    The effect of protocol for disinfection of extracted teeth recommended by center for disease control (CDC) on microhardness of enamel and dentin

    Background: According to the guideline of the United States center for disease control (CDC), the extracted teeth should be sterilized by autoclaving or storage in 10% formalin before using for educational or research purposes. The objective of this study was to evaluate the effect of this protocol on microhardness of dentin and enamel. Material and Methods: Thirty extracted single-root teeth were used in this study. The crowns were resected, and the roots were longitudinally sectioned into two halves. The Vickers microhardness (VHN) of specimens was measured on polished canal dentin and buccal enamel surfaces. The crowns were randomly divided into three groups (n=10). Group 1 and 2 were sterilized using autoclave and formalin, respectively while group 3 (control) was stored in synthetic tissue fluid. The root halves were also randomly divided into 3 groups (n=20) which were treated as mentioned above for crown samples. Following sterilization, VHN of samples was measured again. ANOVA and paired samples t-tests were used to analyze the data. Results: Autoclaving caused a significant reduction in microhardness of dentin ( P <0.001, 12.04% decreases in VHN). However, there were no significant differences for before and after sterilization within other groups. Conclusions: Based on the results of this study, the CDC protocol is recommended in studies related to enamel microhardness. However, Autoclaving is not an appropriate sterilization method in studies related to dentin microhardness. In these studies, two-week immersion in 10% formalin is recommended

    Use of poultry pre-cooked slaughterhouse waste as ruminant feed to prevent environmental pollution

    The nine samples from each slaughterhouse were pooled and sub-sampled to make three samples per slaughterhouse. Dried PSW samples were ground through a 1 mm screen (Wiley mill, Arthur H. Thomas, Philadelphia, PA), and samples analyzed for amino acids, total nitrogen, fat, ash and organic matter (AOAC, 1990). The CNCPS protein fractions of the PSW was determined according to standardized procedure of Licitra et al. (1996) at the University of Bahonar, Kerman. The B2 fraction was calculated by difference and results are reported as CP percentage. Phosphate buffer soluble nitrogen (PBSN) was determined using the phosphate buffer. Neutral detergent insoluble nitrogen (NDIN) and acid detergent insoluble nitrogen (ADIN) were determined as the nitrogen content of the residual after neutral and acid detergent procedures. The analysis of 11 amino acids i.e. arginine, cysteine, histidine, isoleucine, leucine, lysine, methionine, phenylalanine, threonine, tryptophan and valine in three different PSW samples was performed using NIRS, FOSS 5000 Denmark at the Paya Amin Mehr Company (Tehran, Iran).The generation of poultry slaughterhouse waste from poultry production is not only unavoidable but the amount and kinds of waste can cause environmental problems. Rumen digestion of poultry slaughterhouse waste was compared with two common protein sources (fish meal and roasted soybean). Three poultry slaughterhouse waste samples were collected from industrial poultry slaughter-houses and the in situ degradation was done using rumen cannulated sheep. The protein (50e63%), ether extracts (18e27%) and ash (9e15.5%) contents of different poultry slaughterhouse waste samples were different (P < 0.05). Methionine and lysine contents were similar among different poultry slaughterhouse waste sources. Difference were observed for cystine (1.2e1.7%), threonine (1.9e2.2%), arginine (3e3.5%), leucine (3.5e4.1%) and valine (2.8e3.3%) (P < 0.05). Ruminal degradation rate for dry matter, organic matter and protein were different among poultry slaughterhouse waste, fish meal and roasted soybean. The degradation parameter for protein degradation was 76% for poultry slaughterhouse waste, 79% for fish meal and 98% for roasted soybean (P < 0.05). Results revealed that there was great variation in chemical composition, protein fractioning, and amino acid profiles of different poultry slaughterhouse waste sources. Poultry slaughterhouse waste is slowly-degraded protein in the rumen and thus can be an economical and rich source of rumen undegradable protein in ruminant nutrition. This implies that the use of poultry slaughterhouse waste in ruminant nutrition has a huge potential as a cleaner product of animal feeding and prevention environmental pollution. However, further studies are warranted to evaluate the digestibility of poultry slaughterhouse waste amino acids escaping the rumen into the intestine in ruminants and to compare the biological values for the amino acids in these waste material with common ruminant feedstuffs

    The effects of dietary supplementation with different levels of Microzist as newly developed probiotics on growth performance, carcass characteristics, and immunological organs of broiler chicks

    The current research was conducted to evaluate the influence of newly developed probiotic (Microzist) and commercial probiotic (Primalac) on growth performance, carcass characteristics, and immunity of broiler chickens. A total of 225 one-day-old broiler chicks Ross-308 was randomly assigned into 5 groups – control group, group with 0.2, 0.25, and 0.3 g/kg of Microzist, and group constituting Primalac (starter: 0.9 g/kg, grower: 0.454 g/kg, and finisher: 0.225 g/kg). At 42nd day of age, there were no statistically significant differences among treatments on daily weight gain and feed conversion ratio (P > .05), but treatment’s effect on the feed intake was found to be significant (P < .05). Diet supplementation with various concentrations of Microzist showed growth performance of broiler chickens more or less similar to the Primalac additives. There were statistically significant differences (P < .05) among treatments on the carcass characteristics. Microzist (0.2 g/kg) reduced the abdominal fat content in broiler chickens in a comparison with control as well as Primalac (P < .05). Furthermore, Microzist showed no influence on the immunological organ weights. The present investigation indicates that Microzist may be a potential alternative to antibiotic growth promoters as well as widely used Primalac in broiler industries

    The effects of antibiotic, probiotic, organic acid, vitamin C, and Echinacea purpurea extract on performance, carcass characteristics, blood chemistry, microbiota, and immunity of broiler chickens

    The present context investigated the comparative study on the supplementation of antibiotic, probiotic, organic acid, vitamin C, and herbal extract after vaccination into drinking water and their effects on performance, carcass quality, blood biochemical parameters, immune system, and intestinal flora in broiler chicks for 42 days. A total of 420 one-day-old male broiler chicks (Ross 308) were randomly assigned into 7 treatments with 3 replicates (pens) per treatment and 20 male chicks for each replicate (pen). The experimental treatments consisted of drinking water (control, without additive); drinking water + antibiotic sulfamet; drinking water + CVet- 50; drinking water + antibiotic sulfamet + C-Vet-50; drinking water + probiotic Primalac; drinking water + butyric acid; and drinking water + extract of Echinacea purpurea Moench (coneflower). There were no differences observed among the treatments for feed intake, but during the whole experimental period, the highest body weight gain was found in the chicks fed with drinking water + antibiotic sulfamet + 50 cc vitamin C (P 0.05) observed among the treatments for feed conversion ratio (P > 0.05). Moreover, there were no differences reported among treatments for carcass characteristics at the end of the experiment. Among the treatments, drinking water + 50 cc vitamin C, and drinking water + extract of E. purpurea reduced (P < 0.05) the levels of cholesterol, triglycerides, and low-density lipoproteins. Drinking water + 50 cc vitamin C, drinking water + Primalac, and drinking water + extract of E. purpurea increased (P < 0.05) the lymphocytes count and decreased the heterophils count and heterophil:lymphocyte ratio. The highest Escherichia coli count and lowest Lactobacillus count in ileal content of the broilers were observed in the control group (P < 0.05)

    Peas may be a candidate crop for integrating silvoarable systems and dairy buffalo farming in southern Italy

    Integration of trees with agricultural crops and livestock systems may offer advantages in term of productivity, economic return, and sustainability. In order to identify a candidate crop for Mediterranean silvoarable systems to be used in livestock farming as a protein source locally produced, a study was undertaken to evaluate the effect of feeding peas (Pisum sativum L.) as main protein source on milk yield and in vivo digestibility of primiparous buffaloes. Two almost isonitrogenous concentrates (on average, crude protein 240 g/kg dry matter) were formulated to contain, as fed basis, either 350 g/kg of soybean cake (SoyC) or 450 g/kg of extruded peas (PeaC) as the main protein source. Twenty primiparous buffaloes were blocked by age and body weight into two dietary treatments (Soyand Pea) from 10 to 100 day in milk. All cows were fed in the barn a total mixed ration containing 3 kg of SoyC and in the milking parlour they were individually supplemented by either 3 kg of SoyC or PeaC according to the group assignment. The substitution rate of soybean protein by pea protein was approximately 30%. Milk yield was not affected by the dietary use of extruded peas, as well as, milk fat and protein percentages, and clotting properties. Moreover, in vivo digestibility did not differ between the two dietary groups. Results support the partial substitution of soybean cake with extruded peas in diets for lactating buffaloe

    Molecular Identification of Candida albicans in Endodontic Retreatment Cases by SYBR Green I Real-time Polymerase Chain Reaction and its Association with Endodontic Symptoms

    Statement of the Problem: Recent microbiological studies have expressed ever-increasing concerns about Candida albicans as a causal factor in the failure of endodontic treatments. Real-time quantitative polymerase chain reaction (qPCR), including the SYBR Green I system, is a technique in which a fluorescent dye is incorporated into the double-stranded DNA that is produced during DNA polymerase activity.Purpose: This study aimed to determine the relative prevalence of Candida albicans in root canals of retreatment cases and its association with endodontic symptoms.Materials and Method: In the present cross-sectional/analytical study fifty subjects were selected. Clinical features and radiographic status of the teeth were also evaluated. After access cavity preparation, the retrieved material and dentinal chips removed from the root canal were transferred into 1.5-mL microtubes, followed by storage at -20ºC until used for DNA extraction. A DNeasy Tissue Kit was used to extract DNA using the DNeasy protocol for animal tissues. Master Plus SYBR Green I (Jena Bioscience, Germany) was used in a Rotor-gene Real-time PCR System for real-time PCR. The relationship between the presence of Candida albicans and the clinical and radiographic features were analyzed using McNemar’s test.Results: There was a significant relationship between the radiographic findings in endodontically treated teeth and the presence of Candida albicans. However, there was no significant relationship between the presence of Candida albicans and any of the clinical symptoms.Conclusion: In spite of the limitations of this study, we concluded that Candida albicans was associated with root canal infections in endodontic retreatment cases, but there was no relationship with the clinical symptoms

    Safety Profile of Gutless Adenovirus Vectors Delivered into the Normal Brain Parenchyma: Implications for a Glioma Phase 1 Clinical Trial

    Abstract Adenoviral vectors (Ads) have been evaluated in clinical trials for glioma. However, systemic immunity against the vectors can hamper therapeutic efficacy. We demonstrated that combined immunostimulation and cytotoxic gene therapy provides long-term survival in preclinical glioma models. Because helper-dependent high-capacity Ads (HC-Ads) elicit sustained transgene expression, in the presence of antiadenoviral immunity, we engineered HC-Ads encoding conditional cytotoxic herpes simplex type 1 thymidine kinase and immunostimulatory cytokine Fms-like tyrosine kinase ligand-3 under the control of the TetOn system. Escalating doses of combined HC-Ads (1?108, 1?109, and 1?1010 viral particles [VP]) were delivered into the rat brain. We assessed neuropathology, biodistribution, transgene expression, systemic toxicity, and behavioral impact at acute and chronic time points after vector delivery. Histopathological analysis did not reveal any evidence of toxicity or long-term inflammation at the lower doses tested. Vector genomes were restricted to the injection site. Serum chemistry did not uncover adverse systemic side effects at any of the doses tested. Taken together, our data indicate that doses of up to 1?109 VP of each HC-Ad can be safely administered into the normal brain.     Global, regional, and national burden of chronic kidney disease, 1990–2017 : a systematic analysis for the Global Burden of Disease Study 2017

    Background Health system planning requires careful assessment of chronic kidney disease (CKD) epidemiology, but data for morbidity and mortality of this disease are scarce or non-existent in many countries. We estimated the global, regional, and national burden of CKD, as well as the burden of cardiovascular disease and gout attributable to impaired kidney function, for the Global Burden of Diseases, Injuries, and Risk Factors Study 2017. We use the term CKD to refer to the morbidity and mortality that can be directly attributed to all stages of CKD, and we use the term impaired kidney function to refer to the additional risk of CKD from cardiovascular disease and gout. Methods The main data sources we used were published literature, vital registration systems, end-stage kidney disease registries, and household surveys. Estimates of CKD burden were produced using a Cause of Death Ensemble model and a Bayesian meta-regression analytical tool, and included incidence, prevalence, years lived with disability, mortality, years of life lost, and disability-adjusted life-years (DALYs). A comparative risk assessment approach was used to estimate the proportion of cardiovascular diseases and gout burden attributable to impaired kidney function. Findings Globally, in 2017, 1·2 million (95% uncertainty interval [UI] 1·2 to 1·3) people died from CKD. The global all-age mortality rate from CKD increased 41·5% (95% UI 35·2 to 46·5) between 1990 and 2017, although there was no significant change in the age-standardised mortality rate (2·8%, −1·5 to 6·3). In 2017, 697·5 million (95% UI 649·2 to 752·0) cases of all-stage CKD were recorded, for a global prevalence of 9·1% (8·5 to 9·8). The global all-age prevalence of CKD increased 29·3% (95% UI 26·4 to 32·6) since 1990, whereas the age-standardised prevalence remained stable (1·2%, −1·1 to 3·5). CKD resulted in 35·8 million (95% UI 33·7 to 38·0) DALYs in 2017, with diabetic nephropathy accounting for almost a third of DALYs. Most of the burden of CKD was concentrated in the three lowest quintiles of Socio-demographic Index (SDI). In several regions, particularly Oceania, sub-Saharan Africa, and Latin America, the burden of CKD was much higher than expected for the level of development, whereas the disease burden in western, eastern, and central sub-Saharan Africa, east Asia, south Asia, central and eastern Europe, Australasia, and western Europe was lower than expected. 1·4 million (95% UI 1·2 to 1·6) cardiovascular disease-related deaths and 25·3 million (22·2 to 28·9) cardiovascular disease DALYs were attributable to impaired kidney function. Interpretation Kidney disease has a major effect on global health, both as a direct cause of global morbidity and mortality and as an important risk factor for cardiovascular disease. CKD is largely preventable and treatable and deserves greater attention in global health policy decision making, particularly in locations with low and middle SDI
