Label-free tissue proteomics can classify oral squamous cell carcinoma from healthy tissue in a stage-specific manner

Objectives: No prognostic or predictive biomarkers for oral squamous cell carcinoma (OSCC) exist. We aimed to discover novel proteins, altered in OSCC, to be further investigated as potential biomarkers, and to improve understanding about pathways involved in OSCC. Materials and Methods: Proteomic signatures of seven paired healthy and OSCC tissue samples were identified using ultra-definition quantitative mass spectrometry, then analysed and compared using Anova, principal component analysis, hierarchical clustering and OPLS-DA modelling. A selection of significant proteins that were also altered in the serum from a previous study (PMID: 28632724) were validated immunohistochemically on an independent cohort (n = 66) to confirm immunopositivity and location within tumour tissue. Ingenuity Pathways Analysis was employed to identify altered pathways. Results: Of 829 proteins quantified, 257 were significant and 72 were able to classify healthy vs OSCC using OPLS-DA modelling. We identified 19 proteins not previously known to be upregulated in OSCC, including prosaposin and alpha-taxilin. KIAA1217 and NDRG1 were upregulated in stage IVa compared with stage I tumours. Altered pathways included calcium signalling, cellular movement, haematological system development and function, and immune cell trafficking, and involved NF-kappa B and MAPK networks. Conclusions: We found a set of proteins reliably separating OSCC tumour from healthy tissue, and multiple proteins differing between stage I and stage IVa OSCC. These potential biomarkers can be studied and validated in larger cohorts.Peer reviewe

Although several concepts for implementing education exaggerated using blockchain technology, t agreements which are allocated ledgers carefully and thoroughly. Blockchain technology, unlike databases that are processed by certain forces that provide policies in correctness of data through real-time implementation, Blockchain is a digital data storage system containing records that are connected via cryptography. Blockchain technology has now been used by various sectors, one of which is for transactions in cryptocurrencies such as Bitcoin. The benefit of the blockchain technology approach in education is that people who do not have knowledge of blockchain technology can benefit from blockchain technology, because a lot of the data generated by this sector is continuous from education to Bachelor. Blockchain, can be used as, "a machine to create high-level security and reliable security", the high level of security and efficiency of blockchain as a storage place for financial transactions and data has opened up opportunities for innovation in education as a storage for educators and students' data

Psychological interventions in preventing chronicity of sub-acute back pain : a systematic review

    Get PDF
Objectives: There is scarce information on preventive psychological and behavioural methods applicable to sub-acute (4-12 weeks) back pain, a precursor to chronic back pain. We conducted a systematic literature review of the efficacy of psychological interventions in preventing chronicity of sub-acute back pain. Methods: A systematic literature search in CINAHL, CENTRAL, MEDLINE, PubMed(R), PsychINFO, Scopus and Web of Science databases. Results: From a total of 271 records, only three studies met the eligibility criteria. In two of the reviewed studies, the interventions had an insignificant preventive impact on the chronification of back pain. In one study the CBT intervention proved promising in preventing back pain related disability. None of the studies reported a significant impact on pain intensity at follow-up. Conclusions: The psychological interventions did not impact pain outcomes. There is a disproportion between novel knowledge on psychological factors involved in the transition to chronic pain and corresponding preventive treatments. Additional studies on psychological interventions on sub-acute back pain prevention are highly warranted due to the enormous burden that back pain creates when it becomes chronic. The research project has the ethical approval of the Research Ethical Committee at Helsinki University Hospital, HUS/2435/2017.Peer reviewe
