In rural speech communities, where locals know each other, dialectal and unofficial names in which the surname precedes the first name (der Müller Peter, (s) Müllers Peter) are used to refer to absent referents. The occurrence of these forms is highly pragmatic and determined by the degree of familiarity of referent, speaker and hearer. Also, they are only used within the respective speech communities. It is all the more surprising to find these reference forms in internet forums, social media and online newspapers, where they refer to prominent figures. This study explores diatopic as well as diaphasic variation of the structure surname + first name in the German webcorpus DECOW16b, and shows that while, especially referring to Bavarian celebrities, it is used to express familiarity with the referents and to present them in private rather than professional contexts, it can also mark depreciation and a denial of professionality. The latter applies to politicians in particular, while sportspersons are generally depicted positively. The use of dialectal register on the internet can be interpreted as a form of enregisterment, transferring the most prominent pragmatic conditions for surname + first name in dialectal registers to new contexts. In rural speech communities, when speakers refer to persons, their family of origin is omnipresent, not only by passing on the name of the family patriarch, but also in the serialization of surname and first name and in the grammatical structures of reference forms (der Müller Peter, s Müllers Peter). This paper portrays the diachronic development of reference forms and elaborates on their preservation in synchronic reference systems. Furthermore, it explores a referent’s linguistic categorization into social groups and shows that different grammatical structures foreground certain distinctions, which can also overlap. In rural villages, inofficial house names, derived from the patriarch’s first name or profession, are still in use. When an individual marries into another house, the reference to this person can change accordingly

Isoflavone glycosides: Synthesis and evaluation as α-glucosidase inhibitors

On the basis of the structure of 4′,7,8-trihydroxyisoflavone 7-O-α-D-arabinofuranoside (namely A-76202, 1), a Rhodococcus metabolite showing potent inhibitory activities against the α-glucosidases of rat liver microsome (IC 50 = 0.46 ng/mL), 26 analogs, each with minor variations at the sugar moiety and the isoflavone A and B rings, were readily synthesized. Notably, a new and efficient method was developed for the divergent synthesis of the B-ring congeners of the isoflavone glycosides by using Suzuki-Miyaura coupling as the final step. Modifications at the sugar moiety and the isoflavone A ring significantly diminish the activity, whereas variations at the B ring are largely tolerated for retaining the potent α-glucosidase inhibitory activity. © Wiley-VCH Verlag GmbH & Co. Identification and Validation of Novel Cerebrospinal Fluid Biomarkers for Staging Early Alzheimer's Disease

Ideally, disease modifying therapies for Alzheimer disease (AD) will be applied during the 'preclinical' stage (pathology present with cognition intact) before severe neuronal damage occurs, or upon recognizing very mild cognitive impairment. Developing and judiciously administering such therapies will require biomarker panels to identify early AD pathology, classify disease stage, monitor pathological progression, and predict cognitive decline. To discover such biomarkers, we measured AD-associated changes in the cerebrospinal fluid (CSF) proteome.CSF samples from individuals with mild AD (Clinical Dementia Rating [CDR] 1) (n = 24) and cognitively normal controls (CDR 0) (n = 24) were subjected to two-dimensional difference-in-gel electrophoresis. Within 119 differentially-abundant gel features, mass spectrometry (LC-MS/MS) identified 47 proteins. For validation, eleven proteins were re-evaluated by enzyme-linked immunosorbent assays (ELISA). Six of these assays (NrCAM, YKL-40, chromogranin A, carnosinase I, transthyretin, cystatin C) distinguished CDR 1 and CDR 0 groups and were subsequently applied (with tau, p-tau181 and Aβ42 ELISAs) to a larger independent cohort (n = 292) that included individuals with very mild dementia (CDR 0.5). Receiver-operating characteristic curve analyses using stepwise logistic regression yielded optimal biomarker combinations to distinguish CDR 0 from CDR>0 (tau, YKL-40, NrCAM) and CDR 1 from CDR<1 (tau, chromogranin A, carnosinase I) with areas under the curve of 0.90 (0.85-0.94 95% confidence interval [CI]) and 0.88 (0.81-0.94 CI), respectively.Four novel CSF biomarkers for AD (NrCAM, YKL-40, chromogranin A, carnosinase I) can improve the diagnostic accuracy of Aβ42 and tau. Together, these six markers describe six clinicopathological stages from cognitive normalcy to mild dementia, including stages defined by increased risk of cognitive decline. Such a panel might improve clinical trial efficiency by guiding subject enrollment and monitoring disease progression. Further studies will be required to validate this panel and evaluate its potential for distinguishing AD from other dementing conditions
