Instability of vortex array and transitions to turbulent states in rotating helium II
We consider superfluid helium inside a container which rotates at constant
angular velocity and investigate numerically the stability of the array of
quantized vortices in the presence of an imposed axial counterflow. This
problem was studied experimentally by Swanson {\it et al.}, who reported
evidence of instabilities at increasing axial flow but were not able to explain
their nature. We find that Kelvin waves on individual vortices become unstable
and grow in amplitude, until the amplitude of the waves becomes large enough
that vortex reconnections take place and the vortex array is destabilized. The
eventual nonlinear saturation of the instability consists of a turbulent tangle
of quantized vortices which is strongly polarized. The computed results compare
well with the experiments. Finally we suggest a theoretical explanation for the
second instability which was observed at higher values of the axial flow
Control of nuclear-cytoplasmic shuttling of Ankrd54 by PKCδ
AIM
To identify and characterize the effect of phosphorylation on the subcellular localization of Ankrd54.
METHODS
HEK293T cells were treated with calyculin A, staurosporin or phorbol 12-myristate 13-acetate (PMA). Cells were transfected with eGFP-tagged Ankrd54 with or without Lyn tyrosine kinase (wild-type, Y397F mutant, or Y508F mutant). The subcellular localization was assessed by immunofluorescence imaging of cells, immunoblotting of subcellular fractionations. The phosphorylation of Ankrd54 was monitored using Phos-tagTM gel retardation. Phosphorylated peptides were analysed by multiple-reaction-monitoring (MRM) proteomic analysis.
RESULTS
Activation of PKC kinases using PMA promoted nuclear export of Ankrd54 and correlated with increased Ankrd54 phosphorylation, assayed using Phos-tagTM gel retardation. Co-expression of an active form of the PKCδ isoform specifically promoted both phosphorylation and cytoplasmic localization of Ankrd54, while PKCδ, Akt and PKA did not. Alanine mutation of several serine residues in the amino-terminal region of Ankrd54 (Ser14, Ser17, Ser18, Ser19) reduced both PMA induced cytoplasmic localization and phosphorylation of Ankrd54. Using MRM proteomic analysis, phosphorylation of the Ser18 residue of Ankrd54 was readily detectable in response to PMA stimulation. PMA stimulation of cells co-expressing Ankrd54 and Lyn tyrosine kinase displayed increased co-immunoprecipitation and enhanced co-localization in the cytoplasm.
CONCLUSION
We identify phosphorylation by PKCδ as a major regulator of nuclear-cytoplasmic shuttling of Ankrd54, and its interaction with the tyrosine kinase Lyn
Governance of microfinance institutions (MFIs) in Cameroon: What lessons can we learn?
The aim of this paper is to find out the effects of the COBAC regulations regulating the microfinance industry on the governance of microfinance institutions (MFIs) in Cameroon. The paper is based on 35 in-depth interviews carried out from May to June 2011 and June to July 2012 with managers and accountants from MFIs in Cameroon, MFI clients and non-clients, regulatory authorities in the Ministry of Finance, and accounting professionals. The findings show that the regulations have broken down the governance within the MFIs in Cameroon thus turning MFIs into hybrid organizations with managers striving to meet their shareholders' interests
ABC transporters coordinately expressed during lignification of Arabidopsis stems include a set of ABCBs associated with auxin transport
The primary inflorescence stem of Arabidopsis thaliana is rich in lignified cell walls, in both vascular bundles and interfascicular fibres. Previous gene expression studies demonstrated a correlation between expression of phenylpropanoid biosynthetic genes and a subset of genes encoding ATP-binding cassette (ABC) transporters, especially in the ABCB/multi-drug resistance/P-glycoprotein (ABCB/MDR/PGP) and ABCG/pleiotropic drug resistance (ABCG/PDR) subfamilies. The objective of this study was to characterize these ABC transporters in terms of their gene expression and their function in development of lignified cells. Based on in silico analyses, four ABC transporters were selected for detailed investigation: ABCB11/MDR8, ABCB14/MDR12, ABCB15/MDR13, and ABCG33/PDR5. Promoter::glucuronidase reporter assays for each gene indicated that promoters of ABCB11, ABCB14, ABCB15, and ABCG33 transporters are active in the vascular tissues of primary stem, and in some cases in interfascicular tissues as well. Homozygous T-DNA insertion mutant lines showed no apparent irregular xylem phenotype or alterations in interfascicular fibre lignification or morphology in comparison with wild type. However, in abcb14-1 mutants, stem vascular morphology was slightly disorganized, with decreased phloem area in the vascular bundle and decreased xylem vessel lumen diameter. In addition, abcb14-1 mutants showed both decreased polar auxin transport through whole stems and altered auxin distribution in the procambium. It is proposed that both ABCB14 and ABCB15 promote auxin transport since inflorescence stems in both mutants showed a reduction in polar auxin transport, which was not observed for any of the ABCG subfamily mutants tested. In the case of ABCB14, the reduction in auxin transport is correlated with a mild disruption of vascular development in the inflorescence stem
Resistência do algodoeiro à ferrugem tropical potencializada pelo silício
Este trabalho teve como objetivo estudar o efeito do silício (Si) sobre componentes de resistência do algodoeiro à ferrugem tropical causada por Phakopsora gossypii. Plantas das cultivares BRS Buriti e FM 993 cresceram em solução nutritiva contendo zero (-Si) ou 2 mmol Si L- 1 (+Si). A maior concentração foliar de Si aumentou o período de incubação e o período latente em 9% e 14,3%, respectivamente, reduziu a área pustular, a área abaixo da curva de progresso da ferrugem e a área abaixo da curva de progresso da área da pústula em 27,5%, 36% e 22%, respectivamente. Nas plantas das cultivares BRS Buriti e FM 993 supridas com Si houve redução no número de pústulas em 70% e 30% e no número de urédias em 40,3% e 19,5%, respectivamente. A concentração de compostos fenólicos nas plantas da cultivar BRS Buriti supridas com Si aumentou até os 10 dai e, para a cultivar FM993, até os 30 dai. A concentração de derivados da lignina-ácido tioglicólico aumentou depois dos 20 dai para as duas cultivares supridas com Si. Atividade da peroxidas e aumentou até os 25 dai e os 10 dai para as plantas das cultivares BRS Buriti e FM 993 supridas com Si, respectivamente. A atividade da quitinase aumentou até 10 dai para as plantas da cultivar BRS Buriti e aos 10 e 20 dai para as plantas da cultivar supridas com Si. A atividade da glucanase aumentou até os 10 dai para as plantas da cultivar BRS Buriti supridas com Si e até os 20 dai para as plantas da cultivar FM 993 supridas com Si. Em conclusão, o suprimento de Si contribuiu para aumentar a resistência do algodoeiro à ferrugem devido à maior atividade das enzimas de defesa
