    Design of Approaches for Dependability and Initial Prototypes

    The aim of CONNECT is to achieve universal interoperability between heterogeneous Networked Systems. For this, the non-functional properties required at each side of the connection going to be established must be fulfilled. By the one inclusive term "CONNECTability" we comprehend properties belonging to all four non-functional concerns of interest for CONNECT, namely dependability, performance, security and trust. We model such properties in conformance with a meta-model which establishes the relevant concepts and their relations. Then, building on the conceptual models proposed in the first year in Deliverable D5.1, in this document we present the approaches developed for assuring CONNECTability both at synthesis time and at runtime. The contributions include: the Dependability&Performance analysis Enabler, for which we release a modular architecture supporting stochastic verification and state-based analysis; incremental verification and event-based monitoring for runtime analysis; a model-based approach to interoperable trust management; the Security-by-Contract-with-Trust framework, which guarantees and enforces the expected trust levels and security policies

    EuReCa ONE—27 Nations, ONE Europe, ONE Registry A prospective one month analysis of out-of-hospital cardiac arrest outcomes in 27 countries in Europe

    AbstractIntroductionThe aim of the EuReCa ONE study was to determine the incidence, process, and outcome for out of hospital cardiac arrest (OHCA) throughout Europe.MethodsThis was an international, prospective, multi-centre one-month study. Patients who suffered an OHCA during October 2014 who were attended and/or treated by an Emergency Medical Service (EMS) were eligible for inclusion in the study. Data were extracted from national, regional or local registries.ResultsData on 10,682 confirmed OHCAs from 248 regions in 27 countries, covering an estimated population of 174 million. In 7146 (66%) cases, CPR was started by a bystander or by the EMS. The incidence of CPR attempts ranged from 19.0 to 104.0 per 100,000 population per year. 1735 had ROSC on arrival at hospital (25.2%), Overall, 662/6414 (10.3%) in all cases with CPR attempted survived for at least 30 days or to hospital discharge.ConclusionThe results of EuReCa ONE highlight that OHCA is still a major public health problem accounting for a substantial number of deaths in Europe.EuReCa ONE very clearly demonstrates marked differences in the processes for data collection and reported outcomes following OHCA all over Europe. Using these data and analyses, different countries, regions, systems, and concepts can benchmark themselves and may learn from each other to further improve survival following one of our major health care events

    Interactions between detritivores and microfungi during the leaf detritus decomposition in a volcanic lake (Lake Vico - central Italy)

    The role of biota in the mass loss of Phragmites australis (Cav.) Trin. ex Steud was studied in the littoral belt of a central Italy volcanic lake. The research focussed on the feeding interactions between detritivores and decomposing fungi as drivers of the leaf litter decomposition. The litterbag technique was used to assess the leaf mass loss, the number of colonizing fungi and the patterns of leaf colonization by detritivores during 40 days of submersion in 16 sampling sites. Cores of bottom sediment were collected to estimate the organic content and ergosterol concentration as measure of fungal mass. The rate of leaf mass loss showed significant variability among the sampling sites and was non-linearly related to the quantity of organic depositions onto the lake bottom, peaking at about 40% of the dry matter. The rate was also positively correlated with the density of detritivore mass relative to the leaf unit mass, which increased with time. On the 20th day of litterbag immersion, when 40% of the initial leaf litter remained, we observed the best accordance between the two measures as well as the lowest difference in the detritivore mass density among sampling sites. In the absence of animals, the decomposition rate was positively related to the number of fungi on the decaying litter. The feeding activity of detritivores changed both the species richness and composition of the fungal community on the litter. The substrate reduction due to intense animal feeding appeared to limit the ability of fungi to regrow after grazing. As a result, an inverse relationship between the number of fungi and the decomposition rate was observed

    Si306, a pyrazolo[3,4-d]pyrimidine derivative recently identified as promising anticancer agent, has shown favorable in vitro and in vivo activity profile against neuroblastoma (NB) models by acting as a competitive inhibitor of c-Src tyrosine kinase. Nevertheless, Si306 antitumor activity is associated with sub-optimal aqueous solubility, which might hinder its further development. Drug delivery systems were here developed with the aim to overcome this limitation, obtaining suitable formulations for more efficacious in vivo use. Si306 was encapsulated in pegylated stealth liposomes, undecorated or decorated with a monoclonal antibody able to specifically recognize and bind to the disialoganglioside GD2 expressed by NB cells (LP[Si306] and GD2-LP[Si306], respectively). Both liposomes possessed excellent morphological and physio-chemical properties, maintained over a period of two weeks. Compared to LP[Si306], GD2-LP[Si306] showed in vitro specific cellular targeting and increased cytotoxic activity against NB cell lines. After intravenous injection in healthy mice, pharmacokinetic profiles showed increased plasma exposure of Si306 when delivered by both liposomal formulations, compared to that obtained when Si306 was administered as free form. In vivo tumor homing and cytotoxic effectiveness of both liposomal formulations were finally tested in an orthotopic animal model of NB. Si306 tumor uptake resulted significantly higher when encapsulated in GD2-LP, compared to Si306, either free or encapsulated into untargeted LP. This, in turn, led to a significant increase in survival of mice treated with GD2-LP[Si306]. These results demonstrate a promising antitumor efficacy of Si306 encapsulated into GD2-targeted liposomes, supporting further therapeutic developments in pre-clinical trials and in the clinic for NB
