    A reassessment of the role of sucrose synthase in the hypoxic sucrose‐ethanol transition in Arabidopsis

    Plants under low-oxygen availability adapt their metabolism to compensate for the lower ATP production that arises from the limited respiratory activity in mitochondria. Anaerobic glycolysis requires continuous fuelling of carbon units, also provided from sucrose. The anaerobic catabolism of sucrose is thought to require the activity of sucrose synthase, being this enzymatic reaction more energetically favourable than that of invertase. The role of sucrose synthases (SUS) for aerobic sucrose catabolism in Arabidopsis has been recently questioned since SUS mutants fail to show altered phenotype or metabolic profile. In the present paper, we analysed the role of SUS1 and SUS4, both induced by low oxygen, in plant survival and ethanol production. The results showed that mutants lacking both SUS were as tolerant to low oxygen as the wild type in most of the experimental conditions tested. Only under conditions of limiting sugar availability the requirement of SUS1 and SUS4 for ethanol production was evident, although partly compensated by invertase activities, as revealed by the use of a double mutant lacking the two major cytosolic invertases. We conclude that, contrary to general belief, the sucrose synthase pathway is not the preferential route for sucrose metabolism under hypoxia

    The research of standardized protocols for dog involvement in animal-assisted therapy: A systematic review

    Dogs are considered the most important species involved in animal-assisted therapy (AAT), and the scientific literature focuses on the benefits linked to the involvement of dogs in various therapeutic areas. In this study, we carried out a systematic review according to the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines, exploring the scientific literature from the last 5 years (2016–2021) on three databases (PubMed, Scopus, and Web of Science) to highlight the characteristics of the dogs involved in AATs. Based on the scientific literature relevant to such dogs, we considered different parameters (i.e., number, age, sex, breed, temperament, methods of choice and training, health status, research goals, and activities with dogs) to include studies in our paper. After screening 4331 papers identified on the searched databases, we selected 38 articles that met the inclusion criteria. Analysis of the included articles showed that the characteristics of the dogs were neglected. Our findings indicated a lack of information about the dogs, as well as the absence of standardized and univocal criteria for dog selection, training programs, and health protocols

    Distribution of helminths in buffalo farms from central Italy

    The aim of the present study was to obtain up-to-date information on the distribution of helminths in buffalo farms of central Italy. The survey was carried out on a sample of 127 farms (epidemiological units), selected using a grid approach within a Geographical Information System, followed by proportional allocation. In each farm the number of buffaloes tested (adults, heifer/steers and calves) ranged from 9 to 21 based on the number of animals on the farm. The total number of faecal samples collected from the 127 farms was 1883. Copromicroscopic examinations were performed using the FLOTAC technique. The following helminths were detected in the examined farms: gastrointestinal strongyles (33.1%), Strongyloides spp. (3.1%), Fasciola hepatica (7.1%), Dicrocoelium dendriticum (2.4%), Paramphistomidae (7.1%), and Moniezia spp. (2.4%)

    Fibroblast proliferation and migration in wound healing by phytochemicals: Evidence for a novel synergic outcome

    Wound-healing is a dynamic skin reparative process that results in a sequence of events, including inflammation, proliferation, and migration of different cell types as fibroblasts. Fibroblasts play a crucial role in repairing processes, from the late inflammatory phase until the fully final epithelization of the injured tissue. Within this context, identifying tools able to implement cell proliferation and migration could improve tissue regeneration. Recently, plants species from all over the world are coming out as novel tools for therapeutic applications thanks to their phytochemicals, which have antioxidant properties and can promote wound healing. In this paper, we aimed at investigating antioxidant activity of waste extracts from different medicinal plants, endemic of the Mediterranean area, on fibroblast proliferation and wound healing. We determined the amount of total phenols and anti-oxidant activity by ABTS assay. We then evaluated the cytotoxicity of the compounds and the proliferative capabilities of fibroblasts by scratch assay. Our results showed that waste extracts retain antioxidant and regenerative properties, inducing tissue re-establishment after environmental stress exposure. Taken together, our findings suggest that waste material could be used in the future also in combinations to stimulate wound healing processes and antioxidant responses in damaged skin

    Mechanical Stimulation of Fibroblasts by Extracorporeal Shock Waves: Modulation of Cell Activation and Proliferation Through a Transient Proinflammatory Milieu

    Extracorporeal shock waves (ESWTs) are \u201cmechanical\u201d waves, widely used in regenerative medicine, including soft tissue wound repair. Although already being used in the clinical practice, the mechanism of action underlying their biological activities is still not fully understood. In the present paper we tried to elucidate whether a proinflammatory effect may contribute to the regenerative potential of shock waves treatment. For this purpose, we exposed human foreskin fibroblasts (HFF1 cells) to an ESWT treatment (100 pulses using energy flux densities of 0.19 mJ/mm2 at 3 Hz), followed by cell analyses after 5 min, up to 48 h. We then evaluated cell proliferation, reactive oxygen species generation, ATP release, and cytokine production. Cells cultured in the presence of lipopolysaccharide (LPS), to induce inflammation, were used as a positive control, indicating that LPS-mediated induction of a proinflammatory pattern in HFF1 increased their proliferation. Here, we provide evidence that ESWTs affected fibroblast proliferation through the overexpression of selected cytokines involved in the establishment of a proinflammatory program, superimposable to what was observed in LPS-treated cells. The possibility that inflammatory circuits can be modulated by ESWT mechanotransduction may disclose novel hypothesis on their biological underpinning and expand the fields of their biomedical application

    Consensus Statement on Animals’ Relationship with Pediatric Oncohematological Patients, on Behalf of Infectious Diseases and Nurse Working Groups of the Italian Association of Pediatric Hematology-Oncology

    : Contact with animals in pediatric oncohematologic patients is associated with many benefits, but the risk of contracting zoonoses, even if low, must be considered by clinicians. In order to assess the awareness about this topic, we surveyed the Italian pediatric oncohematology centers, which resulted in heterogeneous responses. The Infectious Diseases Working Group and the Nurse Working Group of the Italian Association of Pediatric Hematology-Oncology, together with veterinarians from the National Federation of Italian Veterinarians, drew up a consensus document to unify the indications to be given to families with the aim of guaranteeing a safe interaction between patients and animals and improving the collaboration of clinicians with veterinarians and families

    Affiliative bonds and cruel behavior in childhood interspecific relationships: A conceptual framework on the psychodynamics of affect regulation

    Background: Childhood interspecific relationships can be viewed as a mirror of the person’s capacity to interact with other living beings. The interspecific relationship can involve different attachment styles, affect regulation skills, transitional object dynamics, and self-awareness and mentalization processes. Yet interspecific relationships can also be disrupted, since they can be associated with cruel behavior toward animals, which is in turn related to possible psychopathology. Method: We provided a conceptual framework based on the literature addressing the interspecific relationship in both its adaptive and positive features as well as in its disrupted aspects. Discussion: Pets have often been regarded as attachment figures that can aid children in socialization and growth processes. They have been deemed to represent “social catalysts” that tend to facilitate human relationships, thus increasing prosocial behaviors. On the dark side of the pet-child interaction, childhood abuse of animals tends to be associated with behavioral and emotional problems that have been thought of as underlying psychopathology (e.g., conduct disorder and antisocial personality traits). Childhood cruelty to animals seems to accompany emotion dysregulation, poor social information processing, and low empathy. Conclusions: Since the child-pet relationship has substantial implications for the individual’s mental health and for the potential development of psychopathology, addressing children’s attitudes toward animals can aid in understanding the affective and emotional dimensions of their interpersonal experience

    Process evaluation of animal assisted therapies with children: The role of the human-animal bond on the therapeutic alliance, depth of elaboration, and smoothness of sessions

    Objective: The current study examined the change processes in three Animal Assisted Therapies (AATs) by assessing the relationship between the human-animal bond and the therapeutic alliance, depth of elaboration, and smoothness of treatment sessions. Methods: This was a retrospective study assessing videotaped AATs conducted with three children aged from 7 to 9 years old from the observer perspective. The AATs were based on the Federico II Model of Healthcare Zooanthropology, and each consisted in 8 sessions. A one-way analysis of variance and linear regression models was performed. Results: The human-animal bond, therapeutic alliance, depth of elaboration, and smoothness significantly improved during the sessions. Furthermore, the improvement in therapeutic dimensions proved to be a function of the increase in the human-animal bond. Conclusions: The findings have provided empirical evidence of the change processes occurring during AATs with children, offering some limited but specific insights into the interspecific relationship as a central axis of such treatments

    Vascular Health in American Football Players: Cardiovascular Risk Increased in Division III Players

    Studies report that football players have high blood pressure (BP) and increased cardiovascular risk. There are over 70,000 NCAA football players and 450 Division III schools sponsor football programs, yet limited research exists on vascular health of athletes. This study aimed to compare vascular and cardiovascular health measures between football players and nonathlete controls. Twenty-three athletes and 19 nonathletes participated. Vascular health measures included flow-mediated dilation (FMD) and carotid artery intima-media thickness (IMT). Cardiovascular measures included clinic and 24 hr BP levels, body composition, VO2 max, and fasting glucose/cholesterol levels. Compared to controls, football players had a worse vascular and cardiovascular profile. Football players had thicker carotid artery IMT (0.49 ± 0.06 mm versus 0.46 ± 0.07 mm) and larger brachial artery diameter during FMD (4.3 ± 0.5 mm versus 3.7 ± 0.6 mm), but no difference in percent FMD. Systolic BP was significantly higher in football players at all measurements: resting (128.2 ± 6.4 mmHg versus 122.4 ± 6.8 mmHg), submaximal exercise (150.4 ± 18.8 mmHg versus 137.3 ± 9.5 mmHg), maximal exercise (211.3 ± 25.9 mmHg versus 191.4 ± 19.2 mmHg), and 24-hour BP (124.9 ± 6.3 mmHg versus 109.8 ± 3.7 mmHg). Football players also had higher fasting glucose (91.6 ± 6.5 mg/dL versus 86.6 ± 5.8 mg/dL), lower HDL (36.5±11.2 mg/dL versus 47.1±14.8 mg/dL), and higher body fat percentage (29.2±7.9% versus 23.2±7.0%). Division III collegiate football players remain an understudied population and may be at increased cardiovascular risk
