Freeform Bioprinting of Liver Encapsulated in Alginate Hydrogels Tissue Constructs for Pharmacokinetic Study
An in vitro model that can be realistically and inexpensively used to predict human response to
various drug administration and toxic chemical exposure is needed. By fabricating a microscale
3D physiological tissue construct consisting of an array of channels and tissue-embedded
chambers, one can selectively develop various biomimicking mammalian tissues for a number of
pharmaceutical applications, for example, experimental pharmaceutical screening for drug
efficacy and toxicity along with apprehending the disposition and metabolic profile of a
candidate drug. This paper addresses issues relating to the development and implementation of a
bioprinting process for freeform fabrication of a 3D cell-encapsulated hydrogel-based tissue
construct, the direct integration onto a microfluidic device for pharmacokinetic study, and the
underlying engineering science for the fabrication of a 3D microscale tissue chamber as well as
its application in pharmacokinetic study. To this end, a prototype 3D microfluidic tissue chamber
embedded with liver cells encapsulated within a hydrogel matrix construct is bioprinted as a
physiological in vitro model for pharmacokinetic study. The developed fabrication processes are
further validated and parameters optimized by assessing cell viability and liver cell phenotype, in
which metabolic and synthetic liver functions are quantitated.Mechanical Engineerin
No future in archaeological heritage management?
Although the future is mentioned frequently in overarching aims and visions, and it is a major drive in the daily work of archaeological heritage managers and indeed heritage professionals more generally, it remains unclear precisely how an overall commitment to the future can best inform specific heritage practices. It seems that most archaeologists and other heritage professionals cannot easily express how they conceive of the future they work for, and how their work will impact on that future. The future tends to remain implicit in daily practice which operates in a continuing, rolling present. The authors argue that this needs to change because present-day heritage management may be much less beneficial for the future than we commonly expect
Heritage Futures: Comparative Approaches to Natural and Cultural Heritage Practices
Preservation of natural and cultural heritage is often said to be something that is done for the future, or on behalf of future generations, but the precise relationship of such practices to the future is rarely reflected upon. Heritage Futures draws on research undertaken over four years by an interdisciplinary, international team of 16 researchers and more than 25 partner organisations to explore the role of heritage and heritage-like practices in building future worlds. Engaging broad themes such as diversity, transformation, profusion and uncertainty, Heritage Futures aims to understand how a range of conservation and preservation practices across a number of countries assemble and resource different kinds of futures, and the possibilities that emerge from such collaborative research for alternative approaches to heritage in the Anthropocene. Case studies include the cryopreservation of endangered DNA in frozen zoos, nuclear waste management, seed biobanking, landscape rewilding, social history collecting, space messaging, endangered language documentation, built and natural heritage management, domestic keeping and discarding practices, and world heritage site management
Negative Experiences in Television Programming : A View from the Other Side
Many professional archaeologists will engage with television. There seems to be little guidance- other than trial and error and personal communications - for navigating interactions with media production companies. Here, through a series of informal questions and answers, I present some issues with which one might be confronted in such engagements. I base my discussions on personal experience, literature, and conversations with experienced colleagues. A key point is that media production companies' agendas and priorities often differ from those of archaeologists. While the article draws on experiences from Europe and North America, it has relevance for archaeologists working with television everywhere.Peer reviewe

Patient Preference Studies During Early Drug Development: Aligning Stakeholders to Ensure Development Plans Meet Patient Needs
Although patient preferences have been studied broadly for marketed products or around the time of submission to authorities and launch, patient preference studies have rarely been used during the early drug development phases. In this paper, we formulate three hypotheses supporting the use of patient preference studies in early product development: (1) integration of the patient perspective into the development process from phase 1 onwards will result in healthcare solutions with outcomes that best address patients' needs; (2) a structured process to build patient-based evidence involving partnerships between patients and other key stakeholders will improve alignment of development activities with the needs of patients; (3) quantitative patient preference research built on robust qualitative insights is necessary to strengthen development decisions in the interests of patients. To illustrate such a structured process, we describe qualitative insights research (social media analysis and online bulletin boards) and quantitative patient preference studies in dry eye disease and non-alcoholic steatohepatitis conducted during early product development by a pharmaceutical company to generate patient-based evidence. The outputs from such early patient preference studies are being used to inform patient reported outcome strategies, clinical development strategies, product design and delivery features, and form the basis for early dialog with regulators, health technology assessment (HTA) bodies and payers to ensure focus and alignment on patient-relevant endpoints. Furthermore, to discuss and theoretically substantiate our hypotheses, we review how different groups and organizations are working to embrace fully the patient perspective in product development and healthcare decision-making. The hypotheses are commensurate with the general trend toward patient-centered healthcare and the activities initiated by regulators, HTA agencies, and patient organizations. We advocate that all healthcare players should actively contribute to aligning on best practices concerning choice of methodologies and engage in multi-stakeholder dialog along the entire product development chain, to realize health technologies that best meet the needs of patients
Heritage Futures - Comparative Approaches to Natural and Cultural Heritage Practices
This is the final version. Available on open access from UCL Press via the DOI in this recordWe present the book as a co-authored monograph because we acknowledge the collective contributions to the arguments developed within it, and the collaborative nature of the work. This has in itself been an experiment in finding a format in which diverse voices and views could productively speak to one another, while also acknowledging and foregrounding the diversity and range of different views, academic traditions and writing styles of contributors. As principal investigator, Harrison acted as the lead and coordinating author of the book, taking overall responsibilities for its editing and production. The co-investigators (DeSilvey, Holtorf, Macdonald) shared with Harrison editorial responsibilities for the individual thematic parts they each led, and for shaping the intellectual agenda of the book as a whole. However, we also felt it important to indicate the main authors of individual chapters within the book, to make clear specific contributions to the text and its arguments, and to highlight which named individuals were responsible for the empirical work that underpins them.

Effectiveness of Compounded Bioidentical Hormone Replacement Therapy: An Observational Cohort Study
<p>Abstract</p> <p>Background</p> <p>Bioidentical Hormone Replacement Therapy (BHRT) is believed it to be a safer and equally effective alternative to Conventional Hormone Therapy for the relief of menopausal symptoms; however, data are needed to support these claims. The objective of this study is to evaluate the effectiveness of compounded BHRT provided in six community pharmacies.</p> <p>Methods</p> <p>This was an observational cohort study of women between the ages of 18-89 who received a compounded BHRT product from January 1, 2003 to April 30, 2010 in six community pharmacies. Data included patient demographics, comorbidities, therapeutic outcomes, and hormone therapies. Women self-rated menopausal symptoms as absent, mild, moderate, or severe. Descriptive statistics were used to characterize the patient population, BHRT use, and adverse events. Patient symptom severity was compared at baseline and 3 to 6 months follow-up using the Wilcoxon signed-rank test.</p> <p>Results</p> <p>Women (n = 296) receiving BHRT at Oakdell Pharmacy had a mean (standard deviation) age of 52 (9) years. The most common BHRT dosage forms utilized were topical (71%) and oral (43%). Compounded BHRT regimens were generally initiated at low doses regardless of route. Women experienced a 25% decrease in emotional lability (p < 0.01), a 25% decrease in irritability (p < 0.01), and a 22% reduction in anxiety (p = 0.01) within 3 to 6 months. These women also experienced a 14% reduction in night sweats (p = 0.09) and a 6% reduction in hot flashes (p = 0.50).</p> <p>Conclusions</p> <p>This study demonstrates that compounded BHRT improves mood symptoms. Larger studies are needed to examine the impact on vasomotor symptoms, myocardial infarction and breast cancer.</p
