    Analysis of tomato glycoalkaloids by liquid chromatography coupled with electrospray ionization tandem mass spectrometry.

    Steroidal glycoalkaloids (SGAs) extracted from tomato leaves and berries (Lycopersicon esculentum Mill.) were separated and identified using optimized reversed-phase liquid chromatography with electrospray ionization (ESI) and ion trap mass spectrometry (ITMS). The ESI source polarity and chromatographic conditions were evaluated. The ESI spectra contain valuable information, which includes the mass of SGAs, the mass of the aglycones, and several characteristic fragment ions. Cleavage at the interglycosidic bonds proximal to the aglycones is the most prominent process in the ESI process. A protonated molecule, [M+H]+, accompanied by a mixed adduct ion, [M+H+Na]2+, was observed for a-tomatine (i.e., m/z 1034.7 and 528.9) and dehydrotomatine (i.e., m/z 1032.6 and 527.9) in positive ion mode spectra. The structures of these tomato glycoalkaloids were confirmed using tandem mass spectrometry. The identification of a new a-tomatine isomer glycoalkaloid, named filotomatine (MW 1033), which shares a common tetrasaccharide structure (i.e., lycotretraose) with a-tomatine and dehydrotomatine, and soladulcidine as an aglycone, is described for the first time. It occurs in significant amounts in the extracts of wild tomato foliage.     Fungal contamination in one hotel room: Does carpet coating in the floor enhance fungal contamination?

    Study developed in order to know the carpet influence when used in the floor of a hotel room. Twelve air samples of 250L (six in a room with carpet and six more in a room with wood floor) were collected through an impaction method with a flow rate of 140 L/min onto malt extract agar (MEA) supplemented with chloramphenicol (0.05%), using the Millipore air Tester (Millipore), during cleaning activities. Outdoor sample was also performed to be used as a reference. Surface samples from floor and desks, taken at the same time, were collected by the swabbing method. to 7 days. Besides fungal contamination, we also assessed particulate matter contamination in both rooms during the same cleaning tasks. In the analyzed sur- faces, isolates belonging to Aspergillus fumigatus complex were the only fungi found in the carpeted room, whereas in the other room we found Penicllium sp. (63.6%) and Aspergillus sp. (13.6%) as the most frequent genera. In the case of particles the room with carpet obtained significant higher values for both metrics (PMC and PNC), showing that carpet may has influence on particles’ contamination of the room

    Glucosinolates (GLSs) are sulfur-rich plant secondary metabolites which occur in a variety of cruciferous vegetables and among various classes of them, genus Brassica exhibits a rich family of these phytochemicals at high, medium and low abundances. Liquid chromatography (LC) with electrospray ionization in negative ion mode (ESI-) coupled to a hybrid quadrupole lineariontrap (LTQ) and Fouriertransformioncyclotronresonancemass spectrometer (FTICRMS) was employed for the selective and sensitive determination of intact GLSs in crude sample extracts of broccoli (Brassica oleracea L. Var. italica), cauliflower (B. oleracea L. Var. Botrytis) and rocket salad (Eruca sativa L.) with a wide range of contents. When LTQ and FTICR mass analyzers are compared, the magnitude of the limit of detection was ca. 5/6-fold lower with the FTICR MS. In addition, the separation and detection by LC–ESI-FTICR MS provides a highly selective assay platform for unambiguous identification of GLSs, which can be extended to lower abundance (minor) GLSs without significant interferences of other compounds in the sample extracts. The analysis of Brassicaceae species emphasized the presence of eight minor GLSs, viz. 1-methylpropyl-GLS, 2-methylpropyl-GLS, 2-methylbutyl-GLS, 3-methylbutyl-GLS, n-pentyl-GLS, 3-methylpentyl-GLS, 4-methylpentyl-GLS and n-hexyl-GLS. The occurrence of these GLSs belonging to the saturated aliphatic side chain families C4, C5 and C6, presumably formed by chain elongation of leucine, homoleucine and dihomoleucine as primary amino acid precursors, is described. Based on their retention behavior and tandem MS spectra, all these minor compounds occurring in plant extracts of B. oleracea L. Var. italica, B. oleracea L. Var. Botrytis and E. sativa L. were tentatively identified

    O objetivo deste estudo é compreender a função do ambulatório na dinâmica das relações de poder e na construção de identidade da profissão de terapeuta da biomedicina. Para a realização do trabalho foram feitas entrevistas abertas e observação etnográfica em duas unidades hospitalares da cidade do Rio de Janeiro (Hospital Pedro Ernesto, Posto de Atendimento Médico São Francisco Xavier) com 12 médicos. O ambulatório surge no discurso dos profissionais em início de carreira como instância negativa e monótona, que impede o diagnóstico de novas patologias, tornando lenta a ascensão profissional; surge também como espécie de rito de passagem formador da identidade médica. Marcado pela frequência de pacientes oriundos dos estratos sociais mais baixos, essa dimensão pública das instituições médicas repercute relações de dominação social e simbólica inerentes à nossa sociedade

    El presente artículo refleja una investigación llevada a cabo en Lima (Perú) entre profesores de la Universidad de Granada, de la Universidad Nacional Mayor de San Marcos y profesionales locales que trabajan en el área. El trabajo se orienta al análisis de las niñas y los niños de la calle desde las representaciones sociales, haciendo aproximaciones al contexto, y procurando construir unos rasgos tipológicos del colectivo con relación al consumo de drogas, especialmente del alcohol, tabaco e inhalantes. Los resultados evidencian a unos menores con grandes carencias y necesidades básicas, en un contexto de alto riesgo y vulneración, en el que la relación con las drogas se transmite a través de beneficios percibidos y la satisfacción de tales necesidades. Las representaciones sociales son valoraciones llenas de benignidad y otorgan a las drogas propiedades medicinales, ayudas personales, sociales, etc. No obstante, son barreras que limitan o impiden desarrollar sus potencialidades y disfrutar el hecho de ser niño. Finalmente, el trabajo permite un acercamiento a las representaciones sociales de difícil abordaje, aportando un análisis más allá de los hechos concretos, y ofreciendo las bases para la acción desde modelos socioeducativos de transferencia de los beneficios, símbolos y percepciones atribuidos a las sustancias

    Absolute Hyperglycemia versus Stress Hyperglycemia Ratio for the Prognosis of Hospitalized Patients with COVID-19 in the First Months of the Pandemic: A Retrospective Study

    Diabetes is a risk factor for greater severity of coronavirus disease 2019 (COVID-19). The stress hyperglycemia ratio (SHR) is an independent predictor of critical illness, and it is reported to have a stronger association than absolute hyperglycemia. The aim of this study was to assess the relationship of absolute hyperglycemia and SHR with the severity of COVID-19, since there are no studies investigating SHR in patients with COVID-19. We conducted a retrospective observational study on hospitalized patients with COVID-19 in the first months of the pandemic, regarding absolute hyperglycemia, SHR, and severity outcomes. Of the 374 patients, 28.1% had a previous diagnosis of type 2 diabetes. Absolute hyperglycemia (64.8% versus 22.7%; p < 0.01) and SHR [1.1 (IQR 0.9-1.3) versus 1.0 (IQR 0.9-1.2); p < 0.001] showed a statistically significant association with previous diabetes. Absolute hyperglycemia showed a significant association with clinical severity of COVID-19 (79.0% versus 62.7%; p < 0.001), need for oxygen therapy (74.8% versus 54.4%; p < 0.001), invasive mechanical ventilation (28.6% versus 11.6%; p < 0.001), and intensive care unit (30.3% versus 14.9%; p = 0.002), but not with mortality; by contrast, there was no statistically significant association between SHR and all these parameters. Our results are in agreement with the literature regarding the impact of absolute hyperglycemia on COVID-19 severity outcomes, while SHR was not a significant marker. We therefore suggest that SHR should not be evaluated in all patients admitted in the hospital for COVID-19, and we encourage the standard measures at admission of blood glucose and HbA1c levels.info:eu-repo/semantics/publishedVersio

    Accessing occupational exposure to fungi in a cork industry

    In this study we aimed to access fungal exposure in workers from one cork industry through the mycological analysis of their nasal exudate and the environmental fungal contamination of their surroundings as well. Nasal mucous samples from 127 workers were taken with sterilized cotton swabs.The fungal species identified in the collected nose swabs were shown to be correlated with the results obtained in the environment. Eighty workers (63.0%) presented contamination of their nose nostril with Chrysonilia sitophila, which number of colonies was countless. Within the Aspergillus genus, the complexes Fumigati, Circumdati, Versicolores and Candidi were isolated. No azole-resistant Aspergillus isolates grew in the selective media used (screened itraconazole and voriconazole resistance).This approach allowed us to estimate the risk associated with these tasks performance. Moreover, the cork industry is related to high dust contamination and this can promote exposure to fungi since dust particles can act as carriers of fungi to the worker’s nose. Assessment by molecular tools will ensure the specific targeting of DNA from P. glabrum complex in workers nose

    Habitat and community structure modulate fish interactions in a neotropical clearwater river

    Species interactions can modulate the diversity and enhance the stability of biological communities in aquatic ecosystems. Despite previous efforts to describe fish interactions in tropical rivers, the role of habitat characteristics, community structure, and trophic traits over these interactions is still poorly understood. To investigate among-habitat variation in substratum feeding pressure and agonistic interactions between fishes, we used remote underwater videos in three habitats of a clearwater river in the Central Western, Brazil. We also performed visual surveys to estimate the abundance and biomass of fishes and proposed a trophic classification to understand how these variables can affect fish interactions. Community structure was the main factor affecting the variation in the interactions among the habitats. Biomass was the main variable determining which habitat a fish will feed on, while species abundance determined with how many other species it will interact in the agonistic interaction networks for each habitat. Specific habitats are not only occupied, but also used in distinct ways by the fish community. Overall, our results demonstrate the importance of the heterogeneity of habitats in tropical rivers for the interactions performed by the fishes and how the intensity of these interactions is affected by community structure
