    A calorimeter coupled with a magnetic spectrometer for the detection of primary cosmic antiprotons

    A tracking calorimeter made of 3200 brass streamer tubes together with 3200 pick-up strips has been built to complement a magnetic spectrometer in order to detect cosmic antiprotons in space. The characteristics of such a calorimeter, the results of a preliminary test of a prototype as well as the properties of the whole apparatus are presented. The apparatus, designed to operate on a balloon at an altitude of about 40 km, can be considered as a second generation detector, capable in principle to solve the problem of the presence of low energy (≀1 Ge V/c) antiprotons in the cosmic rays which is still open because of the disagreement between the existent experimental data

    Redistribution of DAT/α-synuclein complexes visualized by “in situ” proximity ligation assay in transgenic mice modelling early Parkinson’s disease

    Alpha-synuclein, the major component of Lewy bodies, is thought to play a central role in the onset of synaptic dysfunctions in Parkinson's disease (PD). In particular, α-synuclein may affect dopaminergic neuron function as it interacts with a key protein modulating dopamine (DA) content at the synapse: the DA transporter (DAT). Indeed, recent evidence from our "in vitro" studies showed that α-synuclein aggregation decreases the expression and membrane trafficking of the DAT as the DAT is retained into α-synuclein-immunopositive inclusions. This notwithstanding, "in vivo" studies on PD animal models investigating whether DAT distribution is altered by the pathological overexpression and aggregation of α-synuclein are missing. By using the proximity ligation assay, a technique which allows the "in situ" visualization of protein-protein interactions, we studied the occurrence of alterations in the distribution of DAT/α-synuclein complexes in the SYN120 transgenic mouse model, showing insoluble α-synuclein aggregates into dopaminergic neurons of the nigrostriatal system, reduced striatal DA levels and an altered distribution of synaptic proteins in the striatum. We found that DAT/α-synuclein complexes were markedly redistributed in the striatum and substantia nigra of SYN120 mice. These alterations were accompanied by a significant increase of DAT striatal levels in transgenic animals when compared to wild type littermates. Our data indicate that, in the early pathogenesis of PD, α-synuclein acts as a fine modulator of the dopaminergic synapse by regulating the subcellular distribution of key proteins such as the DAT

    Spatial Resolution of Double-Sided Silicon Microstrip Detectors for the PAMELA Apparatus

    The PAMELA apparatus has been assembled and it is ready to be launched in a satellite mission to study mainly the antiparticle component of cosmic rays. In this paper the performances obtained for the silicon microstrip detectors used in the magnetic spectrometer are presented. This subdetector reconstructs the curvature of a charged particle in the magnetic field produced by a permanent magnet and consequently determines momentum and charge sign, thanks to a very good accuracy in the position measurements (better than 3 um in the bending coordinate). A complete simulation of the silicon microstrip detectors has been developed in order to investigate in great detail the sensor's characteristics. Simulated events have been then compared with data gathered from minimum ionizing particle (MIP) beams during the last years in order to tune free parameters of the simulation. Finally some either widely used or original position finding algorithms, designed for such kind of detectors, have been applied to events with different incidence angles. As a result of the analysis, a method of impact point reconstruction can be chosen, depending on both the particle's incidence angle and the cluster multiplicity, so as to maximize the capability of the spectrometer in antiparticle tagging.Comment: 28 pages, 18 figures, submitted to Nuclear Instruments and Methods in Physics Research

    Study of the granularity for a tracking calorimeter with optimal rejection of proton background in positron detection

    In this paper we present a Monte Carlo study of a calorimeter response for an experiment to equip the magnetic facility of the USA space station. Main purpose in the design of such a calorimeter is the efficient discrimination between eloctromagnetic and hadronic showers. The estimated rejection power results to be better than 1·10−3 p/e+ for incident particles with energy between 10 GeV and 100GeV

    The Cosmic-Ray Proton and Helium Spectra measured with the CAPRICE98 balloon experiment

    A new measurement of the primary cosmic-ray proton and helium fluxes from 3 to 350 GeV was carried out by the balloon-borne CAPRICE experiment in 1998. This experimental setup combines different detector techniques and has excellent particle discrimination capabilities allowing clear particle identification. Our experiment has the capability to determine accurately detector selection efficiencies and systematic errors associated with them. Furthermore, it can check for the first time the energy determined by the magnet spectrometer by using the Cherenkov angle measured by the RICH detector well above 20 GeV/n. The analysis of the primary proton and helium components is described here and the results are compared with other recent measurements using other magnet spectrometers. The observed energy spectra at the top of the atmosphere can be represented by (1.27+-0.09)x10^4 E^(-2.75+-0.02) particles (m^2 GeV sr s)^-1, where E is the kinetic energy, for protons between 20 and 350 GeV and (4.8+-0.8)x10^2 E^(-2.67+-0.06) particles (m^2 GeV nucleon^-1 sr s)^-1, where E is the kinetic energy per nucleon, for helium nuclei between 15 and 150 GeV nucleon^-1.Comment: To be published on Astroparticle Physics (44 pages, 13 figures, 5 tables

    Two of the most compelling issues facing astrophysics and cosmology today are to understand the nature of the dark matter that pervades the universe and to understand the apparent absence of cosmological antimatter. For both issues, sensitive measurements of cosmic-ray antiprotons and positrons, in a wide energy range, are crucial. Many different mechanisms can contribute to antiprotons and positrons production, ranging from conventional reactions up to exotic processes like neutralino annihilation. The open problems are so fundamental (i.e.: is the universe symmetric in matter and antimatter ?) that experiments in this field will probably be of the greatest interest in the next years.     Human Stem Cell-Derived Neurons: A System to Study Human Tau Function and Dysfunction

    Background: Intracellular filamentous deposits containing microtubule-associated protein tau constitute a defining characteristic of many neurodegenerative disorders. Current experimental models to study tau pathology in vitro do not usually recapitulate the tau expression pattern characteristic of adult human brain. In this study, we have investigated whether human embryonic stem cell-derived neurons could be a good model to study human tau distribution, function and dysfunction. Methodology/Principal Findings: Using RT-PCR, immunohistochemistry, western blotting and cell transfections we have investigated whether all 6 adult human brain tau isoforms are expressed in neurons derived from human embryonic and fetal stem cells and whether 4 repeat tau over-expression alone, or with the F3 tau repeat fragment, (amino acid 258–380 of the 2N4R tau isoform with the DK280 mutation) affects tau distribution. We found that the shortest 3 repeat tau isoform, similarly to human brain, is the first to be expressed during neuronal differentiation while the other 5 tau isoforms are expressed later. Over expression of tau with 4 repeats affects tau cellular distribution and the short tau F3 fragment appears to increase tau phosphorylation but this effect does not appear to be toxic for the cell.     PAMELA is a composite particle detector which will be launched during the first half of 2006 on board the Russian satellite Resurs DK-1 from Baikonur cosmodrome in Kazakhstan. This experiment is mainly conceived for the study of cosmic-ray antiparticles and for the search for light antinuclei, but other issues related to the cosmic-ray physics will be investigated.     Dark Matter Search Perspectives with GAMMA-400

    GAMMA-400 is a future high-energy gamma-ray telescope, designed to measure the fluxes of gamma-rays and cosmic-ray electrons + positrons, which can be produced by annihilation or decay of dark matter particles, and to survey the celestial sphere in order to study point and extended sources of gamma-rays, measure energy spectra of Galactic and extragalactic diffuse gamma-ray emission, gamma-ray bursts, and gamma-ray emission from the Sun. GAMMA-400 covers the energy range from 100 MeV to ~3000 GeV. Its angular resolution is ~0.01 deg(Eg > 100 GeV), and the energy resolution ~1% (Eg > 10 GeV). GAMMA-400 is planned to be launched on the Russian space platform Navigator in 2019.
