    Solaropsis brasiliana, anatomy, range extension and its phylogenetic position within pleurodontidae (Mollusca, gastropoda, stylommatophora)

    A detailed anatomical revision on Solaropsis brasiliana (Deshayes 1832) has been carried out. New characters on shell, anatomy of soft parts, and a review of the genus distribution in South America, as well as clarification on S. brasiliana distributional area are provided in the present study. Solaropsis brasiliana is diagnosed by its globose, solid, and hirsute shell, with periphery obsoletely angular, bursa copulatrix with a thick, long diverticulum, a thick, long flagellum and a penis retractor muscle forked, with the vas deferens passing through it. This compiled information was used to test the phylogenetic position of S. brasiliana within South American Pleurodontidae through a cladistics analysis. In the phylogenetic hypothesis obtained, S. brasiliana is sister group of S. gibboni (Pfeiffer 1846) and the monophyly of the genus Solaropsis Beck is also supported. Here, we sustain that the distribution of S. brasiliana is restricted to Brazil, inhabiting the States of Rio de Janeiro, Espírito Santo, Bahia and Minas Gerais.Fil: Cuezzo, Maria Gabriela.     Psychometric properties of the Disordered Eating Attitude Scale for adult men

    OBJECTIVE: To evaluate psychometric properties of the Disordered Eating Attitude Scale (DEAS) for men. METHODS: Two hundred and twenty-eight undergraduate male students (18-39 years old) answered the DEAS, originally developed and validated for women. Internal consistency was evaluated by Cronbach's Alpha; convergent validity by comparing DEAS and the Eating Attitude (EAT) and Restraint Scale (RS) scores using Pearson's coefficient. Test-retest reliability was evaluated with a subsample (n = 38) in a month interval by means of intraclass correlation coefficient (ICC). Known-groups validity was obtained comparing scores in DEAS among undergraduate students and men with eating disorders (ED) (n = 28). RESULTS: Internal consistency of scale was 0.63. DEAS score correlated with EAT (r = 0.65) and RS (r = 0.51); ICC between test and retest was 0.948. Known-groups analysis differentiated ED patients and undergraduate students (p < 0.001). CONCLUSIONS: The scale presented adequate psychometric properties and could be used in studies with adult men, since the construct is not explored among males. Nevertheless, it is recommended to revise the scale and to develop specific instruments for male public.OBJETIVO: Avaliar as propriedades psicométricas da Escala de Atitudes Alimentares Transtornadas (EAAT) para o sexo masculino. MÉTODOS: Duzentos e vinte e oito universitários (18-39 anos) responderam à EAAT, originalmente desenvolvida e validada para o sexo feminino. A consistência interna foi avaliada pelo Alpha de Cronbach e a validade convergente, por meio do coeficiente de correlação de Pearson comparando os escores da EAAT, do Teste de Atitudes Alimentares (EAT) e da Escala de Restrição (RS). A reprodutibilidade foi avaliada aplicando a escala numa subamostra (n = 38) com um mês de intervalo utilizando o coeficiente de correlação intraclasse (CCI). A validade known-groups foi obtida comparando o escore dos universitários na EAAT com o escore de homens com diagnóstico de transtornos alimentares (TA) (n = 28). RESULTADOS: A consistência interna da escala foi de 0,63. O escore da EAAT foi correlacionado com a EAT (r = 0,65) e RS (r = 0,51), e o CCI entre o teste e o reteste foi de 0,948. A análise known-groups diferenciou pacientes com TA de estudantes universitários (p < 0,001). CONCLUSÕES: A escala apresentou propriedades psicométricas adequadas e pode ser utilizada em estudos com homens adultos - uma vez que o constructo é pouco explorado em homens.     Avaliação de indicadores sociais em agroecossistemas com barragens subterrâneas no semiárido Baiano, Brasil

    O semiárido brasileiro vem passando por uma gama de mudanças sociais proporcionadas por políticas públicas que vem possibilitando o desenvolvimento de territórios em toda a região. Esse trabalho tem por objetivo avaliar esses agroecossistemas utilizando indicadores sociais. Na amostragem feita foram selecionadas três propriedades no Estado da Bahia: uma no município de Serrolândia e as outras duas no município de Canudos, todas localizadas em clima semiárido. O acesso à água possibilitou algumas transformações sociais positivas porque permitiu um aumento da capacidade produtiva no tempo e no espaço. Mas, quando relacionados ao direito a terra e a questões de direitos básicos como saúde e educação, observa-se que as tecnologias e inovações não têm interferido pouco no que se refere a acesso. O desenvolvimento dessas propriedades dentro dos princípios agroecológicos ainda está entre o nível baixo e intermediário, deixando claro que a sustentabilidade desses agroecossistemas depende do desenvolvimento de fatores sociais, econômicos e econômicos de forma integrada.The Brazilian semiarid region is going through a range of social changes caused by public policies that has enabled the development of territories across the region. This study aims to evaluate these agroecosystems using social indicators. In three properties sampled were selected in the state of Bahia, one in the municipality of Serrolândia and the other two in the municipality of Canudos, all located in semi-arid climate. Access to water has enabled some positive social change because it allowed an increase in production capacity in time and space. But when related to land rights and the basic rights issues such as health and education, it is observed that the technologies and innovations have not interfered little with regard to access. The development of these properties within the agro-ecological principles is still among the low and middle, making it clear that the sustainability of agro-ecosystems depends on the development of social, economic and economic factors in an integrated manner.Eje: A1 Sistemas de producción de base agroecológica (Trabajos científicos)Facultad de Ciencias Agrarias y Forestale

    Expression of NES-hTERT in Cancer Cells Delays Cell Cycle Progression and Increases Sensitivity to Genotoxic Stress

    Telomerase is a reverse transcriptase associated with cellular immortality through telomere maintenance. This enzyme is activated in 90% of human cancers, and inhibitors of telomerase are currently in clinical trials to counteract tumor growth. Many aspects of telomerase biology have been investigated for therapy, particularly inhibition of the enzyme, but little was done regarding its subcellular shuttling. We have recently shown that mutations in the nuclear export signal of hTERT, the catalytic component of telomerase, led to a mutant (NES-hTERT) that failed to immortalize cells despite nuclear localization and catalytic activity. Expression of NES-hTERT in primary fibroblast resulted in telomere-based premature senescence and mitochondrial dysfunction. Here we show that expression of NES-hTERT in LNCaP, SQ20B and HeLa cells rapidly and significantly decreases their proliferation rate and ability to form colonies in soft agar while not interfering with endogenous telomerase activity. The cancer cells showed increased DNA damage at telomeric and extra-telomeric sites, and became sensitive to ionizing radiation and hydrogen peroxide exposures. Our data show that expression of NES-hTERT efficiently counteracts cancer cell growth in vitro in at least two different ways, and suggest manipulation with the NES of hTERT or its subcellular shuttling as a new strategy for cancer treatment

    The germline mutational landscape of BRCA1 and BRCA2 in Brazil

    The detection of germline mutations in BRCA1 and BRCA2 is essential to the formulation of clinical management strategies, and in Brazil, there is limited access to these services, mainly due to the costs/availability of genetic testing. Aiming at the identification of recurrent mutations that could be included in a low-cost mutation panel, used as a first screening approach, we compiled the testing reports of 649 probands with pathogenic/likely pathogenic variants referred to 28 public and private health care centers distributed across 11 Brazilian States. Overall, 126 and 103 distinct mutations were identified in BRCA1 and BRCA2, respectively. Twenty-six novel variants were reported from both genes, and BRCA2 showed higher mutational heterogeneity. Some recurrent mutations were reported exclusively in certain geographic regions, suggesting a founder effect. Our findings confirm that there is significant molecular heterogeneity in these genes among Brazilian carriers, while also suggesting that this heterogeneity precludes the use of screening protocols that include recurrent mutation testing only. This is the first study to show that profiles of recurrent mutations may be unique to different Brazilian regions.
