    Low-Cost Double Curvature: Geometrical and Structural Potentials of Rectangular, Cold-Bent Glass Construction

    The realization of doubly curved façades often requires large investments in fabrication equipment and produces additional waste through subtractive fabrication processes and non-reusable molds. In glass construction, elastic bending techniques can be used for small curvatures. This paper continues previous research of the authors on bending rectangular glass elements into irregularly curved panels. First, we analyze the stresses occurring in cold bent glass during assembly, thus defining a particlespring model which is able to compute approximate stresses in real-time during the bending procedure. In a second step, we compare the structural performance of the bent glass with that of flat panels using FE-analysis. Finally, we illustrate the implementations on multi-panel façade layouts. We analyze the dependencies between curvature, gap-tolerance and panelization. We present a method to minimize gap-tolerances by optimizing the distribution of surface curvature. Our results highlight the structural and geometrical potentials and possible applications for curved glass construction

    An intuitionistic approach to scoring DNA sequences against transcription factor binding site motifs

    Background: Transcription factors (TFs) control transcription by binding to specific regions of DNA called transcription factor binding sites (TFBSs). The identification of TFBSs is a crucial problem in computational biology and includes the subtask of predicting the location of known TFBS motifs in a given DNA sequence. It has previously been shown that, when scoring matches to known TFBS motifs, interdependencies between positions within a motif should be taken into account. However, this remains a challenging task owing to the fact that sequences similar to those of known TFBSs can occur by chance with a relatively high frequency. Here we present a new method for matching sequences to TFBS motifs based on intuitionistic fuzzy sets (IFS) theory, an approach that has been shown to be particularly appropriate for tackling problems that embody a high degree of uncertainty. Results: We propose SCintuit, a new scoring method for measuring sequence-motif affinity based on IFS theory. Unlike existing methods that consider dependencies between positions, SCintuit is designed to prevent overestimation of less conserved positions of TFBSs. For a given pair of bases, SCintuit is computed not only as a function of their combined probability of occurrence, but also taking into account the individual importance of each single base at its corresponding position. We used SCintuit to identify known TFBSs in DNA sequences. Our method provides excellent results when dealing with both synthetic and real data, outperforming the sensitivity and the specificity of two existing methods in all the experiments we performed. Conclusions: The results show that SCintuit improves the prediction quality for TFs of the existing approaches without compromising sensitivity. In addition, we show how SCintuit can be successfully applied to real research problems. In this study the reliability of the IFS theory for motif discovery tasks is proven

    Fetuin-A Induces Cytokine Expression and Suppresses Adiponectin Production

    BACKGROUND: The secreted liver protein fetuin-A (AHSG) is up-regulated in hepatic steatosis and the metabolic syndrome. These states are strongly associated with low-grade inflammation and hypoadiponectinemia. We, therefore, hypothesized that fetuin-A may play a role in the regulation of cytokine expression, the modulation of adipose tissue expression and plasma concentration of the insulin-sensitizing and atheroprotective adipokine adiponectin. METHODOLOGY AND PRINCIPAL FINDINGS: Human monocytic THP1 cells and human in vitro differenttiated adipocytes as well as C57BL/6 mice were treated with fetuin-A. mRNA expression of the genes encoding inflammatory cytokines and the adipokine adiponectin (ADIPOQ) was assessed by real-time RT-PCR. In 122 subjects, plasma levels of fetuin-A, adiponectin and, in a subgroup, the multimeric forms of adiponectin were determined. Fetuin-A treatment induced TNF and IL1B mRNA expression in THP1 cells (p<0.05). Treatment of mice with fetuin-A, analogously, resulted in a marked increase in adipose tissue Tnf mRNA as well as Il6 expression (27- and 174-fold, respectively). These effects were accompanied by a decrease in adipose tissue Adipoq mRNA expression and lower circulating adiponectin levels (p<0.05, both). Furthermore, fetuin-A repressed ADIPOQ mRNA expression of human in vitro differentiated adipocytes (p<0.02) and induced inflammatory cytokine expression. In humans in plasma, fetuin-A correlated positively with high-sensitivity C-reactive protein, a marker of subclinical inflammation (r = 0.26, p = 0.01), and negatively with total- (r = -0.28, p = 0.02) and, particularly, high molecular weight adiponectin (r = -0.36, p = 0.01). CONCLUSIONS AND SIGNIFICANCE: We provide novel evidence that the secreted liver protein fetuin-A induces low-grade inflammation and represses adiponectin production in animals and in humans. These data suggest an important role of fatty liver in the pathophysiology of insulin resistance and atherosclerosis

    Multiplying Labour, Multiplying Resistance: Class Composition in Buenos Aires’ Clandestine Textile Workshops

    Buenos Aires’ talleres clandestinos (clandestine textile workshops) are powerful sites of accumulation and resistance; a complex and communitarian migrant economy. The economy’s complexity is, however, masked by its spatiality, clandestinity, and the promotion of culturalist analyses that ignore intra-collective class differentials. This paper considers the “autonomy of migration” approach through the lens of “class composition” to explore the talleres’ contours. Witnessed in the talleres is a clear “multiplication of labour”, yet approaching this multiplication compositionally highlights the multiple examples of resistance and refusal immanent to the workshop economy. But this dialectic of control and resistance transcends the workplace, with the talleres one node in a wider, socially reproductive borderscape. By developing a framework that neither condemns nor celebrates economic structures like the talleres, but instead unpacks their antagonistic nature, the paper highlights the benefits of (a)analysing the autonomy of migration approach compositionally, and (b)further geographical engagement with autonomist thinking

    Modulation of vascular reactivity by perivascular adipose tissue (PVAT)

    Purpose of Review: In this review we discuss the role of perivascular adipose tissue (PVAT) in the modulation of vascular contractility and arterial pressure, focusing on the role of the renin-angiotensin-aldosterone system and oxidative stress/inflammation. Recent Findings: PVAT possesses an relevant endocrine-paracrine activity, which may be altered in several pathophysiological and clinical conditions. During the last two decades it has been shown PVAT may modulate vascular reactivity. It has also been previously demonstrated that inflammation in adipose tissue may be implicated in vascular dysfunction. In particular, adipocytes secrete a number of adipokines with various functions, as well as several vasoactive factors, together with components of the renin-angiotensin system which may act at local or at systemic level. It has been shown that the anticontractile effect of PVAT is lost in obesity, probably as a consequence of the development of adipocyte hypertrophy, inflammation, and oxidative stress. Summary: Adipose tissue dysfunction is interrelated with inflammation and oxidative stress, thus contributing to endothelial dysfunction observed in several pathological and clinical conditions such as obesity and hypertension. Decreased local adiponectin level, macrophage recruitment and infiltration, and activation of renin-angiotensin-aldosterone system could play an important role in this regards

    The Benefits of Titling Indigenous Communities in the Peruvian Amazon: A Stated Preference Approach

    We conduct a discrete choice experiment with leaders of 164 Peruvian Indigenous communities (ICs) to elicit their preferences about and valuation of land titles—to our knowledge, the first use of rigorous stated preference methods to analyze land titling. We find that on average, IC leaders are willing to pay US35,000US35,000–US45,000 for a title, roughly twice the per community administrative cost of titling; willingness to pay is positively correlated with the value of IC land and the risk of land grabbing; and leaders prefer titling processes that involve Indigenous representatives and titles that encompass land with cultural value
