    Self-assembly of proteins into a three-dimensional multilayer system: investigation of the surface of the human fungal pathogen Aspergillus fumigatus.

    Hydrophobins are small surface active proteins that fulfil a wide spectrum of functions in fungal growth and development. The human fungal pathogen Aspergillus fumigatus expresses RodA hydrophobins that self-assemble on the outer conidial surface into tightly organized nanorods known as rodlets. AFM investigation of the conidial surface allows us to evidence that RodA hydrophobins self-assemble into rodlets through bilayers. Within bilayers, hydrophilic domains of hydrophobins point inward, thus making a hydrophilic core, while hydrophobic domains point outward. AFM measurements reveal that several rodlet bilayers are present on the conidial surface thus showing that proteins self-assemble into a complex three-dimensional multilayer system. The self-assembly of RodA hydrophobins into rodlets results from attractive interactions between stacked ÎČ-sheets, which conduct to a final linear cross-ÎČ spine structure. A Monte Carlo simulation shows that anisotropic interactions are the main driving forces leading the hydrophobins to self-assemble into parallel rodlets, which are further structured in nanodomains. Taken together, these findings allow us to propose a mechanism, which conducts RodA hydrophobins to a highly ordered rodlet structure. The mechanism of hydrophobin assembly into rodlets offers new prospects for the development of more efficient strategies leading to disruption of rodlet formation allowing a rapid detection of the fungus by the immune system

    Penelitian ini bertujuan untuk mengetahui Pengaruh Kualitas Sistem Informasi (BSGTouch) Terhadap Kepuasan Pengguna. Metode penelitian yang digunakan dalam penelitian ini adalah metode kuantitatif dengan pendekatan kuantitatif deskriptif. Data yang digunakanadalah data primer yang diperoleh dari penyebaran kuesioner kepada nasabah Bank SulutGo Cabang Boroko. Penarikan sampel dalam penelitian menggunakan rumus Lameshow dikarenakan jumlah populasi tidak diketahui banyaknya dan jumlah responden berjumlah 96. Analisis data dalam penelitian ini menggunakan analisis regresi linier sederhana dengan bantuan program SPSS 21. Hasil penelitian menunjukan kualitas system informasi berpengaruh positif signifikan terhadap kepuasan pengguna. Koefisien determinasi dalam penelitian ini sebesar 68,4% yang berarti variabilitas kepuasan pengguna dapat dijelaskan oleh kualitas system informasi sebesar 68,4%

    Penelitian ini bertujuan untuk mengetahui 1) seberapa besar pengaruh system informasi manajemen terhadap kinerja pegawai. 2) seberapa besar pengaruh lingkungan kerja terhadap kinerja pegawai. 3) seberapa besar pengaruh system informasi manajemen dan lingkungan kerja terhadap kinerja pegawai. Populasi dalam penelitian ini sebanyak 138 pegawai. Tehnik pengambilan sampel menggunakan purposive sampling yaitu sebanyak 58 responden. Uji analisis regresi linear berganda. Berdasarkan hasil penelitian uji t (parsial) menunjukkan bahwa variabel 1) system informasi manajemen berpengaruh positif terhadap kinerja pegawai ditunjukkan dengan nilai t-hitung 3,547 t-tabel 2,0040. 2) lingkungan kerja berpengaruh positif terhadap kinerja pegawai ditunjukkan dengan nilai t-hitung 2,436 t-tabel 2,0040. 3) system informasi manajemen dan lingkungan kerja secara simultan mempengaruhi kinerja pegawai ditunjukkan dengan nilai Fhitung 8,999 Ftabel 3,1649 dengan taraf signifikansi 0,05. Berdasarkan perhitungan koefisien determinasi menunjukkan R2 sebesar 24,7% dikategorikan rendah, dapat disimpulkan bahwa system informasi manajemen dan lingkungan kerja secara simultan berpengaruh terhadap kinerja pegawai sedangkan 75,3% dipengaruhi oleh variabel lain yang tidak diteliti dalam penelitian ini seperti motivasi, dan pengambilan keputusan

    Evaluation of Antibiotic Use Among Sepsis Patients in an Intensive Care Unit: A cross-sectional study at a referral hospital in Indonesia

    Objectives: This study aimed to evaluate the appropriateness of antibiotic use and factors associated with outcomes among sepsis patients in an ntensive care unit (ICU). Methods: This cross-sectional study was carried out from February to May 2017 and included all adult patients with sepsis or septic shock admitted to the ICU of Dharmais Cancer Hospital, Jakarta, Indonesia. Data were collected from the patients’ medical records. Results: A total of 60 patients with sepsis or septic shock were admitted to the ICU during the study period. The most common source of infection was hospital-acquired pneumonia (61.7%) and the majority had two or more comorbidities (93.3%). There were 115 antibiotic regimens prescribed. Overall, 33.3% of patients were prescribed inappropriate types of antibiotics and 51.7% were given an inappropriate dosage. The mortality rate was 68.3%. There was a statistically significant association between patient outcome and inappropriate doses of antibiotics (P = 0.034), although not inappropriate types of antibiotics (P = 0.050). A multivariate analysis indicated that the main factors influencing patient outcome were septic shock and the presence of at least two comorbidities (P <0.050 each). Conclusion: Inappropriate doses of antibiotics, a diagnosis of septic shock and the presence of at least two comorbidities were found to significantly increase the mortality rate of sepsis patients admitted to an ICU in Indonesia.     Fish is an important source of animal protein in Africa and is used in the composition of several national dishes. This literature review examines fishing, aquaculture and fish reared in the whÚdos of the upper Ouémé delta in Benin. It addresses socio-economic aspects, nutritional importance and traditional fish preservation technology. The sources of information used are publications, books, reports of dissertations and theses and technical notes. It was found that Clarias gariepinus and Oreochromis niloticus, among others, are two species of fish most often raised in the whÚdos of the Ouémé River in Benin.&nbsp; These fish adapt to large variations of ecological factors of the aquatic environment found everywhere in Africa and reproduce easily in captivity, which favors their breeding in ponds. These species present multiple fish farming qualities: resistance to high densities, resistance to diseases and low oxygen levels, easy adaptation to artificial feeding and high growth. Microbiological contamination of fish flesh occurs only after capture and can be endogenous or exogenous contamination. Generally, several processes are used to overcome this problem of contamination of fish. Hot smoking and drying are very often used. The lack of hygiene in the traditional technology of smoking fish sometimes favor a considerable microbial contamination of the obtained products. Thus, contaminated fish can be the cause of foodborne diseases such as food poisoning. Most of the pathogenic strains responsible for these diseases are resistant to certain antibiotics

    Circulating adrenomedullin estimates survival and reversibility of organ failure in sepsis: the prospective observational multinational Adrenomedullin and Outcome in Sepsis and Septic Shock-1 (AdrenOSS-1) study

    Background: Adrenomedullin (ADM) regulates vascular tone and endothelial permeability during sepsis. Levels of circulating biologically active ADM (bio-ADM) show an inverse relationship with blood pressure and a direct relationship with vasopressor requirement. In the present prospective observational multinational Adrenomedullin and Outcome in Sepsis and Septic Shock 1 (, AdrenOSS-1) study, we assessed relationships between circulating bio-ADM during the initial intensive care unit (ICU) stay and short-term outcome in order to eventually design a biomarker-guided randomized controlled trial. Methods: AdrenOSS-1 was a prospective observational multinational study. The primary outcome was 28-day mortality. Secondary outcomes included organ failure as defined by Sequential Organ Failure Assessment (SOFA) score, organ support with focus on vasopressor/inotropic use, and need for renal replacement therapy. AdrenOSS-1 included 583 patients admitted to the ICU with sepsis or septic shock. Results: Circulating bio-ADM levels were measured upon admission and at day 2. Median bio-ADM concentration upon admission was 80.5 pg/ml [IQR 41.5-148.1 pg/ml]. Initial SOFA score was 7 [IQR 5-10], and 28-day mortality was 22%. We found marked associations between bio-ADM upon admission and 28-day mortality (unadjusted standardized HR 2.3 [CI 1.9-2.9]; adjusted HR 1.6 [CI 1.1-2.5]) and between bio-ADM levels and SOFA score (p &lt; 0.0001). Need of vasopressor/inotrope, renal replacement therapy, and positive fluid balance were more prevalent in patients with a bio-ADM &gt; 70 pg/ml upon admission than in those with bio-ADM ≀ 70 pg/ml. In patients with bio-ADM &gt; 70 pg/ml upon admission, decrease in bio-ADM below 70 pg/ml at day 2 was associated with recovery of organ function at day 7 and better 28-day outcome (9.5% mortality). By contrast, persistently elevated bio-ADM at day 2 was associated with prolonged organ dysfunction and high 28-day mortality (38.1% mortality, HR 4.9, 95% CI 2.5-9.8). Conclusions: AdrenOSS-1 shows that early levels and rapid changes in bio-ADM estimate short-term outcome in sepsis and septic shock. These data are the backbone of the design of the biomarker-guided AdrenOSS-2 trial. Trial registration: ClinicalTrials.gov, NCT02393781. Registered on March 19, 2015
