    Distribution, morphology, seasonal dynamics, and molecular characterization of Tylenchulus semipenetrans from citrus orchards in southern Iran

    Tylenchulus semipenetrans was identified morphologically from root samples collected from citrus orchards in Shahdad region, southeastern Iran. The number of juveniles and females averaged 648/100 g soil and 65/10 g root, respectively, in infected samples. Monitoring these orchards for two years (2012-2013) indicated no significant correlation between soil pH (7.21-7.70) and EC (0.56-1.98) values and populations densities of T. semipenetrans. With respect to seasonal distribution, T. semipenetrans attained the highest density during fall and the lowest in spring. Sequence analysis of the D2-D3 segment of 28S rDNA confirmed this population as T. semipenetrans. Nblast indicates that the sequence from this population differs in only three or five nucleotides from corresponding sequences from T. semipenetrans populations from Korea (FJ969715, FJ060713, and FJ969712), with 99% identity.     The distribution of nematodes was studied in rice fields in Guilan province, Iran, from 2014 to 2016. Nematode biodiversity of 250 soil and root samples was examined. Thirty nematode species were identified morphologically, including plant parasites, microbivores and mycetophagous and predator species. Molecular techniques were also used for further identification of three plant parasitic species. Indicators of population were also estimated. Spiral nematodes (Helicotylenchus crenacauda Sher, 1966 and H. digitiformis Ivanova, 1967) and stunt nematode (Tylenchorhynchus agri) were the predominant parasitic species identified. Among other species, three mononchid species were identified namely Mononchus aquaticus, Mylonchulus sigmaturus and M. polonicus. The species M. polonicus was found and reported in Iran for the first time. The two plant parasitic species T. agri and Xiphinema index were reported in association with rice in Iran for the first time. To evaluate the distribution and incidence of Aphelenchoides besseyi (rice white-tip nematode) in different regions of Guilan province, a total of 255 fresh seed samples were collected/inspected, of which, about 40% of them were infested with A. besseyi. Of the 16 studied counties, the highest percentage of infected seeds came from fields around the city of Astara (69.2%) and the second highest infection was observed near the city of Anzali (60%)

    Reaction of Commercial Cultivars of Kiwifruit to Infection by Root-knot Nematode and Its Biocontrol Using Endophytic Bacteria

    Root-knot nematodes (RKN) cause considerable economic losses to kiwifruit production annually. Screening of resistant cultivars has been one of the long-standing methods to manage root-knot nematodes. Here, the reaction of the four most common commercial cultivars of kiwifruit, namely, Actinidia chinensis var. deliciosa cv. Hayward, A. chinensis var. deliciosa cv. Abbott, A. chinensis var. deliciosa cv. Bruno, and A. chinensis var. chinensis cv. Haegeum (commonly known as ‘Golden’ kiwifruit) to infection by the RKN, Meloidogyne incognita, was evaluated. Among examined cultivars ‘Golden’ was the most susceptible, having on average 52.8 galls, 56.1 egg masses per gram of root, and 642 J2 population per 200 gram of soil. ‘Bruno’ showed the highest resistance, with 3.3 galls, 4.1 egg masses per gram of root, and 79 J2 in 200 g of soil. Then, two potential biological control agents, namely Priestia megaterium 31.en and Agrobacterium tumefaciens 19.en were used on ‘Hayward’ seedlings against M. incognita and showed a significant reduction in the number of galls and egg masses on roots, juvenile population in the soil, and increased the growth parameters of the plants compared to non-treated seedlings. We demonstrated that integrated management using resistant cultivars and biological control can provide a safe and economic method to control RKN, and these resistant cultivars can be used in breeding programs

    Molecular Characterization of Three B-1,4-Endoglucanase Genes in Pratylenchus loosi and Functional Analysis of Pl-eng-2 Gene

    Pratylenchus loosi is an important root-lesion nematode that causes damage to tea plantations in Iran and all over the world. The present study reports on the characterization and evolution of three ß-1,4-endoglucanase genes: Pl-eng-2, Pl-eng-3 and Pl-eng-4. The gene structure of Pl-eng-2 was fully determined with the predicted signal peptide and devoid of the linker domain and carbohydrate-binding domain, while Pl-eng-3 and Pl-eng-4 were only partially sequenced. The transcription of Pl-eng-2 was localized in the secretory esophageal glands of all life stages, but it was upregulated in male and female stages. The exon/intron structures of Pl-eng-2, Pl-eng-3 and Pl-eng-4 confirmed that they resulted from gene duplication followed by sequence and gene structure diversification with loss of the linker domain and carbohydrate-binding domain during evolution. A phylogenetic analysis further confirmed that nematode endoglucanases resulted from the horizontal gene transfer of a bacterial gene, as Pl-eng-3 showed sister relationships with the CelB cellulase of Bacillus subtilis. Silencing Pl-eng-2 by in vitro RNA interference produced a 60% decrease of the transcript level. The reproductive ability of silenced P. loosi showed a 35% reduction of eggs and larval stages compared to untreated nematodes, suggesting that this gene is involved in the early steps of invasion
