    Investigation of Metal and Organic Contaminant Distributions and Sedimentation Rates in Backwater Lakes along the Illinois River

    Systematic sub-sampling of sediment cores in sections of uniform thickness is necessary in order to evaluate historic changes in sediment quality, to determine the vertical extent of contamination, and to measure sedimentation rates. With these objectives in mind, fourteen sediment cores were collected during March 2002 using the Illinois State Water Survey vibracorer. Concentrations of metals and total organic carbon were measured using standard techniques. Concentrations of chlorinated pesticides, phenolic compounds, polycyclic aromatic hydrocarbons (PAHs), and polychlorinated biphenyls (PCBs) were measured by gas chromatography/mass spectrometry (GC-MS). The concentrations of chlorinated pesticides, phenolic compounds and polychlorinated biphenyls (PCBs) were below the method detection limit in all sediment samples analyzed. However, there was a wide range in concentrations of polycyclic aromatic hydrocarbons (PAHs) which were detected in all sediment samples. Also, a wide range of metal concentrations was noted in the sediments evaluated. Lower concentrations of metals were found in the upper 0.5 m of sediment but concentrations were elevated at depths ranging from 1.0 m to 1.5 m. Sedimentation rates were estimated using cesium-137 radiometric dating on 14 vibracores. Sedimentation rates range from < 0.1 to 1.9 cm/yr, with an average of 0.9 cm/yr.     A novel method of non-clinical dispatch is associated with a higher rate of critical Helicopter Emergency Medical Service intervention

    Background - Helicopter Emergency Medical Services (HEMS) are a scarce resource that can provide advanced emergency medical care to unwell or injured patients. Accurate tasking of HEMS is required to incidents where advanced pre-hospital clinical care is needed. We sought to evaluate any association between non-clinically trained dispatchers, following a bespoke algorithm, compared with HEMS paramedic dispatchers with respect to incidents requiring a critical HEMS intervention. Methods - Retrospective analysis of prospectively collected data from two 12-month periods was performed (Period one: 1st April 2014 – 1st April 2015; Period two: 1st April 2016 – 1st April 2017). Period 1 was a Paramedic-led dispatch process. Period 2 was a non-clinical HEMS dispatcher assisted by a bespoke algorithm. Kent, Surrey & Sussex HEMS (KSS HEMS) is tasked to approximately 2500 cases annually and operates 24/7 across south-east England. The primary outcome measure was incidence of a HEMS intervention.Results - A total of 4703 incidents were included; 2510 in period one and 2184 in period two. Variation in tasking was reduced by introducing non-clinical dispatchers. There was no difference in median time from 999 call to HEMS activation between period one and two (period one; median 7 min (IQR 4–17) vs period two; median 7 min (IQR 4–18). Non-clinical dispatch improved accuracy of HEMS tasking to a mission where a critical care intervention was required (OR 1.25, 95% CI 1.04–1.51, p = 0.02).Conclusion - The introduction of non-clinical, HEMS-specific dispatch, aided by a bespoke algorithm improved accuracy of HEMS tasking.     Development and validation of the child post-traumatic cognitions inventory (CPTCI)

    Background: Negative trauma-related cognitions have been found to be a significant factor in the maintenance of post-traumatic stress disorder (PTSD) in adults. Initial studies of such appraisals in trauma-exposed children and adolescents suggest that this is an important line of research in youth, yet empirically validated measures for use with younger populations are lacking. A measure of negative trauma-related cognitions for use with children and adolescents, the Child Post-Traumatic Cognitions Inventory (CPTCI), is presented. The measure was devised as an age-appropriate version of the adult Post-Traumatic Cognitions Inventory (Foa et al., 1999). Methods: The CPTCI was developed and validated within a large (n = 570) sample, comprising community and trauma-exposed samples of children and adolescents aged 6-18 years. Results: Principal components analysis suggested a two-component structure. These components were labelled 'permanent and disturbing change' and 'fragile person in a scary world', and were each found to possess good internal consistency, test-retest reliability, convergent validity, and discriminative validity. The reliability and validity of these sub-scales was present regardless of whether the measure was completed in the acute phase or several months after a trauma. Scores on these sub-scales did not vary with age. Conclusions: The CPTCI is a reliable and valid measure that is not specific to the type of trauma exposure, and shows considerable promise as a research and clinical tool. The structure of this measure suggests that appraisals concerning the more abstract consequences of a trauma, as well as physical threat and vulnerability, are pertinent factors in trauma-exposed children and adolescents, even prepubescent children

    Structural basis of meiotic chromosome synaptic elongation through hierarchical fibrous assembly of SYCE2-TEX12

    The synaptonemal complex (SC) is a supramolecular protein assembly that mediates synapsis between homologous chromosomes during meiosis. SC elongation along the chromosome length (up to 24 ÎŒm) depends on its midline α-fibrous component SYCE2-TEX12. Here, we report X-ray crystal structures of human SYCE2-TEX12 as an individual building-block and upon assembly within a fibrous lattice. We combine these structures with mutagenesis, biophysics and electron microscopy to reveal the hierarchical mechanism of SYCE2-TEX12 fibre assembly. SYCE2-TEX12’s building-blocks are 2:2 coiled-coils which dimerise into 4:4 hetero-oligomers and interact end-to-end and laterally to form 10-nm fibres, which intertwine within 40-nm bundled micrometre-long fibres that define the SC’s midline structure. This assembly mechanism bears striking resemblance with intermediate filament proteins vimentin, lamin and keratin. Thus, SYCE2-TEX12 exhibits behaviour typical of cytoskeletal proteins to provide an α-fibrous SC backbone that structurally underpins synaptic elongation along meiotic chromosomes

    Multiplex stress resistance in cells from long‐lived dwarf mice

    Mutations that extend nematode longevity by interference with IGF‐I/insulin sensing pathways also lead to resistance to multiple forms of stress. Here, we report that skin‐derived fibroblasts from Snell dwarf mice, already known to show increased longevity and delayed aspects of aging, are resistant to multiple forms of cellular stress, including UV light, heat, paraquat, H2O2, and the toxic metal cadmium.     We defend the thesis that every necessarily true proposition is always true. Since not every proposition that is always true is necessarily true, our thesis is at odds with theories of modality and time, such as those of Kit Fine and David Kaplan, which posit a fundamental symmetry between modal and tense operators. According to such theories, just as it is a contingent matter what is true at a given time, it is likewise a temporary matter what is true at a given possible world; so a proposition that is now true at all worlds, and thus necessarily true, may yet at some past or future time be false in the actual world, and thus not always true. We reconstruct and criticize several lines of argument in favor of this picture, and then argue against the picture on the grounds that it is inconsistent with certain sorts of contingency in the structure of time
