    Assessing uncertainty in the American Indian Trust Fund

    Fiscal year-end balances of the Individual Indian Money System (a part of the Indian Trust) were constructed from data related to money collected in the system and disbursed by the system from 1887 to 2007. The data set of fiscal year accounting information had a high proportion of missing values, and much of the available data did not satisfy basic accounting relationships. Instead of just calculating a single estimate and arguing to the Court that the assumptions needed for the computation were reasonable, a distribution of calculated balances was developed using multiple imputation and time series models.     Kundenbindung von Jugendlichen am Beispiel der Jugendbibliothek freestyle der StadtbĂĽchereien DĂĽsseldorf

    Die vorliegende Arbeit beschäftigt sich mit der Jugendbibliothek freestyle der Stadtbüchereien Düsseldorf. Das Konzept zielt auf eine langfristige Kundenbindung jugendlicher Nutzer ab 14 Jahre. Das Projekt und seine Durchführung werden ausführlich vorgestellt. Ferner wird eine Kundenbefragung unter den freestyle-Nutzern durchgeführt. Diese ermittelt die Kundenzufriedenheit und liefert Rückschlüsse auf den Erfolg des Konzepts. Darüber hinaus werden das Freizeit- und Medienverhalten Jugendlicher aufgegriffen und Ursachen für den Wegfall dieser Zielgruppe in Öffentlichen Bibliotheken ermittelt. Zusätzlich verdeutlicht diese Arbeit die zunehmende Bedeutung von Kundenbindung und Marketing für öffentliche Einrichtungen und stellt praktische Kundenbindungsinstrumente aus Wirtschaft und Kultur vor, die als Anregung für ein langfristiges Kundenbindungsprogramm in Öffentlichen Bibliotheken dienen sollen

    Beep Tones Attenuate Pain following Pavlovian Conditioning of an Endogenous Pain Control Mechanism

    Heterotopic noxious counter-stimulation (HNCS) is commonly used to study endogenous pain control systems. The resulting pain inhibition is primarily based on spinal cord-brainstem loops. Recently, functional imaging studies have shown that limbic structures like the anterior cingulate cortex and amygdala are also implicated. Since these structures are involved in learning processes, it is possible that the HNCS-induced pain inhibition may depend on specific cues from the environment that have been associated with pain reduction through associative learning. We investigated the influence of Pavlovian conditioning on HNCS-induced pain inhibition in 32 healthy subjects by using a differential conditioning paradigm in which two different acoustic stimuli were either repeatedly paired or unpaired with HNCS. Series of noxious electrical pulse trains delivered to the non-dominant foot served as test stimuli. Diffuse noxious inhibitory control (DNIC)-like effects were induced by concurrent application of tonic HNCS (immersion of the contralateral hand in ice water). Subjective pain intensity and pain unpleasantness ratings and electromyographic recordings of the facial corrugator muscle and the nocifensive RIII flexion reflex were used to measure changes in pain sensitivity. HNCS induced significant pain and reflex inhibitions. In the post-conditioning phase, only the paired auditory cue was able to significantly reduce pain perceptions and corrugator muscle activity. No conditioned effect could be observed in RIII reflex responses. Our results indicate that the functional state of endogenous pain control systems may depend on associative learning processes that, like in the present study, may lead to an attenuation of pain perception. Similar albeit opposite conditioning of pain control mechanisms may significantly be involved in the exacerbation and chronification of pain states

    Neutron Yield Predictions with Artificial Neural Networks: A Predictive Modeling Approach

    The development of compact neutron sources for applications is extensive and features many approaches. For ion-based approaches, several projects with different parameters exist. This article focuses on ion-based neutron production below the spallation barrier for proton and deuteron beams with arbitrary energy distributions with kinetic energies from 3 MeV to 97 MeV. This model makes it possible to compare different ion-based neutron source concepts against each other quickly. This contribution derives a predictive model using Monte Carlo simulations (an order of 50,000 simulations) and deep neural networks. It is the first time a model of this kind has been developed. With this model, lengthy Monte Carlo simulations, which individually take a long time to complete, can be circumvented. A prediction of neutron spectra then takes some milliseconds, which enables fast optimization and comparison. The models’ shortcomings for low-energy neutrons (<0.1 MeV) and the cut-off prediction uncertainty (±3 MeV) are addressed, and mitigation strategies are proposed

    The Opposition to Latin American Liberation Theology and the Transformation of Christianity, 1960-1990

    This thesis aims to explore the political, social and religious opposition to Liberation Theology in Latin America during the 1960s to 1990s, and the transformation of Christianity. During this period, most Latin American countries underwent social struggles and political repression in which opposition and persecution arose from dictatorial and military governments who labeled those committed to the poor as communists. Liberation Theology emerged as an ecclesial and theological trend committed to the poor, in the late 1960’s and early 1970’s in Latin America. This thesis traces the origins, development, expansion and repression of Liberation Theology. This work maintains that under the Cold War context and the National Security Doctrine, Liberation Theology became a target of political repression because its commitment with the poor placed it as subversive and communist. This research reveals how it was repressed with violence and the promotion of counteracting religious groups, leading to changes in Christianity

    Pain-autonomic measures reveal nociceptive sensitization in complex regional pain syndrome

    BACKGROUND Allodynia and hyperalgesia are common signs in individuals with complex regional pain syndrome (CRPS), mainly attributed to sensitization of the nociceptive system. Appropriate diagnostic tools for the objective assessment of such hypersensitivities are still lacking, which are essential for the development of mechanism-based treatment strategies. OBJECTIVES This study investigated the use of pain-autonomic readouts to objectively detect sensitization processes in CRPS. METHODS Twenty individuals with chronic CRPS were recruited for the study alongside 16 age- and sex-matched healthy controls (HC). All individuals underwent quantitative sensory testing and neurophysiological assessments. Sympathetic skin responses (SSRs) were recorded in response to 15 pinprick and 15 noxious heat stimuli of the affected (CRPS hand/foot) and a control area (contralateral shoulder/hand). RESULTS Individuals with CRPS showed increased mechanical pain sensitivity and increased SSR amplitudes compared with HC in response to pinprick and heat stimulation of the affected (p  0.05). Habituation of pinprick-induced SSRs was reduced in CRPS compared to HC in both the affected (p = 0.018) and slightly in the control area (p = 0.048). Habituation of heat-induced SSR was reduced in CRPS in the affected (p = 0.008), but not the control area (p = 0.053). CONCLUSIONS This is the first study demonstrating clinical evidence that pain-related autonomic responses may represent objective tools to quantify sensitization processes along the nociceptive neuraxis in CRPS (e.g. widespread hyperexcitability). Pain-autonomic readouts could help scrutinize mechanisms underlying the development and maintenance of chronic pain in CRPS and provide valuable metrics to detect mechanism-based treatment responses in clinical trials. SIGNIFICANCE This study provides clinical evidence that autonomic measures to noxious stimuli can objectively detect sensitization processes along the nociceptive neuraxis in complex regional pain syndrome (CRPS) (e.g. widespread hyperexcitability). Pain-autonomic readouts may represent valuable tools to explore pathophysiological mechanisms in a variety of pain patients and offer novel avenues to help guide mechanism-based therapeutic strategies

    R-loop landscape in mature human sperm: Regulatory and evolutionary implications

    R-loops are three-stranded nucleic acid structures consisting of an RNA:DNA hybrid and a displaced DNA strand. While R-loops pose a potential threat to genome integrity, they constitute 5% of the human genome. The role of R-loops in transcriptional regulation, DNA replication, and chromatin signature is becoming increasingly clear. R-loops are associated with various histone modifications, suggesting that they may modulate chromatin accessibility. To potentially harness transcription-coupled repair mechanisms in the germline, nearly the entire genome is expressed during the early stages of male gametogenesis in mammals, providing ample opportunity for the formation of a transcriptome-dependent R-loop landscape in male germ cells. In this study, our data demonstrated the presence of R-loops in fully mature human and bonobo sperm heads and their partial correspondence to transcribed regions and chromatin structure, which is massively reorganized from mainly histone to mainly protamine-packed chromatin in mature sperm. The sperm R-loop landscape resembles characteristic patterns of somatic cells. Surprisingly, we detected R-loops in both residual histone and protamine-packed chromatin and localize them to still-active retroposons, ALUs and SINE-VNTR-ALUs (SVAs), the latter has recently arisen in hominoid primates. We detected both evolutionarily conserved and species-specific localizations. Comparing our DNA-RNA immunoprecipitation (DRIP) data with published DNA methylation and histone chromatin immunoprecipitation (ChIP) data, we hypothesize that R-loops epigenetically reduce methylation of SVAs. Strikingly, we observe a strong influence of R-loops on the transcriptomes of zygotes from early developmental stages before zygotic genome activation. Overall, these findings suggest that chromatin accessibility influenced by R-loops may represent a system of inherited gene regulation
