Constraints on the Quasiparticle Density of States in High- Superconductors
In this Letter we present new tunneling data on YBaCuO thin films
by low temperature scanning tunneling spectroscopy. Unusual peak-dip-hump
features, previously reported in BiSrCaCuO, are also
found in YBaCuO. To analyse these common signatures we propose a
new heuristic model in which, in addition to the d-wave symmetry, the gap
function is energy dependent. A simple expression for the quasiparticle density
of states is derived, giving an excellent agreement with the experiment. The
dynamics of the quasiparticle states and the energy scales involved in the

Cosmic Calibration: Constraints from the Matter Power Spectrum and the Cosmic Microwave Background
Several cosmological measurements have attained significant levels of
maturity and accuracy over the last decade. Continuing this trend, future
observations promise measurements of the statistics of the cosmic mass
distribution at an accuracy level of one percent out to spatial scales with
k~10 h/Mpc and even smaller, entering highly nonlinear regimes of gravitational
instability. In order to interpret these observations and extract useful
cosmological information from them, such as the equation of state of dark
energy, very costly high precision, multi-physics simulations must be
performed. We have recently implemented a new statistical framework with the
aim of obtaining accurate parameter constraints from combining observations
with a limited number of simulations. The key idea is the replacement of the
full simulator by a fast emulator with controlled error bounds. In this paper,
we provide a detailed description of the methodology and extend the framework
to include joint analysis of cosmic microwave background and large scale
structure measurements. Our framework is especially well-suited for upcoming
large scale structure probes of dark energy such as baryon acoustic
oscillations and, especially, weak lensing, where percent level accuracy on

Scanning Tunneling Spectroscopy on the novel superconductor CaC6
We present scanning tunneling microscopy and spectroscopy of the newly
discovered superconductor CaC. The tunneling conductance spectra, measured
between 3 K and 15 K, show a clear superconducting gap in the quasiparticle
density of states. The gap function extracted from the spectra is in good
agreement with the conventional BCS theory with = 1.6 0.2
meV. The possibility of gap anisotropy and two-gap superconductivity is also
discussed. In a magnetic field, direct imaging of the vortices allows to deduce
a coherence length in the ab plane 33 nm
TagDigger: user-friendly extraction of read counts from GBS and RAD-seq data
Background: In genotyping-by-sequencing (GBS) and restriction site-associated DNA sequencing (RAD-seq), read depth is important for assessing the quality of genotype calls and estimating allele dosage in polyploids. However, existing pipelines for GBS and RAD-seq do not provide read counts in formats that are both accurate and easy to access. Additionally, although existing pipelines allow previously-mined SNPs to be genotyped on new samples, they do not allow the user to manually specify a subset of loci to examine. Pipelines that do not use a reference genome assign arbitrary names to SNPs, making meta-analysis across projects difficult.
Results: We created the software TagDigger, which includes three programs for analyzing GBS and RAD-seq data. The first script, tagdigger_interactive.py, rapidly extracts read counts and genotypes from FASTQ files using user-supplied sets of barcodes and tags. Input and output is in CSV format so that it can be opened by spreadsheet software. Tag sequences can also be imported from the Stacks, TASSEL-GBSv2, TASSEL-UNEAK, or pyRAD pipelines, and a separate file can be imported listing the names of markers to retain. A second script, tag_manager.py, consolidates marker names and sequences across multiple projects. A third script, barcode_splitter.py, assists with preparing FASTQ data for deposit in a public archive by splitting FASTQ files by barcode and generating MD5 checksums for the resulting files.
Conclusions: TagDigger is open-source and freely available software written in Python 3. It uses a scalable, rapid search algorithm that can process over 100 million FASTQ reads per hour. Low temperature exposure induces browning of bone marrow stem cell derived adipocytes in vitro
Brown and beige adipocytes are characterised as expressing the unique mitochondrial uncoupling protein (UCP)1 for which the primary stimulus in vivo is cold exposure. The extent to which cold-induced UCP1 activation can also be achieved in vitro, and therefore perform a comparable cellular function, is unknown. We report an in vitro model to induce adipocyte browning using bone marrow (BM) derived mesenchymal stem cells (MSC), which relies on differentiation at 32 °C instead of 37 °C. The low temperature promoted browning in adipogenic cultures, with increased adipocyte differentiation and upregulation of adipogenic and thermogenic factors, especially UCP1. Cells exhibited enhanced uncoupled respiration and metabolic adaptation. Cold-exposed differentiated cells showed a marked translocation of leptin to adipocyte nuclei, suggesting a previously unknown role for leptin in the browning process. These results indicate that BM-MSC can be driven to forming beige-like adipocytes in vitro by exposure to a reduced temperature. This in vitro model will provide a powerful tool to elucidate the precise role of leptin and related hormones in hitherto functions in the browning process
A proposed approach to monitor private-sector policies and practices related to food environments, obesity and non-communicable disease prevention
Private-sector organizations play a critical role in shaping the food environments
of individuals and populations. However, there is currently
very limited independent monitoring of private-sector actions related to
food environments. This paper reviews previous efforts to monitor the
private sector in this area, and outlines a proposed approach to monitor
private-sector policies and practices related to food environments, and
their influence on obesity and non-communicable disease (NCD) prevention.
A step-wise approach to data collection is recommended, in which
the first (‘minimal’) step is the collation of publicly available food and
nutrition-related policies of selected private-sector organizations. The
second (‘expanded’) step assesses the nutritional composition of each
organization’s products, their promotions to children, their labelling
practices, and the accessibility, availability and affordability of their
products. The third (‘optimal’) step includes data on other commercial
activities that may influence food environments, such as political lobbying
and corporate philanthropy. The proposed approach will be further
developed and piloted in countries of varying size and income levels.
There is potential for this approach to enable national and international
benchmarking of private-sector policies and practices, and to inform
efforts to hold the private sector to account for their role in obesity and
NCD prevention
Market strategies used by processed food manufacturers to increase and consolidate their power: a systematic review and document analysis
Background
The public health community has become increasingly critical of the role that powerful corporations play in driving unhealthy diets, one of the leading contributors to the global burden of disease. While a substantial amount of work has examined the political strategies used by dominant processed food manufacturers that undermine public health, less attention has been paid to their use of market strategies to build and consolidate power. In this light, this paper aimed to systematically review and synthesise the market strategies deployed by dominant processed food manufacturers to increase and consolidate their power.
Methods
A systematic review and document analysis of public health, business, legal and media content databases (Scopus, Medline, ABI Inform, Business Source Complete, Thomas Reuters Westlaw, Lexis Advance, Factiva, NewsBank), and grey literature were conducted. Data extracted were analysed thematically using an approach informed by Porter’s ‘Five Forces’ framework.
Results
213 documents met inclusion criteria. The market strategies (n=21) and related practices of dominant processed food manufacturers identified in the documents were categorised into a typological framework consisting of six interconnected strategic objectives: i) reduce intense competition with equivalent sized rivals and maintaining dominance over smaller rivals; ii) raise barriers to market entry by new competitors; iii) counter the threat of market disruptors and drive dietary displacement in favour of their products; iv) increase firm buyer power over suppliers; v) increase firm seller power over retailers and distributors; and vi) leverage informational power asymmetries in relations with consumers.
Conclusions
The typological framework is well-placed to inform general and jurisdiction-specific market strategy analyses of dominant processed food manufacturers, and has the potential to assist in identifying countervailing public policies, such as those related to merger control, unfair trading practices, and public procurement, that could be used to address market-power imbalances as part of efforts to improve population diets

IQGAP1 and IQGAP2 are Reciprocally Altered in Hepatocellular Carcinoma
<p>Abstract</p> <p>Background</p> <p>IQGAP1 and IQGAP2 are homologous members of the IQGAP family of scaffold proteins. Accumulating evidence implicates IQGAPs in tumorigenesis. We recently reported that IQGAP2 deficiency leads to the development of hepatocellular carcinoma (HCC) in mice. In the current study we extend these findings, and investigate IQGAP1 and IQGAP2 expression in human HCC.</p> <p>Methods</p> <p>IQGAP1 and IQGAP2 protein expression was assessed by Western blotting and immunohistochemistry. IQGAP mRNA was measured by quantitative RT-PCR. The methylation status of the <it>Iqgap2 </it>promoter was determined by pyrosequencing of bisulfite-treated genomic DNA.</p> <p>Results</p> <p>IQGAP1 and IQGAP2 expression was reciprocally altered in 6/6 liver cancer cell lines. Similarly, immunohistochemical staining of 82 HCC samples showed that IQGAP2 protein expression was reduced in 64/82 (78.0%), while IQGAP1 was present in 69/82 (84.1%). No IQGAP1 staining was detected in 23/28 (82.1%) normal livers, 4/4 (100.0%) hepatic adenomas and 23/23 (100.0%) cirrhosis cases, while IQGAP2 was increased in 22/28 (78.6%), 4/4 (100.0%) and 23/23 (100.0%), respectively. Although the <it>Iqgap2 </it>promoter was not hypermethylated in HCC at any of the 25 CpG sites studied (N = 17), IQGAP2 mRNA levels were significantly lower in HCC specimens (N = 23) than normal livers (N = 6).</p> <p>Conclusions</p> <p>We conclude that increased IQGAP1 and/or decreased IQGAP2 contribute to the pathogenesis of human HCC. Furthermore, downregulation of IQGAP2 in HCC occurs independently of hypermethylation of the <it>Iqgap2 </it>promoter. Immunostaining of IQGAP1 and IQGAP2 may aid in the diagnosis of HCC, and their pharmacologic modulation may represent a novel therapeutic strategy for the treatment of liver cancer.</p
FcγRIII and FcγRIV are indispensable for acute glomerular inflammation induced by switch variant monoclonal antibodies
The relative ability of IgG subclasses to cause acute inflammation, and the roles of specific effector mechanisms in this process is not clear. We explored this in an in vivo model of glomerular inflammation in the mouse. TNP was planted on the glomerular basement membrane after conjugation to nephrotoxic antibody. The relative nephritogenicity of anti-TNP switch-variant monoclonal antibodies was then explored and shown to be IgG2a>IgG2b, with no disease caused by IgG1. Using knockout mice, we showed that FcγRIII was necessary for both neutrophil influx and glomerular damage induced by IgG2a and IgG2b. Surprisingly IgG1 did not cause disease although it binds to FcγRIII. Using blocking antibodies, we showed that this was explained by an additional requirement for FcγRIV which does not bind to IgG1. IgG2a or IgG2b induced neutrophil influx was not affected by deficiency of either FcγRI or C3. Bone marrow chimeras were constructed to test the effect of combined deficiency of FcγRI and C3, and there was no effect on IgG2a or IgG2b mediated neutrophil influx. However, IgG2b-induced albuminuria and thrombosis was reduced in C3 deficient mice, showing an additional role for complement in IgG2b-mediated glomerular damage. The results show that IgG2a and IgG2b are the pathogenic subclasses in acute neutrophil-mediated glomerular inflammation, with an indispensible role for both FcγRIII and FcγRIV. In addition complement contributes to IgG2b induced glomerular injury
