In Times of Global Planetary Challenges: Metanoia
We grow up in a perspective of an existence, which we call reality. We identify our-selves with this reality as something absolute and we reject the experiences that do not belong to it. Each reality is valid only within certain limits (such as family’s traditions, religious beliefs, culture etc). So, “this is how our own state of consciousness perceives reality and we can never explore reality without exploring ourselves the reality that we explore” reminds us of Ram Dass. Therefore, the numerous crises that arise in our time, ecological, humanitarian, economic, religious, political, show us how we perceive our-selves: mostly as indifferent and without being aware of nature. This work leads us to deeply think about the fact, that as we are living in the Anthropocene era we can prac-tice a different committed science that helps us clarify and manifest proposals, that will help us to deal with this planetary crisis, from new perspectives or metanoia (from Greek μετανοιεν, metanoien a change of mind or direction, new mind).Fil: Castañeda Cataña, Mayra Alejandra. The omnipresent power of the invisible: From viruses to being conscious
According to Mayan cosmology, the law of time is represented as the energy factored by time equals to art. Time is a form of biological information. Time in-forms life, in such a way that life forms processes time as information and externalize it specific forms into the three-dimensional world, then time is the principle ordering of life. Indeed, everything in the natural order of the cosmos is beauty and harmony. All life forms on planet earth have their phases of morphological development, this is real even in the social structure of communities of living beings that also may have an aesthetic or artistic quality. This form and measurements of all things constitutes the entire or holistic order of the universe. So, if life is it nothing more than better-informed matter, where does this information come from? Socrates and Platon held that nothing in nature and in the world can be explained by random or chance, as Democritus would have argued, that nature creations occur because they have a Purpose. According to Platon, the natural world is a designer?s result demiurge or a universal consciousness that sets everything in the best possible place. So, if there is a start point where intelligence creates order, where there is order, there is purpose so; what is the purpose then? This work tends to give an answer and show a different perspective of living, studying and understanding (micro)biological phenomena in order to become conscious and assume that Nature is infinitely more powerful than us, the Whole is more than the sum of parts, we are part of it.Fil: Castañeda Cataña, Mayra Alejandra. Generating a lexicon of errors in Portuguese to support an error identification system for Spanish native learners
Portuguese is a less resourced language in what concerns foreign language learning. Aiming to inform a module of a system designed to support scientific written production of Spanish native speakers learning Portuguese, we developed an approach to automatically generate a lexicon of wrong words, reproducing language transfer errors made by such foreign learners. Each item of the artificially generated lexicon contains, besides the wrong word, the respective Spanish and Portuguese correct words. The wrong word is used to identify the interlanguage error and the correct Spanish and Portuguese forms are used to generate the suggestions. Keeping control of the correct word forms, we can provide correction or, at least, useful suggestions for the learners. We propose to combine two automatic procedures to obtain the error correction: i) a similarity measure and ii) a translation algorithm based on aligned parallel corpus. The similarity-based method achieved a precision of 52%, where as the alignment-based method achieved a precision of 90%. In this paper we focus only on interlanguage errors involving suffixes that have different forms in both languages. The approach, however, is very promising to tackle other types of errors, such as gender errors.
We report in this paper the development of a module dedicated to discourse markers in HABLA (Hispanofalantes Purchasing an Academic Lin-guistic Base), a tool designed to support native Spanish speakers in their aca-demic writing in Portuguese. HABLA is conceived to meet the needs of native Spanish speakers who are enrolled in Brazilian federal and state institutions and must write a dissertation or thesis in Portuguese. The diagnosis of difficulties faced by the learners in the use of discourse markers is based on the analysis of a learners’ corpus. Part of these difficulties are addressed by two procedures al-ready implemented that identify the problems automatically and present sugges-tions. The development of the module encompasses the compilation of a bilin-gual lexicon of discourse markers – Spanish-Portuguese - as well as a list of false friends discourse markers
Production of recombinant carbohydrate-binding modules in Pichia pastoris and their application in the modification of paper fibers
Dissertação de mestrado em BioengenhariaEnzymes have been widely applied in the paper industry for the modification of pulp and paper
properties. Although they have the potential to replace hazardous chemicals, their use in the
hydrolysis of polysaccharides reduces both fiber strength and mass. To overcome these negative
effects, the potential use of recombinant carbohydrate-binding modules (CBMs) in papermaking,
has been receiving growing attention. A CBM is defined as a contiguous amino-acid sequence
within a carbohydrate-active enzyme, presenting an independent fold and function, and
carbohydrate-binding activity. In addition, glycosylation in fungal CBMs has been suggested to be
important for the modification of pulp and paper properties
In order to improve cellulose fibers from the paper industry using an enzyme-free process,
consisting of recombinant CBMs, we aimed to: (1) clone and produce in Pichia pastoris
recombinant CBM1 from Trichoderma reesei (fungus) and CBM3 from Clostridium thermocellum
(bacteria), (2) optimize the production and purification of the recombinant CBMs by immobilized
metal ion affinity chromatography (IMAC) and (3) characterize the cellulose-binding capacity of
the recombinant CBMs and study their effects in the properties of pulp (drainability) and paper
fibers (wetability, bursting and tensile strengths, lengthening, tearing and air permeability).
Recombinant CBM1 fused to enhanced green fluorescent protein (eGFP), directly or by the native
glycosylated CBM linker, was produced at high levels but mainly as intracellular protein. Their
purification from P. pastoris cell-free extracts by IMAC resulted in low amounts, which
compromised their characterization and application. Nevertheless, the two purified recombinant
CBM-fusions were able to bind to cellulose. On the other hand, both recombinant versions of
CBM3, glycosylated and non-glycosylated, were produced as secreted proteins with the nonglycosylated
CBM3 exhibiting higher cellulose-binding affinity than the glycosylated version.
Regardless of the lack of effect on pulp drainability, both CBM3 versions improved significantly
the mechanical properties of Eucaliptus globulus papersheets, namely the burst and tensile
strength indexes, up to 12% and 10% (in papersheets made of E. globulus pulp treated with nonglycosylated
CBM3), respectively. In addition, non-glycosylated CBM3 decreased significantly the
wetability and air permeability of papersheets. However, both recombinant CBM3 versions did
not have any effect on papersheets made of a mixture of E. globulus and Pinus sylvestris (30:70)
pulp. The modification of paper fibers by recombinant CBM3 was dependent on its concentration
but not on its glycosylation. These results indicate that recombinant CBM3 play an important role
in the improvement of surface/interface properties of cellulose fibers, contributing to paper
products with higher quality.
papel.
No
entanto, as proteínas de fusão eGFP-CBM purificadas apresentaram capacidade de ligação à
celulose.
Além disso, a versão
Fundação para a Ciência e Tecnologia (FCT) - Project GlycoCBMs PTDC/AGR-FOR/3090/2012 – FCOMP-01-0124-FEDER-02794
