    Comparing Clinical Profiles in Alzheimer\u27s Disease and Parkinson\u27s Disease Dementia

    BACKGROUND: Greater understanding of differences in baseline impairment and disease progression in patients with Alzheimer\u27s disease (AD) and Parkinson\u27s disease dementia (PDD) may improve the interpretation of drug effects and the design of future studies. METHODS: This was a retrospective analysis of three randomized, double-blind rivastigmine databases (one in PDD, two in AD). Impairment on the Alzheimer\u27s Disease Assessment Scale-cognitive subscale (ADAS-cog), Alzheimer\u27s Disease Cooperative Study-Activities of Daily Living (ADCS-ADL) scale, 10-item Neuropsychiatric Inventory (NPI-10) and the ADCS-Clinical Global Impression of Change (CGIC) was compared [standardized difference (Cohen\u27s d), similar if \u3c0.1]. RESULTS: Patients with AD or PDD had similar levels of impairment on the ADAS-cog and NPI-10. Scores on the ADCS-ADL scale (standardized difference = 0.47) and the ADAS-cog memory domain (total, 0.33; items, 0.10-0.58) were higher in AD; PDD patients were more impaired in the language (0.23) and praxis (0.34) domains. AD patients receiving placebo showed greater deterioration on the ADAS-cog (0.14) and improvement on the NPI-10 (0.11) compared with patients with PDD. CONCLUSION: Differing patterns of impairment occur in AD and PDD

    Phosphorus fractions in apple orchards in southern Brazil

    Phosphorus (P) applications at dosages higher than the necessary may cause P buildup in the soil labile fractions and, consequently, pollution of water sources. This study aimed to assess accumulation of P fractions as well as the parameters of adsorption isotherms in soil profiles having a history of application of phosphate fertilizers and cultivated with apple trees. Soil samples were collected from an area with no history of cultivation and 2 apple orchards cultivated for 8 (P8) and 18 (P18) years, in the city of Urubici, Santa Catarina, southern Brazil. Soil samples were air dried, sieved in a 2-mm mesh, and subjected to chemical analyses (P adsorption) besides P chemical fractionation. Applications of phosphate fertilizers to the orchards, particularly P18, increased the inorganic and organic concentrations of P in all fractions. The distribution of P in organic and inorganic forms in the planted soils was similar to the distribution observed for the native vegetation soil. The highest inorganic labile P fraction, the lowest maximum P adsorption capacity, and the highest equilibrium concentration values at P18 are indicators of a higher environmental pollution risk of the orchards soils compared to the native soil

    Desenvolvimento rural sustentável na região do município de Ibirubá- RS

    Trabalho apresentado no 31º SEURS - Seminário de Extensão Universitária da Região Sul, realizado em Florianópolis, SC, no período de 04 a 07 de agosto de 2013 - Universidade Federal de Santa Catarina.O referido programa busca através de diferentes ações integradas na relação ensino-pesquisa-extensão, desenvolver ações voltadas aos sistemas familiares de produção e comercialização, através dos princípios da interdisciplinaridade. Fazem parte dos parceiros deste programa organizações públicas e privadas do município de Ibirubá-RS. Este programa faz parte de uma iniciativa do Banco Brasil, em conjunto com diversas entidades do setor público e privado (IFRS, COOPEAGRI,Prefeitura Municipal, EMATER, entre outros) que visa qualificar a produção leiteira realizada por pequenos produtores através da prestação de serviços de assistência técnica e extensão rural. Com o foco na sustentabilidade o programa também prevê incrementar a diversificação produtiva nas propriedades rurais, facilitando o contato e a aprendizagem de novas tecnologias adaptadas as condições da agricultura familiar. Uma primeira etapa já fora realizada, em ação conjunta com professores e alunos do IFRS e entidades participantes do programa, efetuaram-se reuniões com as famílias convidadas, da Localidade Linha Pulador Sul, Ibirubá, RS. Esta localidade foi escolhida por motivos de melhor acessibilidade e boa aceitabilidade dos produtores para novas técnicas. Na primeira reunião, realizada na comunidade, foram apresentados os objetivos do programa para as famílias convidadas. Na segunda reunião houve participação das famílias rurais que aderiram ao programa, em um total de oito, na qual as entidades idealizadoras apresentaram aos presentes o modelo de questionário que será aplicado com intenção de fazer um diagnóstico da propriedade, e também foram determinadas as equipes responsáveis pela aplicação deste junto as famílias participantes. A partir de então foram determinadas as datas das visitas, as quais estão em andamento. A próxima etapa prevista será sistematizar os diagnósticos e realizar novos encontros com os produtores, a fim de validar os dados obtidos e criar as estratégias de ação que serão aplicadas nas propriedades rurais no decorrer do Programa

    Radiosensitization of mammary carcinoma cells by telomere homolog oligonucleotide pretreatment

    Introduction: Ionizing radiation (IR) is a widely used approach to cancer therapy, ranking second only to surgery in rate of utilization. Responses of cancer patients to radiotherapy depend in part on the intrinsic radiosensitivity of the tumor cells. Thus, promoting tumor cell sensitivity to IR could significantly enhance the treatment outcome and quality of life for patients. Methods: Mammary tumor cells were treated by a 16-base phosphodiester-linked oligonucleotide homologous to the telomere G-rich sequence TTAGGG (T-oligo: GGTTAGGTGTAGGTTT) or a control-oligo (the partial complement, TAACCCTAACCCTAAC) followed by IR. The inhibition of tumor cell growth in vitro was assessed by cell counting and clonogenic cell survival assay. The tumorigenesis of tumor cells after various treatments was measured by tumor growth in mice. The mechanism underlying the radiosensitization by T-oligo was explored by immunofluorescent determination of phosphorylated histone H2AX (γ\gammaH2AX) foci, β\beta-galactosidase staining, comet and Terminal deoxynucleotidyl transferase dUTP Nick End Labeling (TUNEL) assays. The efficacy of the combined treatment was assessed in a spontaneous murine mammary tumor model. Results: Pretreatment of tumor cells with T-oligo for 24 hours in vitro enhanced both senescence and apoptosis of irradiated tumor cells and reduced clonogenic potential. Radiosensitization by T-oligo was associated with increased formation and/or delayed resolution of γ\gammaH2AX DNA damage foci and fragmented DNA. T-oligo also caused radiosensitization in two in vivo mammary tumor models. Indeed, combined T-oligo and IR-treatment in vivo led to a substantial reduction in tumor growth. Of further significance, treatment with T-oligo and IR led to synergistic inhibition of the growth of spontaneous mammary carcinomas. Despite these profound antitumor properties, T-oligo and IR caused no detectable side effects under our experimental conditions. Conclusions: Pretreatment with T-oligo sensitizes mammary tumor cells to radiation in both in vitro and in vivo settings with minimal or no normal tissue side effects

    Alterações musculares e esqueléticas cervicais em mulheres disfônicas

    Termo clínico, a disfonia envolve a todas as transformações e dificuldades durante a emissão vocal, as quais resultam no impedimento da produção normal da voz. Pacientes como esse problema, podem apresentar desequilíbrio da musculatura crâniocervical e laríngea e lesão orgânica subjacente. A disfonia resulta em modificações fonatórias, limitando atividades diárias relacionadas ao uso da voz, impactando na vida social e na qualidade de vida do indivíduo. Este estudo teve como objetivo analisar alterações musculares e esqueléticas cervicais em mulheres com disfonia, conforme identificado na literatura científica sobre o tema. Para isso, realizou-se uma revisão integrativa de literatura, selecionando estudos nas bases de dados Literatura Latino-americana e do Caribe em Ciências da Saúde (Lilacs) e Medical Literature Analysis and Retrieval System Online (Medline). A partir da análise qualitativa dos resultados, concluiu-se que dor intensa na região posterior do pescoço e na laringe se manifestam em mulheres disfônicas. Contribuem para isso a função prejudicada da articulação cervical e alterações da amplitude de movimento cervical. Com isso, compreende-se que o abuso vocal e o mau uso da voz como fatores mais comuns para a disfonia

    An integrative approach for building personalized gene regulatory networks for precision medicine

    Only a small fraction of patients respond to the drug prescribed to treat their disease, which means that most are at risk of unnecessary exposure to side effects through ineffective drugs. This inter-individual variation in drug response is driven by differences in gene interactions caused by each patient's genetic background, environmental exposures, and the proportions of specific cell types involved in disease. These gene interactions can now be captured by building gene regulatory networks, by taking advantage of RNA velocity (the time derivative of the gene expression state), the ability to study hundreds of thousands of cells simultaneously, and the falling price of single-cell sequencing. Here, we propose an integrative approach that leverages these recent advances in single-cell data with the sensitivity of bulk data to enable the reconstruction of personalized, cell-type- and context-specific gene regulatory networks. We expect this approach will allow the prioritization of key driver genes for specific diseases and will provide knowledge that opens new avenues towards improved personalized healthcare
