Trust and Civic Engagement in the Italian COVID-19 Lockdown
The paper presents considerations on the possible consequences associated with the lockdown condition that ensued on the coronavirus pandemic. On the one hand, the infection brought about a notable intensification of primary relationships, in particular within the family and the domestic space in general, while, on the other hand and especially at the start, it generated strong symbolic integration, shown for example by events such as flashmobs and other forms of the grassroot involvement of individuals. Through empirical research data, we will attempt to understand to what extent the cultural climate prevalent in Italy through March and April influenced the attitudes of the Italians in terms of general interpersonal trust or trust in the institutions, as well as in terms of culture and civic engagement. For this purpose, the paper uses data taken from two empirical research on a sample of about 1,000 cases among the Italian population. It will be compared the evidence resulting from a first survey realized at the end of 2017 and the data collected in a second survey carried out on a sample of the same size (that presents a panel quota of 700 cases) ending on April 30 2020, i.e., at the end of the lockdown period. The variables to be analysed are relative to general interpersonal trust, trust in the institutions and civic engagement activities. Such items will function as dependent variables and will be analysed in a diachronic perspective comparing the two panel samples. Furthermore, they will be observed in relation to their distribution throughout Italy, to the age of the interviewees and to their civil status, in an attempt to discover any connection between family life and the attitude towards civil society, keeping in mind the dynamics triggered in everyday life by the pandemic

Literature about the consequences of economic crisis on people\u2019s well-being has highlighted some statistical connections. In particular, researchers agree on the fact that economic crises bring an increase in suicides, states of psychological depression, and abuse of alcohol along with them. This work deals with the issue of human impact of economic crisis, and, through a panel of data concerning Italian population, it searches for any possible connection with less critical but anyway significant phenomena, such as the general state of well-being and subjectively perceived health
Molecular mechanisms underlying inherited photoreceptor degeneration as targets for therapeutic intervention
Retinitis pigmentosa (RP) is a form of retinal degeneration characterized by primary degeneration of rod photoreceptors followed by a secondary cone loss that leads to vision impairment and finally blindness. This is a rare disease with mutations in several genes and high genetic heterogeneity. A challenging effort has been the characterization of the molecular mechanisms underlying photoreceptor cell death during the progression of the disease. Some of the cell death pathways have been identified and comprise stress events found in several neurodegenerative diseases such as oxidative stress, inflammation, calcium imbalance and endoplasmic reticulum stress. Other cell death mechanisms appear more relevant to photoreceptor cells, such as high levels of cGMP and metabolic changes. Here we review some of the cell death pathways characterized in the RP mutant retina and discuss preclinical studies of therapeutic approaches targeting the molecular outcomes that lead to photoreceptor cell demise
Rapid detection of copy number variations and point mutations in BRCA1/2 genes using a single workflow by ion semiconductor sequencing pipeline
Molecular analysis of BRCA1 (MIM# 604370) and BRCA2 (MIM #600185) genes is essential for familial breast and ovarian cancer prevention and treatment. An efficient, rapid, cost-effective accurate strategy for the detection of pathogenic variants is crucial. Mutations detection of BRCA1/2 genes includes screening for single nucleotide variants (SNVs), small insertions or deletions (indels), and Copy Number Variations (CNVs). Sanger sequencing is unable to identify CNVs and therefore Multiplex Ligation Probe amplification (MLPA) or Multiplex Amplicon Quantification (MAQ) is used to complete the BRCA1/2 genes analysis. The rapid evolution of Next Generation Sequencing (NGS) technologies allows the search for point mutations and CNVs with a single platform and workflow. In this study we test the possibilities of NGS technology to simultaneously detect point mutations and CNVs in BRCA1/2 genes, using the OncomineTM BRCA Research Assay on Personal Genome Machine (PGM) Platform with Ion Reporter Software for sequencing data analysis (Thermo Fisher Scientific). Comparison between the NGS-CNVs, MLPA and MAQ results shows how the NGS approach is the most complete and fast method for the simultaneous detection of all BRCA mutations, avoiding the usual time consuming multistep approach in the routine diagnostic testing of hereditary breast and ovarian cancers
Personal Networks as Social Capital: a Research Strategy to Measure Contents and Forms of Social Support
The basic idea of social capital consists of the belief that individuals\u2019 lives can be qualitatively improved by social relationships, or rather by the social resources that these relationships manage to mobilize. So, personal networks can provide a kind of capital for individuals. This essay examines the characteristics of personal networks that mobilize social resources in a sample of 307 individuals, representative of the population of Verona (Italy). By using some structural indicators of social capital, the authors describe the contents and the forms of different kinds of social circles (family, work colleagues, members in third sector organizations, friends, neighbours). This study rejects a hypothesis according to which stronger ties are better vehicles for symbolic and expressive resources, and confirms a hypothesis on the similarity of the forms of different social circles that provide individuals with social support

Dementia-related genetic variants in an Italian population of early-onset Alzheimer’s disease
Early-onset Alzheimer’s disease (EOAD) is the most common form of early-onset dementia. Although three major genes have been identified as causative, the genetic contribution to the disease remains unsolved in many patients. Recent studies have identified pathogenic variants in genes representing a risk factor for developing Alzheimer’s disease (AD) and in causative genes for other degenerative dementias as responsible for EOAD. To study them further, we investigated a panel of candidate genes in 102 Italian EOAD patients, 45.10% of whom had a positive family history and 21.74% with a strong family history of dementia. We found that 10.78% of patients carried pathogenic or likely pathogenic variants, including a novel variant, in PSEN1, PSEN2, or APP, and 7.84% showed homozygosity for the ε4 APOE allele. Additionally, 7.84% of patients had a moderate risk allele in PSEN1, PSEN2, or TREM2 genes. Besides, we observed that 12.75% of our patients carried only a variant in genes associated with other neurodegenerative diseases. The combination of these variants contributes to explain 46% of cases with a definite familiarity and 32% of sporadic forms. Our results confirm the importance of extensive genetic screening in EOAD for clinical purposes, to select patients for future treatments and to contribute to the definition of overlapping pathogenic mechanisms between AD and other forms of dementia
Diagnostic value of plasma p-tau181, NfL, and GFAP in a clinical setting cohort of prevalent neurodegenerative dementias
Background: Increasing evidence supports the use of plasma biomarkers of neurodegeneration and neuroinflammation to screen and diagnose patients with dementia. However, confirmatory studies are required to demonstrate their usefulness in the clinical setting. Methods: We evaluated plasma and cerebrospinal fluid (CSF) samples from consecutive patients with frontotemporal dementia (FTD) (n = 59), progressive supranuclear palsy (PSP) (n = 31), corticobasal syndrome (CBS) (n = 29), dementia with Lewy bodies (DLB) (n = 49), Alzheimer disease (AD) (n = 97), and suspected non-AD physiopathology (n = 51), as well as plasma samples from 60 healthy controls (HC). We measured neurofilament light chain (NfL), phospho-tau181 (p-tau181), and glial fibrillary acid protein (GFAP) using Simoa (all plasma biomarkers and CSF GFAP), CLEIA (CSF p-tau181), and ELISA (CSF NfL) assays. Additionally, we stratified patients according to the A/T/N classification scheme and the CSF α-synuclein real-time quaking-induced conversion assay (RT-QuIC) results. Results: We found good correlations between CSF and plasma biomarkers for NfL (rho = 0.668, p < 0.001) and p-tau181 (rho = 0.619, p < 0.001). Plasma NfL was significantly higher in disease groups than in HC and showed a greater increase in FTD than in AD [44.9 (28.1–68.6) vs. 21.9 (17.0–27.9) pg/ml, p < 0.001]. Conversely, plasma p-tau181 and GFAP levels were significantly higher in AD than in FTD [3.2 (2.4–4.3) vs. 1.1 (0.7–1.6) pg/ml, p < 0.001; 404.7 (279.7–503.0) vs. 198.2 (143.9–316.8) pg/ml, p < 0.001]. GFAP also allowed discriminating disease groups from HC. In the distinction between FTD and AD, plasma p-tau181 showed better accuracy (AUC 0.964) than NfL (AUC 0.791) and GFAP (AUC 0.818). In DLB and CBS, CSF amyloid positive (A+) subjects had higher plasma p-tau181 and GFAP levels than A− individuals. CSF RT-QuIC showed positive α-synuclein seeding activity in 96% DLB and 15% AD patients with no differences in plasma biomarker levels in those stratified by RT-QuIC result. Conclusions: In a single-center clinical cohort, we confirm the high diagnostic value of plasma p-tau181 for distinguishing FTD from AD and plasma NfL for discriminating degenerative dementias from HC. Plasma GFAP alone differentiates AD from FTD and neurodegenerative dementias from HC but with lower accuracy than p-tau181 and NfL. In CBS and DLB, plasma p-tau181 and GFAP levels are significantly influenced by beta-amyloid pathology
