    Demographic effects on fruit set in the dioecious shrub Canada buffaloberry (Shepherdia canadensis)

    The effects of pollen limitation on reproductive success in plants have been well-documented using pollen supplementation experiments. However, the role of local demographics in determining pollen limitation, particularly in terms of the additive and interactive effects of pollen availability and competition are not well known. We measured fruit set in the dioecious shrub Canada buffaloberry (Shepherdia canadensis) in Central Alberta, Canada to evaluate whether local demographics measured at three spatial scales (25, 50, and 100 m2) affect fruit set in buffaloberry. We test whether density-dependence (population density), pollen donor (measured as male density, distance to nearest male plant and size of nearest male plant), female competitor (measured as female density and distance to nearest female plant), or the combined pollen donor and competitor hypotheses best explain natural variations in fruit set for a population of Canada buffaloberry. Support was highest for the combined pollen donor and competitor hypothesis at an intermediate spatial scale of 50 m2. Proportion fruit set increased with male shrub density (pollen donors) and decreased with female shrub density (pollen competitors), but was more affected by the presence of males than females. This illustrates that access to male shrubs within a 3.99 m radius affects pollen availability, while nearby females compete intra-specifically for pollen

    Live-birth rate associated with repeat in vitro fertilization treatment cycles

Importance The likelihood of achieving a live birth with repeat in vitro fertilization (IVF) is unclear, yet treatment is commonly limited to 3 or 4 embryo transfers. Objective To determine the live-birth rate per initiated ovarian stimulation IVF cycle and with repeated cycles. Design, Setting, and Participants Prospective study of 156 947 UKwomen who received 257 398 IVF ovarian stimulation cycles between 2003 and 2010 and were followed up until June 2012. Exposures In vitro fertilization, with a cycle defined as an episode of ovarian stimulation and all subsequent separate fresh and frozen embryo transfers. Main Outcomes and Measures Live-birth rate per IVF cycle and the cumulative live-birth rates across all cycles in all women and by age and treatment type. Optimal, prognosis-adjusted, and conservative cumulative live-birth rates were estimated, reflecting 0%, 30%, and 100%, respectively, of women who discontinued due to poor prognosis and having a live-birth rate of 0 had they continued. Results Among the 156 947 women, the median age at start of treatment was 35 years (interquartile range, 32-38; range, 18-55), and the median duration of infertility for all 257 398 cycles was 4 years (interquartile range, 2-6; range

    Regression discontinuity analysis for pharmacovigilance:statin example reflected trial findings showing little evidence of harm

    OBJECTIVES: The study aims to explore the use of regression discontinuity analysis (RDA) to examine effects of prescription of statins on total cholesterol and adverse outcomes (type 2 diabetes, rhabdomyolysis and myopathy, myalgia and myositis, liver disease, CVD, and mortality). STUDY DESIGN AND SETTING: We conducted a prospective cohort study using the Clinical Practice Research Datalink including patients with QRISK scores of 10 to 30 in 2010 to 2013 who were last followed-up in October 2016. Comparing patients with QRISK≥20 and QRISK<20, we explored RDA assumptions, provided proof of concept analyses (total cholesterol as outcome), and investigated the effect of statins prescription on adverse outcomes. RESULT: RDA confirmed statin prescription reduced total cholesterol (Mean difference (MD) -1.33 mmol/L, 95%Confidence Interval (CI) -1.93 to -0.73). RDA provided little evidence for adverse effects on diabetes, myalgia and myositis, liver disease, CVD, or mortality. The RDA analysis findings are similar to RCT results. Findings from non-RDA analysis agree with published observational studies. CONCLUSION: RDA can be used with large routine clinical datasets to provide evidence on effects of medications which are prescribed according to a threshold. Testable RDA assumptions were satisfied, but confidence intervals were wide, partly due to the low compliance with the prescribing threshold

    Drivers of understory plant communities in Sierra Nevada mixed conifer forests with pyrodiversity

    Background: Fire suppression in western North America increased and homogenized overstory cover in conifer forests, which likely affected understory plant communities. We sought to characterize understory plant communities and their drivers using plot-based observations from two contemporary reference sites in the Sierra Nevada, USA. These sites had long-established natural fire programs, which have resulted in restored natural fire regimes. In this study, we investigated how pyrodiversity—the diversity of fire size, severity, season, and frequency—and other environment factors influenced species composition and cover of forest understory plant communities. Results: Understory plant communities were influenced by a combination of environmental, plot-scale recent fire history, and plot-neighborhood pyrodiversity within 50 m. Canopy cover was inversely proportional to understory plant cover, Simpson’s diversity, and evenness. Species richness was strongly influenced by the interaction of plot-based fire experience and plot-neighborhood pyrodiversity within 50 m. Conclusions: Pyrodiversity appears to contribute both directly and indirectly to diverse understory plant communities in Sierra Nevada mixed conifer forests. The indirect influence is mediated through variability in tree canopy cover, which is partially related to variation in fire severity, while direct influence is an interaction between local and neighborhood fire activity
