    Effectiveness of Large Scale Water and Sanitation Interventions: the One Million Initiative in Mozambique

    The One Million Initiative aims for 70% of the population using safe drinking water and 50% using improved sanitation in 18 districts in three provinces of central Mozambique. This paper evaluates health effects of the initiative using a panel survey, analyzing the contributions of latrine ownership, hand washing, and use of improved water sources to health. It is unique in providing rigorous evaluation of such a large scale WASH program. The authors find a large impact of CLTS on adults and older children, and a similar impact in improved water sources on health of young children

    Membrane fluidity matters: Hyperthermia from the aspects of lipids and membranes

    Hyperthermia is a promising treatment modality for cancer in combination both with radio- and chemotherapy. In spite of its great therapeutic potential, the underlying molecular mechanisms still remain to be clarified. Due to lipid imbalances and 'membrane defects' most of the tumour cells possess elevated membrane fluidity. However, further increasing membrane fluidity to sensitise to chemo-or radiotherapy could have some other effects. In fact, hyperfluidisation of cell membrane induced by membrane fluidiser initiates a stress response as the heat shock protein response, which may modulate positively or negatively apoptotic cell death. Overviewing some recent findings based on a technology allowing direct imaging of lipid rafts in live cells and lipidomics, novel aspects of the intimate relationship between the 'membrane stress' of tumour cells and the cellular heat shock response will be highlighted. Our findings lend support to both the importance of membrane remodelling and the release of lipid signals initiating stress protein response, which can operate in tandem to control the extent of the ultimate cellular thermosensitivity. Overall, we suggest that the fluidity variable of membranes should be used as an independent factor for predicting the efficacy of combinational cancer therapies

    The agency of liminality: army wives in the DR Congo and the tactical reversal of militarization

    The inherently unstable boundaries between military and civilian worlds have emerged as a main object of study within the field of critical military studies. This article sheds light on the (re)production of these boundaries by attending to a group that rarely features in the debates on the military/civilian divide: army wives in a ‘non-Northern’ context, more specifically the Democratic Republic of the Congo (DRC). Drawing upon the ‘analytical toolbox’ of governmentality, we explore how civilian and military positionalities are called upon, articulated and subverted in the governing and self-governing of Congolese army wives. By foregrounding the relevance of studying Congolese army wives and their militarization with an analytical toolbox often reserved for so called ‘advanced militaries/societies’, and by revealing numerous similarities between the Congolese and ‘Northern’ contexts, the article also sets out to counter the Euro/US-centrism and ‘theoretical discrimination’ that mark present-day (critical) military studies

    The Crystal Structure of the Hsp90-LA1011 Complex and the Mechanism by Which LA1011 May Improve the Prognosis of Alzheimer's Disease

    Functional changes in chaperone systems play a major role in the decline of cognition and contribute to neurological pathologies, such as Alzheimer's disease (AD). While such a decline may occur naturally with age or with stress or trauma, the mechanisms involved have remained elusive. The current models suggest that amyloid-& beta; (A & beta;) plaque formation leads to the hyperphosphorylation of tau by a Hsp90-dependent process that triggers tau neurofibrillary tangle formation and neurotoxicity. Several co-chaperones of Hsp90 can influence the phosphorylation of tau, including FKBP51, FKBP52 and PP5. In particular, elevated levels of FKBP51 occur with age and stress and are further elevated in AD. Recently, the dihydropyridine LA1011 was shown to reduce tau pathology and amyloid plaque formation in transgenic AD mice, probably through its interaction with Hsp90, although the precise mode of action is currently unknown. Here, we present a co-crystal structure of LA1011 in complex with a fragment of Hsp90. We show that LA1011 can disrupt the binding of FKBP51, which might help to rebalance the Hsp90-FKBP51 chaperone machinery and provide a favourable prognosis towards AD. However, without direct evidence, we cannot completely rule out effects on other Hsp90-co-chaprone complexes and the mechanisms they are involved in, including effects on Hsp90 client proteins. Nonetheless, it is highly significant that LA1011 showed promise in our previous AD mouse models, as AD is generally a disease affecting older patients, where slowing of disease progression could result in AD no longer being life limiting. The clinical value of LA1011 and its possible derivatives thereof remains to be seen

    Playing the victim? Human trafficking, African youth, and geographies of structural inequality

    This article explores the role of agency in determining who is and is not considered to be a legitimate victim of human trafficking. It draws on critical human trafficking scholarship and research on the life chances of West African youth. This is complemented by qualitative data from youth embroiled in football-related human trafficking. The insights from these analyses are brought into conversation with theoretical work on the geographies of commodities. This results in the concept of ‘unveiling the football trafficking fetish’, which is used to theorise how and why the agency of mobile youthful African male bodies undermines their claims of being trafficked human beings. The findings that emerge are significant in two ways. First, they generate theoretical insights on the coexistence of agency and exploitation in young lives, and how young people’s aspirations and agency can be (mis)read and work against them. Second, they provide a unique illustration of how human trafficking is a product of capitalism yet can be presented as a form of behaviour that lies outside of capitalist social relations. To centre these social relations and foster new forms of critical dialogue within and beyond population geography, the article concludes by recommending we consider the implications of conceptualising people as susceptible to rather than vulnerable to human trafficking

    Electrosprayed core-shell nanoparticles of PVP and shellac for furnishing biphasic controlled release of ferulic acid

    Coaxial electrospraying was explored to organize polymer excipients in a core-shell manner for providing biphasic controlled release of active ingredient. With ferulic acid (FA) as a model drug, and shellac and polyvinylpyrrolidone (PVP) as the core and shell polymeric matrices, core-shell nanoparticles were successfully fabricated. A series of tests were carried out to characterize the prepared core-shell nanoparticles and also the nanoparticles prepared using a single fluid electrospraying of the shell or core fluids alone. The core-shell nanoparticles had an average diameter of 530 ± 80 nm with clear core-shell structure. The contained FA was converted to an amorphous state both in the core and the shell parts due to the favorable hydrogen bonding between the components. In vitro dissolution tests demonstrated that the core-shell nanoparticles were able to provide the desired biphasic drug-controlled release profiles.     Heat stress causes spatially-distinct membrane re-modelling in K562 leukemia cells

    Cellular membranes respond rapidly to various environmental perturbations. Previously we showed that modulations in membrane fluidity achieved by heat stress (HS) resulted in pronounced membrane organization alterations which could be intimately linked to the expression and cellular distribution of heat shock proteins. Here we examine heat-induced membrane changes using several visualisation methods. With Laurdan two-photon microscopy we demonstrate that, in contrast to the enhanced formation of ordered domains in surface membranes, the molecular disorder is significantly elevated within the internal membranes of cells preexposed to mild HS. These results were compared with those obtained by anisotropy, fluorescence lifetime and electron paramagnetic resonance measurements. All probes detected membrane changes upon HS. However, the structurally different probes revealed substantially distinct alterations in membrane heterogeneity. These data call attention to the careful interpretation of results obtained with only a single label. Subtle changes in membrane microstructure in the decision-making of thermal cell killing could have potential application in cancer therapy

    Electrospinning scale-up and formulation development of PVA nanofibers aiming oral delivery of biopharmaceuticals

    Electrospinning is a promising drying technology providing a rapid and gentle drying at ambient temperature, thus electrospinning of polyvinyl alcohol aqueous solutions was investigated for the solid formulation of biopharmaceuticals. The commonly used single-needle electrospinning does not have adequate productivity to satisfy the industrial requirements, therefore our aim was to study the scale-up of the technology by using high-speed electrospinning. High molecular weight polyethylene oxide as a secondary polymer was applied to enhance the fiber formation of polyvinyl alcohol. While polyvinyl alcohol-polyethylene oxide formulations resulted in adequate fiber formation it was not possible to process them further as the friability of the fibers was too low. In order to increase the friability, the effect of adding various sugars (mannitol, glucose, lactose, saccharose, and trehalose) was investigated. The results showed that mannitol was the best friability enhancing excipient because of its crystallinity and low moisture content in the fibrous sample. In contrast, glucose, lactose, saccharose, and trehalose were amorphous with higher moisture content and fibers containing these were grindable only after post-drying
