    Randomized Dynamical Decoupling Techniques for Coherent Quantum Control

    The need for strategies able to accurately manipulate quantum dynamics is ubiquitous in quantum control and quantum information processing. We investigate two scenarios where randomized dynamical decoupling techniques become more advantageous with respect to standard deterministic methods in switching off unwanted dynamical evolution in a closed quantum system: when dealing with decoupling cycles which involve a large number of control actions and/or when seeking long-time quantum information storage. Highly effective hybrid decoupling schemes, which combine deterministic and stochastic features are discussed, as well as the benefits of sequentially implementing a concatenated method, applied at short times, followed by a hybrid protocol, employed at longer times. A quantum register consisting of a chain of spin-1/2 particles interacting via the Heisenberg interaction is used as a model for the analysis throughout.Comment: 7 pages, 2 figures.     Colchicine and amiprophos-methyl (APM) in polyploidy induction in banana plant

    The objective was to assess the colchicine and amiprophos-methyl (APM) concentration and exposure period in the chromosome duplication of breed banana plants diploids. Banana stem tips were used from the following genotypes: breed diploids (1304-04 [Malaccensis x Madang (Musa acuminata spp. banksii)] and 8694-15 [0337-02 (Calcutta x Galeo) x SH32-63]). Colchicine was used at concentrations of 0 (control treatment), 1.25, 2.5 and 5.0 mM, while APM was used at 0 (control treatment), 40 and 80 μM, in solution under agitation (20 rpm), for 24 and 48 h periods. With the use of APM, 66.67% tetraploid plants were obtained in the 1304-04 genotype using 40 μM for 24 h and 18.18% in 80 μM for 48 h, while in the 8694-15 genotype using 40 and 80 μM colchicine for 48 h, 27.27 and 21.43% tetraploid plants were observed, respectively. For colchicine, in the 1304-04 genotype, only the 1.25 mM treatment for 48 h presented 25% tetraploid plants and in the 8694-15 genotype, the 5.0 mM concentration for 48 h produced 50% tetraploid plants. APM for 24 h enabled the tetraploid plant of the 1304-04 genotype to be obtained, while colchicine for 48 h resulted in tetraploid plants in the 8694-15 genotype.Key words: Musa acuminata, antimitotic, flow cytometry, tissue culture

    Osmotic tolerance and freezability of isolated caprine early-staged follicles

    Isolated caprine early-staged follicles were submitted to osmotic tolerance tests in the presence of sucrose, ethylene glycol (EG), or NaCl solutions and were exposed to and cryopreserved (by slow or rapid cooling) in MEM alone or MEM supplemented with sucrose, EG (1.0 or 4.0 M), or both. When follicles were exposed to 1.5 M NaCl, only 2% of the follicles were viable, whereas 87% of the follicles were viable after exposure to 4.0 M EG. Regarding exposure time, the highest percentage of viable follicles was obtained when follicles were exposed for 10 min to 1.0 M EG + 0.5 M sucrose; exposure for 60 s to 4.0 M EG + 0.5 M sucrose also maintained high percentage viability in follicles. Slow cooling in the presence of 1.0 M EG + 0.5 M sucrose (75%) or rapid cooling in the presence of 4.0 M EG + 0.5 M sucrose (71%) resulted in a significantly higher proportion of viable follicles than all other treatments (P < 0.05). A 24-h culture of frozen-thawed follicles was used to assess survival; only slow-frozen follicles showed viability rates similar to control follicles (64% vs. 69% respectively; P > 0.05). Interestingly, the percentage of viable rapid-cooled follicles (59%) was similar to that obtained after in vitro culture of conventional slow-cooled follicles but was significantly lower than that in controls. Thus, in addition to determining improved procedures for the exposure of follicles to EG and sucrose before and after freezing of caprine early-staged follicles, we report the development of rapid- and slow-cooling protocols

    Exploitation promotes earlier sex change in a protandrous patellid limpet, Patella aspera Röding, 1798

    Exploitation of organisms can prompt the reduction in the number and size of target populations consequently affecting reproductive output and replenishment. Here, we investigated the effects of exploitation on the population structure of a protandrous patellid limpet, Patella aspera, an overexploited Macaronesian endemic. Timed dives were used to collect animals across eleven islands of Macaronesia. Individuals were inspected for sex, size, and gonad stage. Using catch effort (time per person) per island coastal perimeter as a surrogate for exploitation intensity, we found that limpet abundance (CPUE) and mean size tended to decrease with exploitation intensity. When considering the sex of animals separately, the size of the largest male, but not females, decreased with exploitation. In contrast, the size of the smallest male remained relatively consistent, whereas the size of the smallest female decreased significantly with exploitation. As exploitation is mostly targeting larger individuals, results suggest that males are compensating the removal of larger females, by undergoing sex change at smaller and presumably earlier sizes. These results have wider implications for the conservation of P. aspera, as a reduction in female size will likely affect the numbers of oocytes produced, hence fecundity. Regulations promoting the protection of the larger-sized animals should be enforced to safeguard the replenishment of the population

    In order to assess a new strategy of DNA vaccine for a more complete understanding of its action in immune response, it is important to determine the in vivo biodistribution fate and antigen expression. In previous studies, our group focused on the prophylactic and therapeutic use of a plasmid DNA encoding the Mycobacterium leprae 65-kDa heat shock protein (Hsp65) and achieved an efficient immune response induction as well as protection against virulent M. tuberculosis challenge. In the present study, we examined in vivo tissue distribution of naked DNA-Hsp65 vaccine, the Hsp65 message, genome integration and methylation status of plasmid DNA. The DNA-Hsp65 was detectable in several tissue types, indicating that DNA-Hsp65 disseminates widely throughout the body. The biodistribution was dose-dependent. In contrast, RT-PCR detected the Hsp65 message for at least 15 days in muscle or liver tissue from immunized mice. We also analyzed the methylation status and integration of the injected plasmid DNA into the host cellular genome. The bacterial methylation pattern persisted for at least 6 months, indicating that the plasmid DNA-Hsp65 does not replicate in mammalian tissue, and Southern blot analysis showed that plasmid DNA was not integrated. These results have important implications for the use of DNA-Hsp65 vaccine in a clinical setting and open new perspectives for DNA vaccines and new considerations about the inoculation site and delivery system

    Enhanced lithium depletion in Sun-like stars with orbiting planets

    The surface abundance of lithium on the Sun is 140 times less than protosolar, yet the temperature at the base of the surface convective zone is not hot enough to burn Li. A large range of Li abundances in solar type stars of the same age, mass and metallicity is observed, but theoretically difficult to understand. An earlier suggestion that Li is more depleted in stars with planets was weakened by the lack of a proper comparison sample of stars without detected planets. Here we report Li abundances for an unbiased sample of solar-analogue stars with and without detected planets. We find that the planet-bearing stars have less than 1 per cent of the primordial Li abundance, while about 50 per cent of the solar analogues without detected planets have on average 10 times more Li. The presence of planets may increase the amount of mixing and deepen the convective zone to such an extent that the Li can be burned.Comment: 13 pages, 2 figure

    Identification of archaeal proteins that affect the exosome function in vitro

    <p>Abstract</p> <p>Background</p> <p>The archaeal exosome is formed by a hexameric RNase PH ring and three RNA binding subunits and has been shown to bind and degrade RNA <it>in vitro</it>. Despite extensive studies on the eukaryotic exosome and on the proteins interacting with this complex, little information is yet available on the identification and function of archaeal exosome regulatory factors.</p> <p>Results</p> <p>Here, we show that the proteins PaSBDS and PaNip7, which bind preferentially to poly-A and AU-rich RNAs, respectively, affect the <it>Pyrococcus abyssi </it>exosome activity <it>in vitro</it>. PaSBDS inhibits slightly degradation of a poly-rA substrate, while PaNip7 strongly inhibits the degradation of poly-A and poly-AU by the exosome. The exosome inhibition by PaNip7 appears to depend at least partially on its interaction with RNA, since mutants of PaNip7 that no longer bind RNA, inhibit the exosome less strongly. We also show that FITC-labeled PaNip7 associates with the exosome in the absence of substrate RNA.</p> <p>Conclusions</p> <p>Given the high structural homology between the archaeal and eukaryotic proteins, the effect of archaeal Nip7 and SBDS on the exosome provides a model for an evolutionarily conserved exosome control mechanism.</p

    Light propagation and Anderson localization in disordered superlattices containing dispersive metamaterials: Effects of correlated disorder

We consider and compare the cases where disorder is uncorrelated to situations where it is totally correlated and anticorrelated. The photonic gaps of the corresponding periodic structure are not completely destroyed by the presence of disorder, which leads to minima in the localization length. In the vicinities of a gap, the behavior of the localization length depends crucially on the physical origin of the gap (Bragg or non-Bragg gaps). Within a Bragg gap, the localization length increases as the degree of disorder increases, an anomalous behavior that only occurs for the uncorrelated and completely correlated cases. In these cases, minima of the localization length at the positions of Bragg gaps are shifted by increasing disorder, which does not occur for the anticorrelated case, where the positions of the minima remain unaltered. Minima in the localization length corresponding to non-Bragg gaps are not shifted by increasing disorder, albeit the widths of these minima are changed. We have found that the asymptotic behavior for the localization length xi proportional to lambda(6) for disordered metamaterials is not affected by correlations.
