    Smooth tensionful higher-codimensional brane worlds with bulk and brane form fields

    Completely regular tensionful codimension-n brane world solutions are discussed, where the core of the brane is chosen to be a thin codimension-(n-1) shell in an infinite volume flat bulk, and an Einstein-Hilbert term localized on the brane is included (Dvali-Gabadadze-Porrati models). In order to support such localized sources we enrich the vacuum structure of the brane by the inclusion of localized form fields. We find that phenomenological constraints on the size of the internal core seem to impose an upper bound to the brane tension.     The problematic backreaction of SUSY-breaking branes

    In this paper we investigate the localisation of SUSY-breaking branes which, in the smeared approximation, support specific non-BPS vacua. We show, for a wide class of boundary conditions, that there is no flux vacuum when the branes are described by a genuine delta-function. Even more, we find that the smeared solution is the unique solution with a regular brane profile. Our setup consists of a non-BPS AdS_7 solution in massive IIA supergravity with smeared anti-D6-branes and fluxes T-dual to ISD fluxes in IIB supergravity.Comment: 27 pages, Latex2e, 5 figure

    Fractional branes, warped compactifications and backreacted orientifold planes

    The standard extremal p-brane solutions in supergravity are known to allow for a generalisation which consists of adding a linear dependence on the world-volume coordinates to the usual harmonic function. In this note we demonstrate that remarkably this generalisation goes through in exactly the same way for p-branes with fluxes added to it that correspond to fractional p-branes. We relate this to warped orientifold compactifications by trading the Dp-branes for Op-planes that solve the RR tadpole condition. This allows us to interpret the worldvolume dependence as due to lower-dimensional scalars that flow along the massless directions in the no-scale potential. Depending on the details of the fluxes these flows can be supersymmetric domain wall flows.     FAS2FURIOUS: Moderate-Throughput Secreted Expression of Difficult Recombinant Proteins in <em>Drosophila</em> S2 Cells

Recombinant protein expression in eukaryotic insect cells is a powerful approach for producing challenging targets. However, due to incompatibility with standard baculoviral platforms and existing low-throughput methodology, the use of the Drosophila melanogaster “S2” cell line lags behind more common insect cell lines such as Sf9 or High-Five™. Due to the advantages of S2 cells, particularly for secreted and secretable proteins, the lack of a simple and parallelizable S2-based platform represents a bottleneck, particularly for biochemical and biophysical laboratories. Therefore, we developed FAS2FURIOUS, a simple and rapid S2 expression pipeline built upon an existing low-throughput commercial platform. FAS2FURIOUS is comparable in effort to simple E. coli systems and allows users to clone and test up to 46 constructs in just 2 weeks. Given the ability of S2 cells to express challenging targets, including receptor ectodomains, secreted glycoproteins, and viral antigens, FAS2FURIOUS represents an attractive orthogonal approach for protein expression in eukaryotic cells

    Effects of a Single Intra-Articular Injection of a Microsphere-Formulation of Triamcinolone Acetonide on Knee Osteoarthritis Pain: A Double-Blinded, Randomized, Placebo-Controlled, Multinational Study

    Background: Intra-articular corticosteroids relieve osteoarthritis pain, but rapid systemic absorption limits efficacy. FX006, a novel, microsphere-based, extended-release triamcinolone acetonide (TA) formulation, prolongs TA joint residence and reduces systemic exposure compared with standard TA crystalline suspension (TAcs). We assessed symptomatic benefits and safety of FX006 compared with saline-solution placebo and TAcs. Methods: In this Phase-3, multicenter, double-blinded, 24-week study, adults ≥40 years of age with knee osteoarthritis (Kellgren-Lawrence grade 2 or 3) and average-daily-pain (ADP)-intensity scores of ≥5 and ≤9 (0 to 10 numeric rating scale) were centrally randomized (1:1:1) to a single intra-articular injection of FX006 (32 mg), saline-solution placebo, or TAcs (40 mg). The primary end point was change from baseline to week 12 in weekly mean ADP-intensity scores for FX006 compared with saline-solution placebo. Secondary end points were area-under-effect (AUE) curves of the change in weekly mean ADP-intensity scores from baseline to week 12 for FX006 compared with saline-solution placebo, AUE curves of the change in weekly mean ADP-intensity scores from baseline to week 12 for FX006 compared with TAcs, change in weekly mean ADP-intensity scores from baseline to week 12 for FX006 compared with TAcs, and AUE curves of the change in weekly mean ADP-intensity scores from baseline to week 24 for FX006 compared with saline-solution placebo. Exploratory end points included week-12 changes in Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC) and Knee Injury and Osteoarthritis Outcome Score Quality of Life (KOOS-QOL) subscale scores for FX006 compared with saline-solution placebo and TAcs. Adverse events were elicited at each inpatient visit. Results: The primary end point was met. Among 484 treated patients (n = 161 for FX006, n = 162 for saline-solution placebo, and n = 161 for TAcs), FX006 provided significant week-12 improvement in ADP intensity compared with that observed for saline-solution placebo (least-squares mean change from baseline: −3.12 versus −2.14; p < 0.0001) indicating ∼50% improvement. FX006 afforded improvements over saline-solution placebo for all secondary and exploratory end points (p < 0.05). Improvements in osteoarthritis pain were not significant for FX006 compared with TAcs using the ADP-based secondary measures. Exploratory analyses of WOMAC-A, B, and C and KOOS-QOL subscales favored FX006 (p ≤ 0.05). Adverse events were generally mild, occurring at similar frequencies across treatments. Conclusions: FX006 provided significant, clinically meaningful pain reduction compared with saline-solution placebo at week 12 (primary end point)

    EFT beyond the horizon: stochastic inflation and how primordial quantum fluctuations go classical

    We identify the effective theory describing inflationary super-Hubble scales and show it to be a special case of effective field theories appropriate to open systems. Open systems allow information to be exchanged between the degrees of freedom of interest and those that are integrated out, such as for particles moving through a fluid. Strictly speaking they cannot in general be described by an effective lagrangian; rather the appropriate `low-energy' limit is instead a Lindblad equation describing the evolution of the density matrix of the slow degrees of freedom. We derive the equation relevant to super-Hubble modes of quantum fields in near-de Sitter spacetimes and derive two implications. We show the evolution of the diagonal density-matrix elements quickly approaches the Fokker-Planck equation of Starobinsky's stochastic inflationary picture. This provides an alternative first-principles derivation of this picture's stochastic noise and drift, as well as its leading corrections. (An application computes the noise for systems with a sub-luminal sound speed.) We argue that the presence of interactions drives the off-diagonal density-matrix elements to zero in the field basis. This shows why the field basis is the `pointer basis' for the decoherence of primordial quantum fluctuations while they are outside the horizon, thus allowing them to re-enter as classical fluctuations, as assumed when analyzing CMB data. The decoherence process is efficient, occurring after several Hubble times even for interactions as weak as gravitational-strength. Crucially, the details of the interactions largely control only the decoherence time and not the nature of the final late-time stochastic state, much as interactions can control the equilibration time for thermal systems but are largely irrelevant to the properties of the resulting equilibrium state

    Kahler Moduli Inflation Revisited

    We perform a detailed numerical analysis of inflationary solutions in Kahler moduli of type IIB flux compactifications. We show that there are inflationary solutions even when all the fields play an important role in the overall shape of the scalar potential. Moreover, there exists a direction of attraction for the inflationary trajectories that correspond to the constant volume direction. This basin of attraction enables the system to have an island of stability in the set of initial conditions. We provide explicit examples of these trajectories, compute the corresponding tilt of the density perturbations power spectrum and show that they provide a robust prediction of n_s approximately 0.96 for 60 e-folds of inflation.Comment: 27 pages, 9 figure

    Effects of heavy modes on vacuum stability in supersymmetric theories

    We study the effects induced by heavy fields on the masses of light fields in supersymmetric theories, under the assumption that the heavy mass scale is much higher than the supersymmetry breaking scale. We show that the square-masses of light scalar fields can get two different types of significant corrections when a heavy multiplet is integrated out. The first is an indirect level-repulsion effect, which may arise from heavy chiral multiplets and is always negative. The second is a direct coupling contribution, which may arise from heavy vector multiplets and can have any sign. We then apply these results to the sGoldstino mass and study the implications for the vacuum metastability condition. We find that the correction from heavy chiral multiplets is always negative and tends to compromise vacuum metastability, whereas the contribution from heavy vector multiplets is always positive and tends on the contrary to reinforce it. These two effects are controlled respectively by Yukawa couplings and gauge charges, which mix one heavy and two light fields respectively in the superpotential and the Kahler potential.     On Supersymmetric D7-branes in the Warped Deformed Conifold

    We study the supersymmetric properties of D7-branes in the warped deformed conifold. We consider the κ\kappa-symmetry conditions on D7-branes in this specific warped background, taking into account the background NS-NS 2-form flux. While any holomorphic embedding defines a supersymmetric D7-brane in the absence of background H-flux, most of the D7-brane embeddings considered in the literature do not preserve supersymmetry for the warped deformed conifold without also including brane worldvolume flux. For the simplest such embedding, we construct numerically the worldvolume fluxnecessary to restore supersymmetry. We also comment on the dual field theory descriptions in terms of cascading N=1 supersymmetric gauge theories with flavors.
