    The Neogene Savannas of North America : A Retrospective Analysis on Artiodactyl Faunas

    Savanna-like ecosystems were present at high latitudes in North America during much of the Neogene. Present-day African savannas, like the Serengeti, have been proposed to be modern analogs of these paleosavannas, particularly those from the middle Miocene of the Great Plains region of the United States. Both these extant and extinct savannas contain a preponderance of artiodactyl (even-toed ungulate) species; however, the taxonomic composition of each fauna is different. While present-day African savannas are dominated by ruminants (primarily bovids), the Neogene savannas of North America were dominated by a diversity of both camelid and non-bovid ruminant families. This study provides a quantitative test of the similarity of the artiodactyl faunas of the North American Neogene paleosavannas to those of the modern-day African savannas. A correspondence analysis of ecomorphological features revealed considerable overlap between modern and fossil faunas. The morphospace occupation of the extinct North American ruminants falls within that of the African bovids. Some of the extinct camelids also fall within this same morphospace, but many do not, perhaps indicating an environmental difference such as greater aridity in Neogene North America. The diversity and disparity of artiodactyl faunas through the Neogene of North America changed along with changing temperatures and precipitation regimes. The taxonomic and ecomorphological diversity of the Serengeti ruminant fauna is statistically comparable to those North American paleofaunas occurring during or immediately after the Middle Miocene Climatic Optimum (MMCO), but the later, more depauperate faunas are no longer comparable.     Molecular docking analysis of phytoconstituents of Illicium verum fruit against Caspase 3, MMP-9 and TNF-α

    510-519Present study was designed to evaluate the anti-apoptotic activity and anti-inflammatory of phytocompounds of Illicium verum fruits. Bioactive compounds from the alcoholic, chloroform and ethylacteate fractions of Illicium verum were selected based on GC-MS analysis, which were in silico docked against Caspase 3, Matrix Metalloproteinase 9 and TNF-α. The structures of Caspase 3, Matrix Metalloproteinase 9 (MMP-9) and TNF-α enzymes were obtained from the protein data bank. Molecular docking was done by using Glide XP module of Schrodinger software and binding energies of ligands were also calculated. The phytocompound Strychane,1-acetyl-20α- hydroxy-16-methylene showed good docking score against Caspase 3 and TNF-α. The phytocompound Pyrrolidine, 1-(7-oxo-2,4,6 trimethylheptanoyl) showed good docking score against MMP-9. The results revealed that phytoconstituents obtained from Illicium verum showed good docking against Caspase-3, MMP-9 and TNF-α indicating their potential towards apoptosis and inflammation

    Polycythemia is uncommon in dogs with chronic hypoxic pulmonary disease

    Background Prolonged tissue hypoxia caused by chronic pulmonary disease is commonly regarded as an important mechanism in the development of secondary polycythemia, but little clinical data are available to support this hypothesis. Objective To study the prevalence and severity of erythrocytosis accompanying chronic hypoxic pulmonary disease in dogs. Animals Forty-seven dogs with hypoxic chronic pulmonary disease, 27 dogs with nonhypoxic chronic pulmonary disease, and 60 healthy controls. Methods Dogs with chronic pulmonary disease and chronic hypoxemia (partial pressure of arterial oxygen [PaO2] = 65%) was noted in any of the dogs. Red blood cell parameters were not associated with the severity of hypoxemia (correlation to PaO2: Erytr, r = -.14; Hb, r = -.21; Hct, r = -.14; P > .05 for all). Conclusions and Clinical Importance Polycythemia is uncommon, and usually mild if present, in dogs with chronic hypoxia caused by pulmonary disease.Peer reviewe

    Thoracic high resolution CT using the modified VetMousetrap (TM) device is a feasible method for diagnosing canine idiopathic pulmonary fibrosis in awake West Highland White Terriers

    Canine idiopathic pulmonary fibrosis is a chronic, progressive interstitial lung disease particularly prevalent in West Highland White Terriers. In the present prospective pilot study, we evaluated the feasibility of modified VetMousetrap (TM) device in high resolution CT to detect idiopathic pulmonary fibrosis in West Highland White Terriers. Twelve awake West Highland White Terriers with canine idiopathic pulmonary fibrosis and 24 clinically healthy West Highland White Terriers were scanned using a helical dual slice scanner utilizing VetMousetrap (TM) device without or with minimal chemical restraint with butorphanol. Three evaluators blindly assessed the images for image quality and the presence of canine idiopathic pulmonary fibrosis related imaging findings such as ground glass opacity and reticular opacities. Additionally, the attenuation of the lung was quantified with ImageJ software using histogram analysis of density over the lung fields. Computed tomography was successfully completed and motion artifact ranked in statistical analysis barely noticeable to mild in all dogs. The agreement between imaging findings and clinical status was very good with overall kappa value 0.91 and percentage of agreement of 94%. There was also very good intraobserver (kappa(range) = 0.79-0.91) and interobserver agreement (kappa = 0.94). Moderate to severe ground glass opacity was present in all affected dogs. In the ImageJ analysis, a significant difference in lung attenuation between the study groups was observed. We conclude that modified VetMousetrap (TM) device is applicable in diagnosing canine idiopathic pulmonary fibrosis in awake West Highland White Terriers avoiding anesthetic risk in these often severely hypoxic patients.Peer reviewe

    Ultra-high frequency ultrasound delineated changes in carotid and muscular artery intima-media and adventitia thickness in obese early middle-aged women

    Obesity is linked to increased arterial size, carotid intima-media thickness and arterial stiffness. The effects of obesity and body composition on muscular artery intima-media and adventitia thickness has previously not been established. The aim of this study was to explore associations between carotid and muscular artery wall layer thickness with body composition and cardiovascular risk factors in early middle-aged women. This is a cross-sectional study including 199 women aged 40 +/- 4 years. Arterial lumen (LD), intima-media (IMT) and adventitia thickness (AT) were measured from carotid, brachial and radial arteries using ultra-high frequency ultrasound (22-71 MHz). Women with obesity had increased IMT in carotid (0.47 vs 0.45 mm), brachial (0.19 vs 0.17 mm) and radial arteries (0.16 vs 0.15 mm) and increased brachial AT (0.14 vs 0.13 mm). In multiple regression models all arterial LD (beta-range 0.02-0.03 mm/kg/m(2)), IMT (beta-range 0.91-3.37 mu m/kg/m(2)), AT (beta-range 0.73-1.38 mu m/kg/m(2)) were significantly associated with BMI. The IMT of all arteries were significantly associated with systolic blood pressure (beta-range 0.36-0.85 mu m/mmHg), attenuating the association between IMT and BMI (beta-range 0.18-2.24 mu m/kg/m(2)). Obese early middle-aged women have increased arterial intima media thickness and brachial artery adventitia thickness compared to non-obese counterparts. The association between BMI and intima-media thickness is partly mediated through blood pressure levels.Peer reviewe

    Macronutrient intake during pregnancy in women with a history of obesity or gestational diabetes and offspring adiposity at 5 years of age

    Background/objectives The impact of maternal macronutrient intake during pregnancy on offspring childhood adiposity is unclear. We assessed the associations between maternal macronutrient intake during and after pregnancy with offspring adiposity at 5 years of age. Additionally, we investigated whether gestational diabetes (GDM), BMI, or breastfeeding modified these associations. Subjects/methods Altogether, 301 mother-child dyads with maternal prepregnancy BMI >= 30 and/or previous GDM participated in the Finnish Gestational Diabetes Prevention Study (RADIEL) and its 5 years follow-up. Macronutrient intakes (E%) were calculated from 3-day food records collected at 5-18 weeks' gestation, in the third trimester, and at 12 months and 5 years after pregnancy. Offspring body fat mass (BFM) and fat percentage (BF%) at 5 years were measured by bioimpedance. Statistical analyses were multivariate linear regression. Results Mean (SD) prepregnancy BMI was 33(4) kg/m(2). GDM was diagnosed in 47%. In normoglycemic women, higher first half of pregnancy n-3 PUFA intake was associated with lower offspring BFM (g) (ss -0.90; 95% CI -1.62, -0.18) and BF% (ss -3.45; 95% CI -6.17, -0.72). In women with GDM, higher first half of pregnancy n-3 PUFA intake was associated with higher offspring BFM (ss 0.94; 95% CI 0.14, 1.75) and BF% (ss 3.21; 95% CI 0.43, 5.99). Higher SFA intake in the third trimester and cumulative intake across pregnancy (mean of the first half and late pregnancy) was associated with higher BFM and BF% (across pregnancy: ss 0.12; 95% CI 0.03, 0.20 and ss 0.44; 95% CI 0.15, 0.73, respectively). Higher carbohydrate intake across pregnancy was associated with lower BFM (ss -0.044; 95% CI -0.086, -0.003), and borderline associated with BF% (ss -0.15; 95% CI -0.31, 0.00). Conclusions The macronutrient composition of maternal diet during pregnancy is associated with offspring BFM and BF% at 5 years. GDM modifies the association between prenatal n-3 PUFA intake and offspring anthropometrics.Peer reviewe

    No effect of gestational diabetes or pre-gestational obesity on 6-year offspring left ventricular function-RADIEL study follow-up

    Aims We aimed to investigate associations between pre-pregnancy obesity, gestational diabetes (GDM), offspring body composition, and left ventricular diastolic and systolic function in early childhood. Methods This is an observational study, including 201 mother-child pairs originating from the Finnish Gestational Diabetes Prevention Study (RADIEL; 96 with GDM, 128 with pre-pregnancy obesity) with follow-up from gestation to 6-year postpartum. Follow-up included dyads anthropometrics, body composition, blood pressure, and child left ventricular function with comprehensive echocardiography (conventional and strain imaging). Results Offspring left ventricular diastolic and systolic function was not associated with gestational glucose concentrations, GDM, or pregravida obesity. Child body fat percentage correlated with maternal pre-pregnancy BMI in the setting of maternal obesity (r = 0.23,P = 0.009). After adjusting for child lean body mass, age, sex, systolic BP, resting HR, maternal lean body mass, pre-gestational BMI, and GDM status, child left atrial volume increased by 0.3 ml (95% CI 0.1, 0.5) for each 1% increase in child body fat percentage. Conclusions No evidence of foetal cardiac programming related to GDM or maternal pre-pregnancy obesity was observed in early childhood. Maternal pre-pregnancy obesity is associated with early weight gain. Child adiposity in early childhood is independently associated with increased left atrial volume, but its implications for long-term left ventricle diastolic function and cardiovascular health remain unknown.Peer reviewe

    Maternal obesity and gestational diabetes : Impact on arterial wall layer thickness and stiffness in early childhood - RADIEL study six-year follow-up

    Background and aims: Gestational diabetes (GDM) and maternal obesity are linked to weight gain in childhood and an increased risk of cardiovascular disease later in life. We assessed the effects of GDM and maternal obesity on arterial function and morphology in relation to body anthropometrics and composition in early childhood. Methods: We assessed body size and composition, blood pressure (BP), arterial morphology and stiffness in 201 pairs of obese mothers (pre-pregnancy BMI 30.7 +/- 5.6 kg/m(2), 96 with GDM) and their children at 6.1 years (SD 0.5). Results: Child BMI (z-score 0.45 +/- 0.92; p <0.001) and common carotid intima-media thickness (IMT, z-score 0.15 +/- 0.75, p=0.003) were increased compared with a healthy Finnish reference population. No associations with maternal GDM was found. Carotid IMT and pulse wave velocity were unrelated to child sex, anthropometrics, body composition, BP, as well as maternal anthropometrics and body composition. Carotid stiffness was independently predicted by second trimester fasting glucose. Child lean body mass was the strongest independent predictor for radial (RA), and brachial artery (BA) lumen diameter (LD) and BA IMT (LD: RA: r(2)=0.068, p <0.001; BA: r(2)=0.108, p <0.001; IMT: BA: r(2)=0.161, p <0.001) and carotid LD (r(2)=0.066, p <0.001). Conclusions: Children of obese mothers have increased BMI, blood pressure and carotid IMT suggesting a transgenerational effect of maternal obesity and clustering of cardiovascular risk factors in the population. Arterial dimensions were mainly predicted by child LBM, and not associated with maternal or child adiposity, or GDM. There was a weak association with maternal gestational fasting glucose and increased carotid artery stiffness.Peer reviewe

    Case Study Comparing Effects of Microplastic Derived from Bottle Caps Collected in Two Cities on Triticum aestivum (Wheat)

    As plastic has become an integral component of daily life, microplastic has become a ubiquitous, unavoidable constituent of nearly all ecosystems. Besides monitoring the amount and distribution of microplastic in the environment, it is necessary to understand the possible direct effects, especially toxicity and how it is affected by environmental factors where it is discarded. The present study investigated how microplastic derived from high-density polyethylene bottle caps collected in two climatically different cities, i.e., Singapore (tropical rainforest climate) and Lahti, Finland (continental climate), affected the essential agricultural grain crop, Triticum aestivum (L.). Wheat seedlings were exposed to microplastic derived from these collected bottle caps, as well as new and artificially aged caps, for seven days. Morphological parameters, such as root and shoot length and oxidative stress development, were measured. Exposure to microplastic derived from the caps resulted in reduced seedling root and shoot lengths compared to the controls, as well as enhanced lipid peroxidation and catalase activity. With all parameters tested, microplastic derived from Lahti bottle caps exhibited more severe effects than Singapore, which was similar to that elicited by new microplastic. The Singapore microplastic had possibly leached its toxic substances before collection due to accelerated degradation promoted by the prevailing warmer climate conditions
