Modulation of macrophage cytokine profiles during solid tumor progression: susceptibility to Candida albicans infection
<p>Abstract</p> <p>Background</p> <p>In order to attain a better understanding of the interactions between opportunist fungi and their hosts, we investigated the cytokine profile associated with the inflammatory response to <it>Candida albicans </it>infection in mice with solid Ehrlich tumors of different degrees.</p> <p>Methods</p> <p>Groups of eight animals were inoculated intraperitoneally with 5 × 10<sup>6 </sup><it>C. albicans </it>7, 14 or 21 days after tumor implantation. After 24 or 72 hours, the animals were euthanized and intraperitoneal lavage fluid was collected. Peritoneal macrophages were cultivated and the levels of IFN-γ, TNF-α, IL-12, IL-10 and IL-4 released into the supernatants were measured by ELISA. Kidney, liver and spleen samples were evaluated for fungal dissemination. Tumor-free animals and animals that had only been subjected to <it>C. albicans </it>infection were used as control groups.</p> <p>Results</p> <p>Our results demonstrated that the mice produced more IFN-γ and TNF-α and less IL-10, and also exhibited fungal clearance, at the beginning of tumor evolution. With the tumor progression, this picture changed: IL-10 production increased and IFN-γ and TNF-α release decreased; furthermore, there was extensive fungal dissemination.</p> <p>Conclusion</p> <p>Our results indicate that solid tumors can affect the production of macrophage cytokines and, in consequence, affect host resistance to opportunistic infections.</p
THE RELEVANCE OF NUTRITIONAL STATUS AND HISTOPATHOLOGICAL FINDINGS ON THE INFECTIOUS PROCESS OF BALB/C MICE INOCULATED WITH Lacazia loboi
SUMMARY The aim of this study was to evaluate the effects of the protein-calorie malnutrition in BALB/c isogenic mice infected with Lacazia loboi, employing nutritional and histopathological parameters. Four groups were composed: G1: inoculated with restricted diet, G2: not inoculated with restricted diet, G3: inoculated with regular diet, G4: not inoculated with regular diet. Once malnutrition had been imposed, the animals were inoculated intradermally in the footpad and after four months, were sacrificed for the excision of the footpad, liver and spleen. The infection did not exert great influence on the body weight of the mice. The weight of the liver and spleen showed reduction in the undernourished groups when compared to the nourished groups. The macroscopic lesions, viability index and total number of fungi found in the footpads of the infected mice were increased in G3 when compared to G1. Regarding the histopathological analysis of the footpad, a global cellularity increase in the composition of the granuloma was observed in G3 when compared to G1, with large numbers of macrophages and multinucleated giant cells, discrete numbers of lymphocytes were present in G3 and an increase was observed in G1. The results suggest that there is considerable interaction between Jorge Lobo's disease and nutrition
Tipos de fibras musculares, identificação, caracterÃsticas e qualidade da carne / Types of muscle fibers, identification, characteristics and quality of meat
A musculatura esquelética é constituÃda por fibras musculares que possuem caracterÃsticas fisiológicas e bioquÃmicas distintas. As fibras são o principal componente do músculo gerado a partir de estruturas derivadas do mesoderma. O número de células e o desenvolvimento de fibras musculares esqueléticas estão relacionados à produção de animais.  Basicamente elas podem ser classificadas em fibra do Tipo I e fibras do Tipo II. As fibras Tipo I e IIb, também conhecidas como oxidativa lenta e glicolÃtica rápida, respectivamente, representam dois perfis metabólicos extremos. As fibras do Tipo IIa e IIx são fibras oxidativas-glicolÃticas rápidas intermediárias. O objetivo da pesquisa é realizar uma revisão bibliográfica sobre os tipos de fibras musculares, identificação e caracterÃsticas para melhor entender a sua relação com a qualidade da carne. As fibras musculares podem ser incluÃdas na seleção genética para caracterÃsticas de crescimento e produção devido à alta variabilidade genética e herdabilidade. Os consumidores estão cada vez mais exigentes em relação à qualidade da carne, principalmente no diz respeito a atributos sensoriais. A qualidade da carne é determinada pela marmorização, cor, textura e gordura, e esses atributos de qualidade são influenciados por caracterÃsticas do músculo esquelético, e estão diretamente relacionadas à s propriedades das fibras musculares.
In vitro and in situ activation of the complement system by the fungus Lacazia loboi
Since there are no studies evaluating the participation of the complement system (CS) in Jorge Lobo's disease and its activity on the fungus Lacazia loboi, we carried out the present investigation. Fungal cells with a viability index of 48% were obtained from the footpads of BALB/c mice and incubated with a pool of inactivated serum from patients with the mycosis or with sterile saline for 30 min at 37 ºC. Next, the tubes were incubated for 2 h with a pool of noninactivated AB+ serum, inactivated serum, serum diluted in EGTA-MgCl2, and serum diluted in EDTA. The viability of L. loboi was evaluated and the fungal suspension was cytocentrifuged. The slides were submitted to immunofluorescence staining using human anti-C3 antibody. The results revealed that 98% of the fungi activated the CS by the alternative pathway and no significant difference in L. loboi viability was observed after CS activation. In parallel, frozen histological sections from 11 patients were analyzed regarding the presence of C3 and IgG by immunofluorescence staining. C3 and IgG deposits were observed in the fungal wall of 100% and 91% of the lesions evaluated, respectively. The results suggest that the CS and immunoglobulins may contribute to the defense mechanisms of the host against L. loboi
