This shows that students at the time of answering questions are still low. After giving the action during the post-test cycle I, the results obtained from 32 students were 14 students who completed or 43.59%, with a class average of 58.43, there was an increase in learning completeness in the initial test and post-test I of 27.97 %, and in the first cycle the maximum mastery of individual and classical learning outcomes has not been obtained, then proceed to the second cycle and obtained learning mastery, namely from 32 students there are 29 people who have completed learning or 90.62%, and an average of 79, 06, in the second cycle, students have completed to obtain good learning outcomes, so this research ends here
Ultra-Rare Variants Identify Biological Pathways and Candidate Genes in the Pathobiology of Non-Syndromic Cleft Palate Only
: In recent decades, many efforts have been made to elucidate the genetic causes of non-syndromic cleft palate (nsCPO), a complex congenital disease caused by the interaction of several genetic and environmental factors. Since genome-wide association studies have evidenced a minor contribution of common polymorphisms in nsCPO inheritance, we used whole exome sequencing data to explore the role of ultra-rare variants in this study. In a cohort of 35 nsCPO cases and 38 controls, we performed a gene set enrichment analysis (GSEA) and a hypergeometric test for assessing significant overlap between genes implicated in nsCPO pathobiology and genes enriched in ultra-rare variants in our cohort. GSEA highlighted an enrichment of ultra-rare variants in genes principally belonging to cytoskeletal protein binding pathway (Probability Density Function corrected p-value = 1.57 × 10-4); protein-containing complex binding pathway (p-value = 1.06 × 10-2); cell adhesion molecule binding pathway (p-value = 1.24 × 10-2); ECM-receptor interaction pathway (p-value = 1.69 × 10-2); and in the Integrin signaling pathway (p-value = 1.28 × 10-2). Two genes implicated in nsCPO pathobiology, namely COL2A1 and GLI3, ranked among the genes (n = 34) with nominal enrichment in the ultra-rare variant collapsing analysis (Fisher's exact test p-value < 0.05). These genes were also part of an independent list of genes highly relevant to nsCPO biology (n = 25). Significant overlap between the two sets of genes (hypergeometric test p-value = 5.86 × 10-3) indicated that enriched genes are likely to be implicated in physiological palate development and/or the pathological processes of oral clefting. In conclusion, ultra-rare variants collectively impinge on biological pathways crucial to nsCPO pathobiology and point to candidate genes that may contribute to the individual risk of disease. Sequencing can be an effective approach to identify candidate genes and pathways for nsCPO

Yang-Mills theory that one obtains via the generalisation of discrete
light-cone quantisation to singular homogeneous plane waves. The non-Abelian
nature of this theory is known to be important for physics near the
singularity, at least as far as the number of degrees of freedom is concerned.
We will show that the quartic interaction is always subleading as one
approaches the singularity and that close enough to t=0 the evolution is driven
by the diverging tachyonic mass term. The evolution towards asymptotically flat

Saponin Inhibits Hepatitis C Virus Propagation by Up-regulating Suppressor of Cytokine Signaling 2
Saponins are a group of naturally occurring plant glycosides which possess a wide range of pharmacological properties, including anti-tumorigenic and antiviral activities. To investigate whether saponin has anti-hepatitis C virus (HCV) activity, we examined the effect of saponin on HCV replication. HCV replication was efficiently inhibited at a concentration of 10 µg/ml of saponin in cell culture grown HCV (HCVcc)-infected cells. Inhibitory effect of saponin on HCV replication was verified by quantitative real-time PCR, reporter assay, and immunoblot analysis. In addition, saponin potentiated IFN-α-induced anti-HCV activity. Moreover, saponin exerted antiviral activity even in IFN-α resistant mutant HCVcc-infected cells. To investigate how cellular genes were regulated by saponin, we performed microarray analysis using HCVcc-infected cells. We demonstrated that suppressor of cytokine signaling 2 (SOCS2) protein level was distinctively increased by saponin, which in turn resulted in inhibition of HCV replication. We further showed that silencing of SOCS2 resurrected HCV replication and overexpression of SOCS2 suppressed HCV replication. These data imply that saponin inhibits HCV replication via SOCS2 signaling pathway. These findings suggest that saponin may be a potent therapeutic agent for HCV patients

Response of Upland Rice towards Biological Fertilizer on the Dry Land in South Konawe District, South East Sulawesi. As the productivity of upland rice is still low, it requires technology improvement such as high yield variety and biological fertilizer. The purpose of this research was to determine the adaptability of upland rice varieties combined with biological fertilizer application. The experiment was conducted in the dry land of South Konawe District, South East Sulawesi using a factorial randomized block design with four replications of two factors. The first factor was three upland rice varieties namely Inpago LIPI Go1, Inpago LIPI Go2 and local varieties Kolono; the second factor was the combination of inorganic fertilizers and biological fertilizers. The experiment was carried out from December 2013 to March 2014. The results showed that the highest productivity was achieved by Inpago LIPI Go2 (4.5 to 5.2 t/ha) combined with the fertilizer combination as follow; Biofertilizer; Biofertilizer + 25% of recommended inorganic fertilizer; and Biofertilizer + 50% of recommended inorganic fertilizer without loosing the yield
Epilepsy with auditory features: A heterogeneous clinico-molecular disease
Objective: To identify novel genes implicated in epilepsy with auditory features (EAF) in phenotypically heterogeneous families with unknown molecular basis. Methods: We identified 15 probands with EAF in whom an LGI1 mutation had been excluded. We performed electroclinical phenotyping on all probands and available affected relatives. We used whole-exome sequencing (WES) in 20 individuals with EAF (including all the probands and 5 relatives) to identify single nucleotide variants, small insertions/deletions, and copy number variants. Results: WES revealed likely pathogenic variants in genes that had not been previously associated with EAF: a CNTNAP2 intragenic deletion, 2 truncating mutations of DEPDC5, and a missense SCN1A change. Conclusions: EAF is a clinically and molecularly heterogeneous disease. The association of EAF with CNTNAP2, DEPDC5, and SCN1A mutations widens the phenotypic spectrum related to these genes. CNTNAP2 encodes CASPR2, a member of the voltage-gated potassium channel complex in which LGI1 plays a role. The finding of a CNTNAP2 deletion emphasizes the importance of this complex in EAF and shows biological convergence

Abstract
PURPOSE: SCN1A is the most clinically relevant epilepsy gene, most mutations lead to severe myoclonic epilepsy of infancy (SMEI) and generalized epilepsy with febrile seizures plus (GEFS+). We studied 132 patients with epilepsy syndromes with seizures precipitated by fever, and performed phenotype-genotype correlations with SCN1A alterations.
METHODS: We included patients with SMEI including borderline SMEI (SMEB), GEFS+, febrile seizures (FS), or other seizure types precipitated by fever. We performed a clinical and genetic study focusing on SCN1A, using dHPLC, gene sequencing, and MLPA to detect genomic deletions/duplications on SMEI/SMEB patients.
RESULTS: We classified patients as: SMEI/SMEB = 55; GEFS+= 26; and other phenotypes = 51. SCN1A analysis by dHPLC/sequencing revealed 40 mutations in 37 SMEI/SMEB (67%) and 3 GEFS+ (11.5%) probands. MLPA showed genomic deletions in 2 of 18 SMEI/SMEB. Most mutations were de novo (82%). SMEB patients carrying mutations (8) were more likely to have missense mutations (62.5%), conversely SMEI patients (31) had more truncating, splice site or genomic alterations (64.5%). SMEI/SMEB with truncating, splice site or genomic alterations had a significantly earlier age of onset of FS compared to those with missense mutations and without mutations (p = 0.00007, ANOVA test). None of the remaining patients with seizures precipitated by fever carried SCN1A mutations.
CONCLUSION: We obtained a frequency of 71%SCN1A abnormalities in SMEI/SMEB and of 11.5% in GEFS+ probands. MLPA complements DNA sequencing of SCN1A increasing the mutation detection rate. SMEI/SMEB with truncating, splice site or genomic alterations had a significantly earlier age of onset of FS. This study confirms the high sensitivity of SCN1A for SMEI/SMEB phenotypes
Photonic quantum state transfer between a cold atomic gas and a crystal
Interfacing fundamentally different quantum systems is key to build future
hybrid quantum networks. Such heterogeneous networks offer superior
capabilities compared to their homogeneous counterparts as they merge
individual advantages of disparate quantum nodes in a single network
architecture. However, only very few investigations on optical
hybrid-interconnections have been carried out due to the high fundamental and
technological challenges, which involve e.g. wavelength and bandwidth matching
of the interfacing photons. Here we report the first optical quantum
interconnection between two disparate matter quantum systems with photon
storage capabilities. We show that a quantum state can be faithfully
transferred between a cold atomic ensemble and a rare-earth doped crystal via a
single photon at telecommunication wavelength, using cascaded quantum frequency
conversion. We first demonstrate that quantum correlations between a photon and
a single collective spin excitation in the cold atomic ensemble can be
transferred onto the solid-state system. We also show that single-photon
time-bin qubits generated in the cold atomic ensemble can be converted, stored
and retrieved from the crystal with a conditional qubit fidelity of more than
. Our results open prospects to optically connect quantum nodes with
different capabilities and represent an important step towards the realization
of large-scale hybrid quantum networks
