    An Intermediate-band imaging survey for high-redshift Lyman Alpha Emitters: The Mahoroba-11

    We present results of our intermediate-band optical imaging survey for high-zz Lyα\alpha emitters (LAEs) using the prime focus camera, Suprime-Cam, on the 8.2m Subaru Telescope. In our survey, we use eleven filters; four broad-band filters (BB, RcR_{\rm c}, ii^\prime, and zz^\prime) and seven intermediate-band filters covering from 500 nm to 720 nm; we call this imaging program as the Mahoroba-11. The seven intermediate-band filters are selected from the IA filter series that is the Suprime-Cam intermediate-band filter system whose spectral resolution is R=23R = 23. Our survey has been made in a 34×2734^\prime \times 27^\prime sky area in the Subaru XMM Newton Deep Survey field. We have found 409 IA-excess objects that provide us a large photometric sample of strong emission-line objects. Applying the photometric redshift method to this sample, we obtained a new sample of 198 LAE candidates at 3<z<53 < z < 5. We found that there is no evidence for evolution of the number density and the star formation rate density for LAEs with logL(Lyα)(ergs1)>42.67\log L({\rm Ly}\alpha) ({\rm erg s^{-1}}) > 42.67 between z3z \sim 3 and 5.Comment: 46 pages, 15 figures, PASJ, Vol.57, No.6, in pres

    Cancer chemotherapy regimens, particularly those employing high-dose cytotoxic drugs such as cyclophosphamide (CTX), have been considered to be immune suppressive. However, we observed that a single administration of high-dose CTX abolished tumors arising from subcutaneous injection of a mouse hepatoma cell line and subsequently induced specific tumor immunity. Depletion of T cells, specifically CD4+ T cells, abrogated the CTX-mediated tumor regression. CTX treatment induced the rapid recruitment of CD4+ T cells into the tumors, and these recruited cells initiated expression of LAMP1/CD107a, a cytotoxic granule molecule, and granzyme B in the absence of antigen presentation at draining lymph nodes and proliferation in the tumor tissues. Moreover, CTX enhanced the expression of a CC chemokine, CCL3, in tumor tissues, and CTX-mediated tumor regression was attenuated in mice deficient in CCR5, the receptor for this chemokine. Consistently, less CTX-induced accumulation of intratumoral LAMP1/CD107a-expressing CD4+ T cells was observed in mice receiving splenocytes derived from CCR5-deficient mice than in those receiving splenocytes derived from WT mice. Thus, CTX induces the expression of CCL3, which induces the intratumoral migration of CD4+ T cells expressing cytotoxic molecules, leading to tumor eradication and subsequent specific tumor immunity. © 2015 Elsevier Ireland Ltd.12 months Embarg

    A novel superior factor widely controlling the rice grain quality

    Synthesis of storage starch and protein accumulation is the main action of endosperm organogenesis in term of the economic importance of rice. This event is strongly disturbed by abiotic stresses such as high temperature; thus, the upcoming global warming will cause a crisis with a great impact on food production^1,2^. The enzymes for the protein storage and starch synthesis pathway should work in concert to carry out the organogenesis of rice endosperm^3-5^, but the regulatory mechanism is largely unknown. Here we show that a novel regulatory factor, named OsCEO1, acts as the conductor of endosperm organogenesis during the rice grain filling stage. The physiological properties of _floury-endosperm-2_ (_flo2_) mutants showed many similarities to symptoms of grains developed under high-temperature conditions, suggesting important roles of the responsible gene in sensitivity to high-temperature stress. Our map-based cloning identified the responsible gene for the _flo2_ mutant, _OsCEO1_, which has no homology to any genes of known function. The _OsCEO1_ belongs to a novel conserved gene family and encodes a protein composed of 1,720 amino acid residues containing a TPR (tetratricopeptide repeat) motif, which is considered to mediate a protein-protein interaction. The yeast two-hybrid analysis raised an unknown protein showing homology to a late embryogenesis abundant protein and a putative basic helix-loop-helix protein as candidates for the direct interactor for _OsCEO1_, whereas no enzyme genes for the synthesis of storage substances were detected. The _flo2_ mutant exhibited reduced expression of several genes for putative regulatory proteins as well as many enzymes involved in storage starch and proteins. These results suggest that _OsCEO1_ is a superior conductor of the novel regulatory cascade of endosperm organogenesis and may have important roles in the response to high-temperature stress
