon 4 (apoE ε4) allele is known to be a genetic risk factor for developing AD. However it is still unknown how apoE impacts the process of Aβ oligomerization. Here, we found that the level of Aβ oligomers in APOEε4/ε4 AD patient brains is 2.7 times higher than those in APOEε3/ε3 AD patient brains, matched for total plaque burden, suggesting that apoE4 impacts the metabolism of Aβ oligomers. To test this hypothesis, we examined apoE’s effect on Aβ oligomer formation. Using both synthetic Aβ and a split-luciferase method for monitoring Aβ oligomers, we observed that apoE increased the level of Aβ oligomers in an isoform dependent manner (E2 < E3 < E4). This effect appears to be dependent on the ApoE carboxy-terminal domain. Moreover, these results were confirmed using endogenous apoE isolated from the TBS-soluble fraction of human brain, which increased the formation of Aβ oligomers. Taken together, these data show that lipidated apoE, especially apoE4, increases Aβ oligomers in the brain. Higher levels of Aβ oligomers in the brains of APOEε4/ε4 carriers compared to APOEε3/ε3 carriers may increase the loss of dendritic spines and accelerate memory impairments, leading to earlier cognitive decline in AD

    Development of a Tumor-Selective Approach to Treat Metastatic Cancer

    BACKGROUND: Patients diagnosed with metastatic cancer have almost uniformly poor prognoses. The treatments available for patients with disseminated disease are usually not curative and have side effects that limit the therapy that can be given. A treatment that is selectively toxic to tumors would maximize the beneficial effects of therapy and minimize side effects, potentially enabling effective treatment to be administered. METHODS AND FINDINGS: We postulated that the tumor-tropic property of stem cells or progenitor cells could be exploited to selectively deliver a therapeutic gene to metastatic solid tumors, and that expression of an appropriate transgene at tumor loci might mediate cures of metastatic disease. To test this hypothesis, we injected HB1.F3.C1 cells transduced to express an enzyme that efficiently activates the anti-cancer prodrug CPT-11 intravenously into mice bearing disseminated neuroblastoma tumors. The HB1.F3.C1 cells migrated selectively to tumor sites regardless of the size or anatomical location of the tumors. Mice were then treated systemically with CPT-11, and the efficacy of treatment was monitored. Mice treated with the combination of HB1.F3.C1 cells expressing the CPT-11-activating enzyme and this prodrug produced tumor-free survival of 100% of the mice for >6 months (P<0.001 compared to control groups). CONCLUSIONS: The novel and significant finding of this study is that it may be possible to exploit the tumor-tropic property of stem or progenitor cells to mediate effective, tumor-selective therapy for metastatic tumors, for which no tolerated curative treatments are currently available
