Development of an ultrasonically excited recoating process in laser powder bed fusion to process non-spreadable 316L powder
Part quality in laser powder bed fusion is influenced by the uniformity and density of the powder layer. As a result, requirements for the powder regarding spreadability and flowability are restrictive. Many researchers reported recoating defects for cohesive and agglomerating powders. However, the processing of such powders is highly desirable since e.g. surface roughness can be reduced. Therefore, we propose a novel ultrasonically excited recoater system to process such powder. In order to qualify the new recoater system, five different geometries with various amplitudes and recoating velocities were evaluated and compared to a conventional system. The defect rate, determined with a camera and AI-based analysis, the deposited mass and segregation along the recoated distance and the powder layer density of the resulting powder bed were analyzed. A process window and optimal geometry could be identified. It was possible to reduce segregation to a minimum, while maintaining a smooth, dense layer
Impact of axial active magnetic bearing stiffness coefficient on resonance frequencies of reaction wheel rotor
Разработана математическая модель системы «ротор - электромагнитные подшипники» для электродвигателя-маховика системы ориентации и стабилизации космического аппарата. Модель учитывает собственные частоты изгибных колебаний ротора и коэффициенты жесткости электромагнитных подшипников. Предложен способ повышения угловой жесткости системы путем применения многополюсного осевого электромагнитного подшипника и рассмотрено влияние его коэффициента жесткости на собственные частоты системы.The paper presents the mathematical model of «rotor - active magnetic bearings» system for reaction wheel used in spacecraft attitude control system. Developed model consider the natural frequencies of rotor bending oscillations and stiffness parameters of electromagnetic bearing. Method of angular stiffness increasing by using multipolar axial magnetic bearing is suggested and the results of impact analysis of multipolar axial magnetic bearing stiffness on resonance frequencies of system is considered

Análise de timol em cera de abelha por micro-extracção em fase sólida (SPME)
A aplicação contínua de acaricídas lipofílicos sintéticos no tratamento das
abelhas conduz a uma acumulação que depende da frequência, lipofilicidade e
quantidade de princípio activo utilizada. Este efeito é mais acentuado na cera
de abelha que no mel, no entanto, e porque a persistência destes resíduos é
elevada, provoca o aparecimento de resistências e a perda do seu efeito
acaricida.[1] Esta razão levou à pesquisa de outros compostos alternativos não
tóxicos e não persistentes, com efeito sobre o ácaro das abelhas, Varroa
Jacobsoni. Entre estes compostos encontra-se o timol, um composto fenólico,
volátil, presente no tomilho. Dos diversos componentes dos óleos essenciais
este é sem dúvida o que demonstrou maior efeito acaricida, utilizando-se no
tratamento das abelhas directamente ou como componente de diversas
formulações.[2] Em Portugal, foi introduzido muito recentemente sob a forma
comercial de APIGUARD: um gel, à base de timol, que controla termicamente a
libertação do princípio activo.
O controlo dos resíduos de timol na cera de abelha e no mel é assim um
desafio actual quer do ponto de vista sanitário quer de qualidade alimentar.
A micro-extracção em fase sólida (SPME) é uma técnica de preparação de
amostras que se baseia na sorção de analítos no revestimento de uma fibra de
sílica fundida e posterior desorção térmica no injector de um cromatógrafo em
fase gasosa (GC). Para além de combinar num único processo etapas de
extracção, purificação e concentração dos analitos, a técnica de SPME
apresenta uma série de vantagens relativamente às técnicas de extracção
convencionais, como a extracção líquido-líquido e extracção em fase sólida,
nomeadamente a sua relativa simplicidade e rapidez, reduzido custo e não
utilização de solventes para a extracção de analitos, para além de permitir a
extracção por imersão directa na amostra gasosa ou líquida e extracção por
amostragem do espaço-de-cabeça da amostra líquida ou sólida.[3] Ao contrário
das técnicas tradicionais, que permitem uma extracção quantitativa dos
analitos, a técnica de SPME baseia-se num equilíbrio de partição do analito.
Esta particularidade torna a técnica de SPME bastante sensível a parâmetros
experimentais que possam afectar os coeficientes de partição dos analitos e,
consequentemente, a sensibilidade e reprodutibilidade dos resultados.[4]
O objectivo deste trabalho é o desenvolvimento de uma metodologia para a
análise de timol em ceras contaminadas, utilizando como padrão interno a
benzofenona. Em primeiro lugar, procedeu-se à optimização da técnica através
da determinação da quantidade de cera, temperatura de análise e período de
contacto da fibra com o espaço-de-cabeça da amostra mais adequados para o
caso em estudo. Numa segunda fase, procedeu-se à análise de diversas
lâminas de cera contaminadas propositadamente com timol e sujeitas a
diferentes condições de armazenamento: em frio, ao ar e em estufa.
Finalmente, procedeu-se à construção da curva de calibração e quantificação
do timol presente nas diversas amostras de cera analisadas.
Considerando-se os resultados, para os níveis de contaminação avaliados, as
condições analíticas mais adequadas ocorrem com a utilização de 1 g de cera,
mantendo-se a fibra em contacto com o espaço-de-cabeça durante 40 minutos
a uma temperatura de 60 ºC. Nestas condições experimentais foi possível obter
uma boa correlação linear (r2=0,990) no intervalo de concentrações [3,5-14
mg/g]. A quantidade de timol encontrada nas amostras é significativamente
inferior à colocada durante o processo de fabrico das lâminas, pelo que o
processo de conservação não é o mais adequado, sendo evidente uma menor
quantidade de timol quando a lâmina de cera é colocada na estufa
Imbalanced gut microbiota fuels hepatocellular carcinoma development by shaping the hepatic inflammatory microenvironment
Hepatocellular carcinoma (HCC) is a leading cause of cancer-related deaths worldwide, and therapeutic options for advanced HCC are limited. Here, we observe that intestinal dysbiosis affects antitumor immune surveillance and drives liver disease progression towards cancer. Dysbiotic microbiota, as seen in Nlrp6(-/-) mice, induces a Toll-like receptor 4 dependent expansion of hepatic monocytic myeloid-derived suppressor cells (mMDSC) and suppression of T-cell abundance. This phenotype is transmissible via fecal microbiota transfer and reversible upon antibiotic treatment, pointing to the high plasticity of the tumor microenvironment. While loss of Akkermansia muciniphila correlates with mMDSC abundance, its reintroduction restores intestinal barrier function and strongly reduces liver inflammation and fibrosis. Cirrhosis patients display increased bacterial abundance in hepatic tissue, which induces pronounced transcriptional changes, including activation of fibro-inflammatory pathways as well as circuits mediating cancer immunosuppression. This study demonstrates that gut microbiota closely shapes the hepatic inflammatory microenvironment opening approaches for cancer prevention and therapy. Steatohepatitis is a chronic hepatic inflammation associated with increased risk of hepatocellular carcinoma progression. Here the authors show that intestinal dysbiosis in mice lacking the inflammasome sensor molecule NLRP6 aggravates steatohepatitis and accelerates liver cancer progression, a process that can be delayed by antibiotic treatment.Peer reviewe
EPA guidance on physical activity as a treatment for severe mental illness: a meta-review of the evidence and Position Statement from the European Psychiatric Association (EPA), supported by the International Organization of Physical Therapists in Mental Health (IOPTMH)
Physical activity (PA) may be therapeutic for people with severe mental illness (SMI) who generally have low PA and experience numerous life style-related medical complications. We conducted a meta-review of interventions and their impact on health outcomes for people with SMI, including schizophrenia-spectrum disorders, major depressive disorder (MDD) and bipolar disorder. We searched major electronic databases until January 2018 for systematic reviews with/without meta-analysis that investigated PA for any SMI. We rated the quality of studies with the AMSTAR tool, grading the quality of evidence, and identifying gaps, future research needs and clinical practice recommendations. For MDD, consistent evidence indicated that PA can improve depressive symptoms versus control conditions, with effects comparable to those of antidepressants and psychotherapy. PA can also improve cardiorespiratory fitness and quality of life in people with MDD, although the impact on physical health outcomes was limited. There were no differences in adverse events versus control conditions. For MDD, larger effect sizes were seen when PA was delivered at moderate-vigorous intensity and supervised by an exercise specialist. For schizophrenia-spectrum disorders, evidence indicates that aerobic PA can reduce psychiatric symptoms, improves cognition and various subdomains, cardiorespiratory fitness, whilst evidence for the impact on anthropometric measures was inconsistent. There was a paucity of studies investigating PA in bipolar disorder, precluding any definitive recommendations. No cost effectiveness analyses in any SMI condition were identified. We make multiple recommendations to fill existing research gaps and increase the use of PA in routine clinical care aimed at improving psychiatric and medical outcomes
Understanding the biases to sepsis surveillance and quality assurance caused by inaccurate coding in administrative health data
Purpose
Timely and accurate data on the epidemiology of sepsis are essential to inform policy decisions and research priorities. We aimed to investigate the validity of inpatient administrative health data (IAHD) for surveillance and quality assurance of sepsis care.
Methods
We conducted a retrospective validation study in a disproportional stratified random sample of 10,334 inpatient cases of age ≥ 15 years treated in 2015–2017 in ten German hospitals. The accuracy of coding of sepsis and risk factors for mortality in IAHD was assessed compared to reference standard diagnoses obtained by a chart review. Hospital-level risk-adjusted mortality of sepsis as calculated from IAHD information was compared to mortality calculated from chart review information.
Results
ICD-coding of sepsis in IAHD showed high positive predictive value (76.9–85.7% depending on sepsis definition), but low sensitivity (26.8–38%), which led to an underestimation of sepsis incidence (1.4% vs. 3.3% for severe sepsis-1). Not naming sepsis in the chart was strongly associated with under-coding of sepsis. The frequency of correctly naming sepsis and ICD-coding of sepsis varied strongly between hospitals (range of sensitivity of naming: 29–71.7%, of ICD-diagnosis: 10.7–58.5%). Risk-adjusted mortality of sepsis per hospital calculated from coding in IAHD showed no substantial correlation to reference standard risk-adjusted mortality (r = 0.09).
Conclusion
Due to the under-coding of sepsis in IAHD, previous epidemiological studies underestimated the burden of sepsis in Germany. There is a large variability between hospitals in accuracy of diagnosing and coding of sepsis. Therefore, IAHD alone is not suited to assess quality of sepsis care
Blood pressure control in chronic kidney disease: A cross-sectional analysis from the German Chronic Kidney Disease (GCKD) study
We assessed the prevalence, awareness, treatment and control of hypertension in patients with moderate chronic kidney disease (CKD) under nephrological care in Germany. In the German Chronic Kidney Disease (GCKD) study, 5217 patients under nephrology specialist care were enrolled from 2010 to 2012 in a prospective observational cohort study. Inclusion criteria were an estimated glomerular filtration rate (eGFR) of 30 +/- 60 mL/min/1.73 m 2 or overt proteinuria in the presence of an eGFR> 60 mL/min/1.73 m 2. Office blood pressure was measured by trained study personnel in a standardized way and hypertension awareness and medication were assessed during standardized interviews. Blood pressure was considered as controlled if systolic 90%. However, only 2456 (49.3%) of the hypertensive patients had controlled blood pressure. About half (51.0%) of the patients with uncontrolled blood pressure met criteria for resistant hypertension. Factors associated with better odds for controlled blood pressure in multivariate analyses included younger age, female sex, higher income, low or absent proteinuria, and use of certain classes of antihypertensive medication. We conclude that blood pressure control of CKD patients remains challenging even in the setting of nephrology specialist care, despite high rates of awareness and medication use
Phosphatidylserine-exposing extracellular vesicles in body fluids are an innate defence against apoptotic mimicry viral pathogens
Some viruses are rarely transmitted orally or sexually despite their presence in saliva, breast milk, or semen. We previously identified that extracellular vesicles (EVs) in semen and saliva inhibit Zika virus infection. However, the antiviral spectrum and underlying mechanism remained unclear. Here we applied lipidomics and flow cytometry to show that these EVs expose phosphatidylserine (PS). By blocking PS receptors, targeted by Zika virus in the process of apoptotic mimicry, they interfere with viral attachment and entry. Consequently, physiological concentrations of EVs applied in vitro efficiently inhibited infection by apoptotic mimicry dengue, West Nile, Chikungunya, Ebola and vesicular stomatitis viruses, but not severe acute respiratory syndrome coronavirus 2, human immunodeficiency virus 1, hepatitis C virus and herpesviruses that use other entry receptors. Our results identify the role of PS-rich EVs in body fluids in innate defence against infection via viral apoptotic mimicries, explaining why these viruses are primarily transmitted via PS-EV-deficient blood or blood-ingesting arthropods rather than direct human-to-human contact
