    ISOLATION OF PHYTOCONSTITUENT, IN VITRO ANTICANCER STUDY IN HELA AND MCF-7 CELL LINES AND MOLECULAR DOCKING STUDIES OF POTHOS SCANDENS LINN

    Objective: This study aims to isolate an active phytoconstituent from ethanolic leaf extract of Pothos scandens Linn., to evaluate in vitro anticancer activity, and to carry out molecular docking studies of the isolated phytoconstituent. Methods: The bioactive constituent 1,1’-(4,5-dihydroxy benzene-1,2-diyl) bisoct-7-en-1-one, a phenolic compound, was isolated by using chromatographic methods and the structure was elucidated by various spectroscopic techniques. In vitro anticancer activity was evaluated against HeLa and MCF 7 cell lines. The viability of cells was evaluated by direct observation of cells by an Inverted phase-contrast microscope and by the MTT assay method. IC50 was calculated using the linear regression model. Results: The results of anticancer studies revealed that different concentrations of the ethanolic extract of leaves of Pothos scandens Linn. exhibited cytotoxic activity against HeLa and MCF 7 cell lines with IC50 of 22.9 and 18.32 ÎĽg/ml, respectively. The anticancer potential of the plant was revalidated by in silico molecular docking study with Vascular Endothelial Growth Factor Receptor 2 (VEGFR2, PDB ID: 4AG8) using Discovery studio 2018. Results of the docking study showed that the ligand exhibited strong interaction with the VEGFR2 kinase with significant binding energy. Conclusion: Pothos scandens linn. can be used for the isolation of potent anticancer agents

    Utilizing Proximity for Increasing Student Knowledge Retention: A Near-Peer Tutoring Program Needs Study

    The purpose of this study was to demonstrate the desire and need for peer-to-peer tutoring programs at Division 1 Dell Medical School at the University of Texas. Materials and Methods. Two sets of surveys were created and sent to students at the Dell Medical School, University of Texas, USA. One survey asking about the need or desire to engage with a peer tutor was sent to first-year students, and another one asking about the desire to provide these services to underclassmen as a potential leadership course option was sent to third-year students. Results. For the first-year student survey, 52.9% of respondents disagreed or strongly disagreed of being aware of near-peer tutoring and 70.5% of respondents either agreed or strongly agreed that it would be an option utilized by students. For the third-year student survey, 75% of students either disagreed or felt neutral in being aware of near-peer tutoring as an option to serve underclassmen, whereas 65% of upperclassmen either agreed or strongly agreed that if near-peer tutoring had been offered, they would have chosen this leadership course option in the effort to teach their underclassmen peers. Conclusions. Numerous studies have demonstrated peer-to-peer tutorial options to be of high utility to students in the medical education space. This particular paper obtained results demonstrating students’ desire to engage in peer tutoring voluntarily for their own course success goals and upperclassmen’s desire to participate as near-peer tutors for the benefit of underclassmen

    Maternal health services in South Africa During the 10th anniversary of the WHO 'Safe Motherhood' initiative

    The tenth anniversary of the World Health Organisation's 'Safe Motherhood' initiative is being celebrated this year and the organisation is using the opportunity to assess critically its gains, its strengths and its weaknesses. South Africa has taken some bold steps to address maternal health services, specifically introducing free health care for pregnant women and children under 5. In this paper we explore what further steps are necessary to ensure improved health outcome for pregnant women. South African health care administrations are, in some cases, engaged in broad health systems interventions at provincial level. This approach to improving health services is nonetheless frustrated by programme-specific initiatives, such as the introduction of female condoms or other piecemeal additions. We argue that making the systems function is the essential, primary step in the success of any intervention. The case of maternal health is explored in this paper

    Creative group performances to assess core competencies in a first-year patient-centered medicine course

    Introduction: A novel assessment of systems-based practice and practice-based learning and improvement learning objectives, implemented in a first-year patient-centered medicine course, is qualitatively described. Methods: Student learning communities were asked to creatively demonstrate a problem and solution for health care delivery. Skits, filmed performances, plays, and documentaries were chosen by the students. Video recordings were reviewed for themes and the presence of course competencies. Results: All performances demonstrated not only the index competencies of team work and facilitation of the learning of others, but many other core objectives of the course. The assignment was rated positively both by the faculty and the students, and has been added to the assessment modalities of the course

    Part II. high-dose methotrexate with leucovorin rescue for severe COVID-19: An immune stabilization strategy for SARS-CoV-2 induced \u27PANIC\u27 attack

    Here, in Part II of a duology on the characterization and potential treatment for COVID-19, we characterize the application of an innovative treatment regimen for the prevention of the transition from mild to severe COVID-19, as well as detail an intensive immunotherapy intervention hypothesis. We propose as a putative randomized controlled trial that high-dose methotrexate with leucovorin (HDMTX-LR) rescue can abolish \u27PANIC\u27, thereby \u27left-shifting\u27 severe COVID-19 patients to the group majority of those infected with SARS-CoV-2, who are designated as having mild, even asymptomatic, disease. HDMTX-LR is endowed with broadly pleiotropic properties and is a repurposed, generic, inexpensive, and widely available agent which can be administered early in the course of severe COVID-19 thus rescuing the critical and irreplaceable gas-exchange alveoli. Further, we describe a preventative treatment intervention regimen for those designated as having mild to moderate COVID-19 disease, but who exhibit features which herald the transition to the severe variant of this disease. Both of our proposed hypothesis-driven questions should be urgently subjected to rigorous assessment in the context of randomized controlled trials, in order to confirm or refute the contention that the approaches characterized herein, are in fact capable of exerting mitigating, if not abolishing, effects upon SARS-CoV-2 triggered \u27PANIC Attack\u27. Confirmation of our immunotherapy hypothesis would have far-reaching ramifications for the current pandemic, along with yielding invaluable lessons which could be leveraged to more effectively prepare for the next challenge to global health
