    Airway structural cells regulate TLR5-mediated mucosal adjuvant activity

    Antigen-presenting cell (APC) activation is enhanced by vaccine adjuvants. Most vaccines are based on the assumption that adjuvant activity of Toll-like receptor (TLR) agonists depends on direct, functional activation of APCs. Here, we sought to establish whether TLR stimulation in non-hematopoietic cells contributes to flagellin’s mucosal adjuvant activity. Nasal administration of flagellin enhanced T-cell-mediated immunity, and systemic and secretory antibody responses to coadministered antigens in a TLR5-dependent manner. Mucosal adjuvant activity was not affected by either abrogation of TLR5 signaling in hematopoietic cells or the presence of flagellin-specific, circulating neutralizing antibodies. We found that flagellin is rapidly degraded in conducting airways, does not translocate into lung parenchyma and stimulates an early immune response, suggesting that TLR5 signaling is regionalized. The flagellin-specific early response of lung was regulated by radioresistant cells expressing TLR5 (particularly the airway epithelial cells). Flagellin stimulated the epithelial production of a small set of mediators that included the chemokine CCL20, which is known to promote APC recruitment in mucosal tissues. Our data suggest that (i) the adjuvant activity of TLR agonists in mucosal vaccination may require TLR stimulation of structural cells and (ii) harnessing the effect of adjuvants on epithelial cells can improve mucosal vaccines.Fil: Van Maele, Laurye.     IL-17A/F-Signaling Does Not Contribute to the Initial Phase of Mucosal Inflammation Triggered by S. Typhimurium

    Salmonella enterica subspecies 1 serovar Typhimurium (S. Typhimurium) causes diarrhea and acute inflammation of the intestinal mucosa. The pro-inflammatory cytokines IL-17A and IL-17F are strongly induced in the infected mucosa but their contribution in driving the tissue inflammation is not understood. We have used the streptomycin mouse model to analyze the role of IL-17A and IL-17F and their cognate receptor IL-17RA in S. Typhimurium enterocolitis. Neutralization of IL-17A and IL-17F did not affect mucosal inflammation triggered by infection or spread of S. Typhimurium to systemic sites by 48 h p.i. Similarly, Il17ra−/− mice did not display any reduction in infection or inflammation by 12 h p.i. The same results were obtained using S. Typhimurium variants infecting via the TTSS1 type III secretion system, the TTSS1 effector SipA or the TTSS1 effector SopE. Moreover, the expression pattern of 45 genes encoding chemokines/cytokines (including CXCL1, CXCL2, IL-17A, IL-17F, IL-1α, IL-1ÎČ, IFNÎł, CXCL-10, CXCL-9, IL-6, CCL3, CCL4) and antibacterial molecules was not affected by Il17ra deficiency by 12 h p.i. Thus, in spite of the strong increase in Il17a/Il17f mRNA in the infected mucosa, IL-17RA signaling seems to be dispensable for eliciting the acute disease. Future work will have to address whether this is attributable to redundancy in the cytokine signaling network

    A new mathematical development has allowed the White and Dantzig method of calculating equilibrium concentrations to be applied to gas mixtures in thermal disequilibrium. The general principle of the method remains the search for the minimum free enthalpy of the mixture, but the calculation conditions have changed since the temperature is no longer uniform: it is therefore possible to deal with cases of mixtures of plasmas where the electrons are generally hotter than the heavier species. The mathematical development and the development of a micro-computer algorithm are described.Un nouveau dĂ©veloppement mathĂ©matique a permis d'adapter au cas d'un mĂ©lange gazeux en dĂ©sĂ©quilibre thermique la mĂ©thode de calcul des concentrations Ă  l'Ă©quilibre de White et Dantzig.     Plasmas jets are studied at low pressure (10-1 - 10 torr) and high temperature (2000 K - 8000 K) to simulate a lot of parameters of re-entry plasma. The experimental apparatus is described and the used diagnostic techniques as electrostatic probes, mass spectrometer, MHD and optical spectrometer are presented. The obtained plasmas are modelized with a two-temperature model (electron temperature and heavy particule temperature).Des jets de plasma sont étudiés à basse pression (10-1 - 10 torr) et à haute température (2000-8000 K) pour simuler certains paramÚtres caractérisant le plasma de ré-entrée d'un engin dans la zone atmosphérique. L'ensemble expérimental est décrit et les méthodes de diagnostic telles que les sondes électrostatiques, le spectromÚtre de masse, la MHD et la spectroscopie optique d'émission sont présentées. Les plasmas obtenus expérimentalement sont modélisés en utilisant une approche à deux températures (température électronique et température des espÚces lourdes)
