    Antioxidant activity of phenolic extracts from different cultivars of Italian onion (Allium cepa) and relative human immune cell proliferative induction

    The total antioxidant activity (TAC) may vary considerably between onion cultivars. Immunological effects of onion phenolic compounds are still underestimated.The objective of this study is to determine the total phenol content (TPC) and the relative TAC of three Allium cepa L. (Liliaceae) onion cultivars cultivated in Cannara (Italy): Rossa di Toscana, Borettana di Rovato, and Dorata di Parma, and to evaluate the phenol extracts ability to induce human immune cell proliferation.TPC was determined by the Folin-Ciocalteu method, TAC with FRAP, TEAC/ABTS, and DPPH methods. Peripheral blood mononuclear cells from healthy human donors were incubated for 24 h at 37 °C with 1 ng/mL of phenolic extract in PBS, immunostained, and then analyzed by 4-color flow cytometry for the phenotypic characterization of T helper cells (CD4+ cells), cytotoxic T lymphocytes (CD8+ cells), T regulatory cells (CD25high CD4+ cells), and natural killer cells/monocytes (CD16+ cells).Rossa di Toscana displayed the highest TPC (6.61 ± 0.87 mg GA equivalents/g onion bulb DW) and the highest TAC with the experienced methods: FRAP, 9.19 ± 2.54 μmol Trolox equivalents/g onion bulb DW; TEAC/ABTS, 21.31 ± 0.41 μmol Trolox equivalents/g onion bulb DW; DPPH, 22.90 ± 0.01 μmol Trolox equivalents/g onion bulb DW. Incubation with Rossa di Toscana extract determined an increase in the frequency of the antitumor/anti-infection NK CD16+ immune cells (23.0 ± 0.4%).Content of health-promoting phenols and the deriving antioxidant and immunostimulating activity vary considerably among the investigated cultivars.     none5Objectives: the aim of this study are to document the occurrence of a cavernous hemangioma of the tympanic membrane (TM) and external auditory canal (EAC) that invaded the middle ear spaces and to review the relevant leterature. Methods: The clinical presentation, imaging studies, operative report, and histologic findings of this new case of cavernous hemangioma are reviewed. Results: A cavernous hemangioma of the TM and EAC involving the middle ear spaces was surgical excised without complications. The pulsatile tinnitus, which affected our patient at the same ear where the lesion was situated, disappeared after surgery. Our case represent the first documented cavernous hemangioma of the TM and EAC that invaded the middle ear spaces and the eighth case of cavernous hemangioma of the EAC/TM. Computed tomography is the method of choice in evaluating this lesion. Conclusion: Hemangioma of the EAC and/or TM are extremely rare entities amenable to surgical excision. With magnetic resonance resonance imaging, there is difficulty in defining the excat location of thr tumor and degree of the tissue involvement.G. MAGLIULO; D. PARROTTO; B. SARDELLA; C.     Carcinosarcoma dell’utero: descrizione di un caso clinico

    Il carcinosarcoma dell’utero è una rara neoplasia altamente maligna per la tendenza a dare metastasi precoci. Caratteristicamente la neoplasia presenta una componente epiteliale e una componente stromale maligna. Si manifesta clinicamente con una metrorragia in epoca postmenopausale talvolta associata a dolore addominale. Gli Autori descrivono un caso di carcinosarcoma in una donna di 82 anni con sanguinamento vaginale da 10 mesi e anemia sideropenica ingravescente. La paziente veniva sottoposta a RM addome e pelvi che mostrava una neoformazione che occupava completamente la cavità uterina, senza segni di interessamento pelvico. Si poneva indicazione alla isterectomia totale con ovaro-salpingectomia bilaterale. Le condizioni cliniche scadute e l’elevato rischio anestesiologico (ASA 3, gruppo III) sconsigliavano la linfoadenectomia pelvica bilaterale con sampling aortico. Il controllo clinico e la TC a 6 mesi dall’intervento chirurgico non evidenziavano segni di ripresa locale o a distanza della malattia. In conclusione: a) controlli specialistici ginecologici in pazienti in postmenopausa con fattori di rischio (obesità e ipertensione arteriosa) possono permettere una diagnosi occasionale o precoce e una terapia tempestiva del carcinosarcoma dell’utero; b) l’ecotomografia sovrapubica e transvaginale, la TC e la RM sono indispensabili per una corretta stadiazione clinica; c) solo la revisione della cavità utrina con esame istologico del tessuto è in grado di porre diagnosi di certezza

    The survival of osteosarcoma patients is connected to metastasis. The ezrin expression is associated with the development of metastasis and poor outcome in osteosarcoma. Ezrin is present in the cytoplasm and after phosphorylation assumes an active form and links F-actin to the cell membrane. This study evaluated ezrin and phosphorylated ezrin at site Tyr354 and Thr567 expression and its subcellular localization in osteosarcoma. We studied 50 osteosarcoma patients (mean follow-up 9.8 years). Ezrin expression was assessed using immunohistochemical and immunofluorescence analysis on tissue microarray and cultured cells of human osteosarcoma 143B. The western blot analysis was carried out on cultured cells. The majority of osteosarcomas, showing cytoplasmic positivity for ezrin, phosphorylated and unphosphorylated, were associated with membranous and nuclear positivity for phosphorylated ezrin Thr567 and phosphorylated ezrin Tyr354, respectively. Ezrin expression was associated with high-grade osteosarcoma (P≤0.04), with metastasis (P≤0.04) and with tumors that developed metastasis (P≤0.04); phosphorylated ezrin Thr567 expression was present mostly in tumors with metastasis (P≤0.01) and in osteosarcomas that did not develop metastasis (P≤0.002). The osteosarcoma patients with ezrin expression have a short survival. The cytoplasmic ezrin expression in osteosarcoma matches its role of membrane-cytoskeleton linker protein. The subcellular trafficking of ezrin is not blocked and it is linked to ezrin phosphorylation, also in cancer. The phosphorylated ezrin Tyr354 nuclear localization suggests its possible role as a nuclear factor in osteosarcoma. The phosphorylated ezrin Thr567 phosphorylation may not be necessary in osteosarcoma metastatic progression but it was modulated. The ezrin expression is associated with more aggressive osteosarcomas and with metastasis. © 2010 USCAP, Inc. All rights reserved

    CARCINOSARCOMA OF THE UTERUS: A CASE-REPORT AND REVIEW OF THE LITERATURE

    Carcinosarcoma is a rare tumor of the uterus with a poor prognosis. We present a case of uterine carcinosarcoma in an 82-year-old woman who suffered from pervaginal bleeding for ten months duration with progressive anemia. Abdominopelvic nuclear magnetic resonance (NMR) imaging showed the presence of an intrauterine mass, infiltrating the myometrium and reaching the cervix. The patient was submitted to total abdominal hysterectomy with bilateral salpingo-oophorectomy. The carcinosarcoma, arising from the lumen of the uterus, infiltrated the inner one-third of the myometrial layer (pT1b, pNx, pMx; FIGO Stage 1B). A CT of the total body performed six months after surgery showed no signs of recurrent and/or metastatic tumor.
