Whereas the neutral systems remain planar in this size range, a 2D-3D transition occurs at Ag5+ for the cationic system and at Ag42+ for the dicationic system. Neutral silver clusters can bind with nitrogen 3 or with the pi system of the base. However, positively charged clusters interact with nitrogen 7 and the neighboring carbonyl group. Thus, the cationic silver-DNA clusters present experimentally may preferentially interact at these sites. © 2017 Author(s)

    The economic implications of introducing new technologies under rainfed farming conditions in Sri Lanka : a case study

    Timing of cultural practices and income distribution throughout the year were considered to be the key factors in the success of rainfed farming. It was felt that new technologies for labour saving at peak periods and a crop that could be harvested during the off period are useful approaches to improving the production capacity of rainfed farms. This study aims to assess the economic benefits of new technologies (NTs) developed for rainfed farms in a range of practical farm situations. A case study approach was employed in exploring this situation. Two sample farmers were selected from a rainfed project conducted in the dry zone of Sri Lanka. Whole farm planning based on Linear Programming (LP) was employed. Essentially, it looks at the farm-household as a whole entity with a common resource base, interrelated with the various activities of the farmer. This was demonstrated using only the historical technologies. The optimal plan generated specified a combination of alternative enterprises subject to a restricted resource base. The impact of introducing NTs was assessed by comparing the previous solution which employs only historical technologies with the new situation. That is, new technologies to be evaluated are included in the farmers' options along with the historical technologies.     A scorpion species proved to be lethal to humans was recently recorded from Jaffna Peninsula (9°40\u270\u27\u27N 80°0\u270\u27\u27E, mean annual temperature 26.2°C), in the northern dry zone of Sri Lanka. This species is morphologically different from all other known scorpions in Sri Lanka. The species was identified as Hottentotta tamulus (Scorpiones: Buthidae), which is commonly found in Maharashtra, India, the closest mainland to Sri Lanka. Small children and housewives were most of the victims. Soon after sting, the patient develops intense pain at the site of sting followed by numbed sensation. Dominant clinical effects include excessive sweating, agitation and palpitation. Blood pressure of the victim goes up, and if not promptly treated leads to acute heart failure. There is a high risk of spreading of this species to the rest of the country due to transport of goods and sand from the area

    Assessing soil erosion hazards using land-use change and landslide frequency ratio method: A case study of Sabaragamuwa province, Sri Lanka

This study aims to identify the vulnerable landscape areas using landslide frequency ratio and land-use change associated soil erosion hazard by employing geo-informatics techniques and the revised universal soil loss equation (RUSLE) model. Required datasets were collected from multiple sources, such as multi-temporal Landsat images, soil data, rainfall data, land-use land-cover (LULC) maps, topographic maps, and details of the past landslide incidents. Landsat satellite images from 2000, 2010, and 2019 were used to assess the land-use change. Geospatial input data on rainfall, soil type, terrain characteristics, and land cover were employed for soil erosion hazard classification and mapping. Landscape vulnerability was examined on the basis of land-use change, erosion hazard class, and landslide frequency ratio. Then the erodible hazard areas were identified and prioritized at the scale of river distribution zones. The image analysis of Sabaragamuwa Province in Sri Lanka from 2000 to 2019 indicates a significant increase in cropping areas (17.96%) and urban areas (3.07%), whereas less dense forest and dense forest coverage are significantly reduced (14.18% and 6.46%, respectively). The average annual soil erosion rate increased from 14.56 to 15.53 t/ha/year from year 2000 to 2019. The highest landslide frequency ratios are found in the less dense forest area and cropping area, and were identified as more prone to future landslides. The river distribution zones Athtanagalu Oya (A-2), Kalani River-south (A-3), and Kalani River-north (A-9), were identified as immediate priority areas for soil conservation

    A Review on Assessing and Mapping Soil Erosion Hazard Using Geo-Informatics Technology for Farming System Management

    Soil erosion is a severe threat to food production systems globally. Food production in farming systems decreases with increasing soil erosion hazards. This review article focuses on geo-informatics applications for identifying, assessing and predicting erosion hazards for sustainable farming system development. Several researchers have used a variety of quantitative and qualitative methods with erosion models, integrating geo-informatics techniques for spatial interpretations to address soil erosion and land degradation issues. The review identified different geo-informatics methods of erosion hazard assessment and highlighted some research gaps that can provide a basis to develop appropriate novel methodologies for future studies. It was found that rainfall variation and land-use changes significantly contribute to soil erosion hazards. There is a need for more research on the spatial and temporal pattern of water erosion with rainfall variation, innovative techniques and strategies for landscape evaluation to improve the environmental conditions in a sustainable manner. Examining water erosion and predicting erosion hazards for future climate scenarios could also be approached with emerging algorithms in geo-informatics and spatiotemporal analysis at higher spatial resolutions. Further, geo-informatics can be applied with real-time data for continuous monitoring and evaluation of erosion hazards to risk reduction and prevent the damages in farming systems.</jats:p

    Novel role of extracellular matrix protein 1 (ECM1) in cardiac aging and myocardial infarction

Introduction The prevalence of heart failure increases in the aging population and following myocardial infarction (MI), yet the extracellular matrix (ECM) remodeling underpinning the development of aging- and MI-associated cardiac fibrosis remains poorly understood. A link between inflammation and fibrosis in the heart has long been appreciated, but has mechanistically remained undefined. We investigated the expression of a novel protein, extracellular matrix protein 1 (ECM1) in the aging and infarcted heart. Methods Young adult (3-month old) and aging (18-month old) C57BL/6 mice were assessed. Young mice were subjected to left anterior descending artery-ligation to induce MI, or transverse aortic constriction (TAC) surgery to induce pressure-overload cardiomyopathy. Left ventricle (LV) tissue was collected early and late post-MI/TAC. Bone marrow cells (BMCs) were isolated from young healthy mice, and subject to flow cytometry. Human cardiac fibroblast (CFb), myocyte, and coronary artery endothelial & smooth muscle cell lines were cultured; human CFbs were treated with recombinant ECM1. Primary mouse CFbs were cultured and treated with recombinant angiotensin-II or TGF-β1. Immunoblotting, qPCR and mRNA fluorescent in-situ hybridization (mRNA-FISH) were conducted on LV tissue and cells. Results ECM1 expression was upregulated in the aging LV, and in the infarct zone of the LV early post-MI. No significant differences in ECM1 expression were found late post-MI or at any time-point post-TAC. ECM1 was not expressed in any resident cardiac cells, but ECM1 was highly expressed in BMCs, with high ECM1 expression in granulocytes. Flow cytometry of bone marrow revealed ECM1 expression in large granular leucocytes. mRNA-FISH revealed that ECM1 was indeed expressed by inflammatory cells in the infarct zone at day-3 post-MI. ECM1 stimulation of CFbs induced ERK1/2 and AKT activation and collagen-I expression, suggesting a pro-fibrotic role. Conclusions ECM1 expression is increased in ageing and infarcted hearts but is not expressed by resident cardiac cells. Instead it is expressed by bone marrow-derived granulocytes. ECM1 is sufficient to induce cardiac fibroblast stimulation in vitro. Our findings suggest ECM1 is released from infiltrating inflammatory cells, which leads to cardiac fibroblast stimulation and fibrosis in aging and MI. ECM1 may be a novel intermediary between inflammation and fibrosis

    A Pan-Dengue Virus Reverse Transcription-Insulated Isothermal PCR Assay Intended for Point-of-Need Diagnosis of Dengue Virus Infection by Use of the POCKIT Nucleic Acid Analyzer

    Dengue virus (DENV) infection is considered a major public health problem in developing tropical countries where the virus is endemic and continues to cause major disease outbreaks every year. Here, we describe the development of a novel, inexpensive, and user-friendly diagnostic assay based on a reverse transcription-insulated isothermal PCR (RT-iiPCR) method for the detection of all four serotypes of DENV in clinical samples. The diagnostic performance of the newly established pan-DENV RT-iiPCR assay targeting a conserved 3′ untranslated region of the viral genome was evaluated. The limit of detection with a 95% confidence was estimated to be 10 copies of in vitro-transcribed (IVT) RNA. Sensitivity analysis using RNA prepared from 10-fold serial dilutions of tissue culture fluid containing DENVs suggested that the RT-iiPCR assay was comparable to the multiplex real-time quantitative RT-PCR (qRT-PCR) assay for DENV-1, -3, and -4 detection but 10-fold less sensitive for DENV-2 detection. Subsequently, plasma collected from patients suspected of dengue virus infection (n = 220) and individuals not suspected of dengue virus infection (n = 45) were tested by the RT-iiPCR and compared to original test results using a DENV NS1 antigen rapid test and the qRT-PCR. The diagnostic agreement of the pan-DENV RT-iiPCR, NS1 antigen rapid test, and qRT-PCR tests was 93.9%, 84.5%, and 97.4%, respectively, compared to the composite reference results. This new RT-iiPCR assay along with the portable POCKIT nucleic acid analyzer could provide a highly reliable, sensitive, and specific point-of-need diagnostic assay for the diagnosis of DENV in clinics and hospitals in developing countries

    Prevalence and risk factors for impaired kidney function in the district of Anuradhapura, Sri Lanka: a cross-sectional population-representative survey in those at risk of chronic kidney disease of unknown aetiology

    BACKGROUND: Over the last 20 years there have been reports of a form of chronic kidney disease of unknown cause (CKDu) affecting rural communities in the North Central Province of Sri Lanka. Valid prevalence estimates, using a standardised methodology, are needed to assess the burden of disease, assess secular trends, and perform international comparisons. METHODS: We conducted a cross-sectional representative population survey in five study areas with different expected prevalences of CKDu. We used a proxy definition of CKDu involving a single measure of impaired kidney function (eGFR< 60 mL/min/1.7m2, using the CKD-Epi formula) in the absence of hypertension, diabetes or heavy proteinuria. RESULTS: A total of 4803 participants (88.7%) took part in the study and 202 (6.0%; 95% CI 5.2-6.8) had a low eGFR in the absence of hypertension, diabetes and heavy proteinuria and hence met the criteria for proxy CKDu. The proportion of males (11.2%; 95% CI 9.2-13.1) were triple than the females (3.7%; 95% CI 2.9-4.5). Advancing age and history of CKD among parents or siblings were risk factors for low GFR among both males and females while smoking was found to be a risk factor among males. CONCLUSIONS: These data, collected using a standardised methodology demonstrate a high prevalence of impaired kidney function, not due to known causes of kidney disease, in the selected study areas of the Anuradhapura district of Sri Lanka. The aetiology of CKDu in Sri Lanka remains unclear and there is a need for longitudinal studies to describe the natural history and to better characterise risk factors for the decline in kidney function
