In planta localisation patterns of MADS domain proteins during floral development in Arabidopsis thaliana
Background: MADS domain transcription factors play important roles in various developmental processes in flowering plants. Members of this family play a prominent role in the transition to flowering and the specification of floral organ identity. Several studies reported mRNA expression patterns of the genes encoding these MADS domain proteins, however, these studies do not provide the necessary information on the temporal and spatial localisation of the proteins. We have made GREEN FLUORESCENT PROTEIN (GFP) translational fusions with the four MADS domain proteins SEPALLATA3, AGAMOUS, FRUITFULL and APETALA1 from the model plant Arabidopsis thaliana and analysed the protein localisation patterns in living plant tissues by confocal laser scanning microscopy (CLSM). Results: We unravelled the protein localisation patterns of the four MADS domain proteins at a cellular and subcellular level in inflorescence and floral meristems, during development of the early flower bud stages, and during further differentiation of the floral organs. The protein localisation patterns revealed a few deviations from known mRNA expression patterns, suggesting a non-cell autonomous action of these factors or alternative control mechanisms. In addition, we observed a change in the subcellular localisation of SEPALLATA3 from a predominantly nuclear localisation to a more cytoplasmic localisation, occurring specifically during petal and stamen development. Furthermore, we show that the down-regulation of the homeodomain transcription factor WUSCHEL in ovular tissues is preceded by the occurrence of both AGAMOUS and SEPALLATA3 proteins, supporting the hypothesis that both proteins together suppress WUSCHEL expression in the ovule. Conclusion: This approach provides a highly detailed in situ map of MADS domain protein presence during early and later stages of floral development. The subcellular localisation of the transcription factors in the cytoplasm, as observed at certain stages during development, points to mechanisms other than transcriptional control. Together this information is essential to understand the role of these proteins in the regulatory processes that drive floral development and leads to new hypotheses
HESS J1507-622: an unique unidentified source off the Galactic Plane
Galactic very high energy (VHE, > 100 GeV) gamma ray sources in the inner
Galaxy H.E.S.S. survey tend to cluster within 1 degree in latitude around the
Galactic plane. HESS J1507-622 instead is unique, since it is located at
latitude of ~3.5 degrees. HESS J1507-622 is slightly extended over the PSF of
the instrument and hence its Galactic origin is clear. The search for
counterparts in other wavelength regimes (radio, infrared and X-rays) failed to
show any plausible counterparts; and given its position off the Galactic plane
and hence the absorption almost one order of magnitude lower, it is very
surprising to not see any counterparts especially at X-rays wavelengths (by
ROSAT, XMM Newton and Chandra). Its latitude implies that it is either rather
close, within about 1 kpc, or is located well off the Galactic plane. And also
the models reflect the uniqueness of this object: a leptonic PWN scenario would
place this source due to its quite small extension to multi-kpc distance
whereas a hadronic scenario would preferentially locate this object at
distances of < 1 kpc where the density of target material is higher
Unidentified Galactic High-Energy Sources as Ancient Pulsar Wind Nebulae in the light of new high energy observations and the new code
In a Pulsar Wind Nebula (PWN), the lifetime of inverse Compton (IC) emitting
electrons exceeds the lifetime of its progenitor pulsar (as well as its
shell-type remnant), but it also exceeds the age of those that emit via
synchrotron radiation. Therefore, during its evolution, the PWN can remain
bright in IC so that its GeV-TeV gamma-ray flux remains high for timescales
much larger (for 10^5 - 10^6 yrs) than the pulsar lifetime and the X-ray PWN
lifetime. In this scenario, the magnetic field in the cavity induced by the
wind of the progenitor star plays a crucial role. This scenario is in line with
the discovery of several unidentified or "dark" sources in the TeV gamma-ray
band without X-ray counterparts; and it is also finding confirmation in the
recent discoveries at GeV gamma rays. Moreover, these consequences could be
also important for reinterpreting the detection of starburst galaxies in the
TeV gamma-ray band when considering a leptonic origin of the gamma-ray signal.
Both theoretical aspects and their observational proofs will be discussed, as

Broadband multi-wavelength campaign on PKS 2005-489
The spectral energy distribution (SED) of high-frequency peaked BL Lac
objects (HBL) is characterized by two peaks: one in the UV-X-ray and one in the
GeV-TeV regime. An interesting object for analyzing these broadband
characteristics is PKS 2005-489, which in 2004 showed the softest TeV spectrum
ever measured. In 2009, a multi-wavelength campaign has been conducted with,
for the first time, simultaneous observations by H.E.S.S. (TeV), Fermi/LAT
(GeV), RXTE (keV), Swift (keV, UV, optical) and ATOM (optical) to cover the two
peaks of the SED. During this campaign PKS 2005-489 underwent a high state in
all wavebands which gives the opportunity to study in detail the emission

Fertility control in a male rabbit using a deslorelin implant. A case report
[EN] Continuous low-dose release of the GnRH analogue deslorelin (Suprelorin, Peptech, Australia) causes a suppression of reproductive function in different species such as dogs, koalas, or kangaroos. No studies concerning the efficacy and safety in male rabbits have yet been conducted. A male rabbit with hypospadia was introduced to the Clinic for Animal Reproduction. The owner intended to keep it together with a fertile female rabbit. To avoid reproduction, a 4.7 mg deslorelin implant was injected subcutaneously. No negative reaction to implant placement was diagnosed by daily examination of the injection site for one week. To test the effectiveness of the treatment, blood samples were taken and the testes size was measured regularly. After an initial rise in testosterone levels for 14 d, a down regulation for seven month with values lower than 0.1 ng/mL was observed. In the same period, the size of the testes declined to 50 % of the original dimensions. Afterwards, both testes regained their former shape and size. Since it was not a stud rabbit, semen parameters were not determined. During the suppression of the reproductive function, no sexual activity was observed by the owners. These findings have yet to be proven by clinical trials with a sufficient number of animals, but this case indicates the efficacy of deslorelin implants in male rabbits.Arlt, S.; Spankowski, S.; Kaufmann, T.; Kostelnik, K.; Heuwieser, W. Toxicity of statins on rat skeletal muscle mitochondria
Abstract.: We investigated mitochondrial toxicity of four lipophilic stains (cerivastatin, fluvastatin, atorvastatin, simvastatin) and one hydrophilic statin (pravastatin). In L6 cells (rat skeletal muscle cell line), the four lipophilic statins (100μmol/l) induced death in 27-49% of the cells. Pravastatin was not toxic up to 1mmol/l. Cerivastatin, fluvastatin and atorvastatin (100μmol/l) decreased the mitochondrial membrane potential by 49-65%, whereas simvastatin and pravastatin were less toxic. In isolated rat skeletal muscle mitochondria, all statins, except pravastatin, decreased glutamate-driven state 3 respiration and respiratory control ratio. Beta-oxidation was decreased by 88-96% in the presence of 100μmol/l of the lipophilic statins, but only at higher concentrations by pravastatin. Mitochondrial swelling, cytochrome c release and DNA fragmentation was induced in L6 cells by the four lipophilic statins, but not by pravastatin. Relationship between hippocampal structure and memory function in elderly humans
With progressing age, the ability to recollect personal events declines, whereas familiarity-based memory remains relatively intact. It has been hypothesized that age-related hippocampal atrophy may contribute to this pattern because of its critical role for recollection in younger humans and after acute injury. Here, we show that hippocampal volume loss in healthy older persons correlates with gray matter loss (estimated with voxel-based morphometry) of the entire limbic system and shows no correlation with an electrophysiological (event-related potential [ERP]) index of recollection. Instead, it covaries with more substantial and less specific electrophysiological changes of stimulus processing. Age-related changes in another complementary structural measure, hippocampal diffusion, on the other hand, seemed to be more regionally selective and showed the expected correlation with the ERP index of recollection. Thus, hippocampal atrophy in older persons accompanies limbic atrophy, and its functional impact on memory is more fundamental than merely affecting recollection
