    First Ever Observations of Mineral Dust in Wintertime over Warsaw, Poland

    The long-range transport of desert dust over the area of the temperate climate zone is associated with the influx of hot air masses due to the location of the sources of this aerosol in the tropical climate zone. Between 24–26 February 2021, such an aerosol outbreak took place and reached Central Europe. The mean temperature of +11.7 °C was recorded during the event. A comparison of this value to the 20-year (2000–2020) average February temperature for Warsaw (−0.2 °C) indicates the uniqueness of the meteorological conditions. It was the first wintertime inflow of Saharan dust over Warsaw, the presence of which was confirmed by lidar and sun-photometer measurements. The properties of the desert dust layers were obtained; the mean values of the particle depolarization for the fully developed mineral dust layer were 13 ± 3% and 22 ± 4% for 355 and 532 nm, respectively. The aerosol optical thickness was high with average values >0.36 for all wavelengths smaller than 500 nm. The three-modal, aerosol size distribution was dominated by coarse-mode particles, with a visible contribution of accumulation-mode particles.     Spatio-temporal evolution of long-range transported mineral desert dust properties over rural and urban sites in Central Europe

    An exceptionally strong and very fast (120h) mineral dust inflow from North Africa to Poland was predicted by NMMB/BSC-Dust and NAAPS models on 10–11 June 2019. Simultaneous measurements with two complex lidar systems at the EARLINET-ACTRIS urban site in Warsaw (Central Poland) and the PolWET peatland site in Rzecin (Western Poland) captured the evolution of this dust event. The advected air masses had different source areas in North Africa, they were reaching each station via independent pathways, and thus, were unlikely mixed with each other. The excellent capabilities of the next generation PollyXT lidar and the mobile EMORAL lidar allowed for the derivation of full datasets of aerosol optical properties profiles that enabled comparative study of the advected dust properties evolution. Within a mere 350 km distance between Warsaw and Rzecin, distinctly different dust properties were measured, respectively: dry mineral dust composed mainly of coarse mode dust particles (50 ± 5 % of the total particle backscattering profile) versus the wet mineral dust dominated by fine dust particles (58 ± 4 %).     Mechanisms generating long-range correlation in nucleotide composition of the Borrelia burgdorferi genome

    Abstract We have analysed protein coding and intergenic sequences in the Borrelia burgdorferi (the Lyme disease bacterium) genome using di erent kinds of DNA walks. Genes occupying the leading strand of DNA have signiÿcantly di erent nucleotide composition from genes occupying the lagging strand. Nucleotide compositional bias of the two DNA strands re ects the aminoacid composition of proteins. 96% of genes coding for ribosomal proteins lie on the leading DNA strand, which suggests that the positions of these as well as other genes are non-random. In the B. burgdorferi genome, the asymmetry in intergenic DNA sequences is lower than the asymmetry in the third positions in codons. All these characters of the B. burgdorferi genome suggest that both replication-associated mutational pressure and recombination mechanisms have established the speciÿc structure of the genome and now any recombination leading to inversion of a gene in respect to the direction of replication is forbidden. This property of the genome allows us to assume that it is in a steady state, which enables us to ÿx some parameters for simulations of DNA evolution. c 1999 Elsevier Science B.V.     The Functional DRD3 Ser9Gly Polymorphism (rs6280) Is Pleiotropic, Affecting Reward as Well as Movement

    Abnormalities of motivation and behavior in the context of reward are a fundamental component of addiction and mood disorders. Here we test the effect of a functional missense mutation in the dopamine 3 receptor (DRD3) gene (ser9gly, rs6280) on reward-associated dopamine (DA) release in the striatum. Twenty-six healthy controls (HCs) and 10 unmedicated subjects with major depressive disorder (MDD) completed two positron emission tomography (PET) scans with [11C]raclopride using the bolus plus constant infusion method. On one occasion subjects completed a sensorimotor task (control condition) and on another occasion subjects completed a gambling task (reward condition). A linear regression analysis controlling for age, sex, diagnosis, and self-reported anhedonia indicated that during receipt of unpredictable monetary reward the glycine allele was associated with a greater reduction in D2/3 receptor binding (i.e., increased reward-related DA release) in the middle (anterior) caudate (p<0.01) and the ventral striatum (p<0.05). The possible functional effect of the ser9gly polymorphism on DA release is consistent with previous work demonstrating that the glycine allele yields D3 autoreceptors that have a higher affinity for DA and display more robust intracellular signaling. Preclinical evidence indicates that chronic stress and aversive stimulation induce activation of the DA system, raising the possibility that the glycine allele, by virtue of its facilitatory effect on striatal DA release, increases susceptibility to hyperdopaminergic responses that have previously been associated with stress, addiction, and psychosis

    Dynamics of the Atmospheric Boundary Layer over two middle-latitude rural sites with Doppler lidar

    The Atmospheric Boundary Layer (ABL) over two middle-latitude rural sites was characterized in terms of mean horizontal wind and turbulence sources using a standard classification methodology based on Doppler lidar. The first location was an irrigated olive orchard in ubeda (Southern Spain), representing one of the most important crops in the Mediterranean basin and a typical site with Mediterranean climate. The second location was PolWET peatland site in Rzecin (Northwestern Poland), representing one of the largest natural terrestrial carbon storages that have a strong interaction with the climate system. The results showed typical situations for non cloud-topped ABL cases, where ABL is fully developed during daytime due to convection, with high turbulent activity and strong positive skewness indicating frequent and powerful updrafts. The cloud-topped cases showed the strong influence that clouds can have on ABL development, preventing it to reach the same maximum height and introducing top-down movements as an important contribution to mixing. The statistical analysis of turbulent sources allowed for finding a common diurnal cycle for convective mixing at both sites, but nocturnal wind shear driven turbulence with marked differences in its vertical distribution. This analysis demonstrates the Doppler lidar measurements and the classification algorithm strong potential to characterize the dynamics of ABL in its full extent and with high temporal resolution. Moreover, some recommendations for future improvement of the classification algorithm were provided on the basis of the experience gained.Peer reviewe

    The effects of preoperative chemotherapy on isolated tumour cells in the blood and bone marrow of gastric cancer patients

    Recent studies in breast cancer suggest that monitoring the isolated tumour cells (ITC) may be used as a surrogate marker to evaluate the efficacy of systemic chemotherapy. In the present study, we have investigated the effects of preoperative chemotherapy on ITC in the blood and bone marrow of patients with potentially resectable gastric cancer. After sorting out the CD45-positive cells, the presence of ITC defined as cytokeratin-positive cells was examined before and after preoperative chemotherapy. The patients received two courses of preoperative chemotherapy with cisplatin (100 mg m−2, day 1) and 5-fluorouracil (1000 mg m−2, days 1–5), administered every 28 days. Fourteen of 32 (44%) patients initially diagnosed with ITC in blood and/or bone marrow were found to be negative (responders) after preoperative chemotherapy (P<0.01). The incidence of ITC in bone marrow was also significantly (P<0.01) reduced from 97 (31 of 32) to 53% (17 of 32). The difference between patients positive for ITC in the blood before (n=7, 22%) and after (n=5, 16%) chemotherapy was statistically insignificant. The overall 3-year survival rates were 32 and 49% in the responders and non-responders, respectively (P=0.683). These data indicate that preoperative chemotherapy can reduce the incidence of ITC in patients with gastric cancer

    Effects of betaine on lipopolysaccharide-induced memory impairment in mice and the involvement of GABA transporter 2

    <p>Abstract</p> <p>Background</p> <p>Betaine (glycine betaine or trimethylglycine) plays important roles as an osmolyte and a methyl donor in animals. While betaine is reported to suppress expression of proinflammatory molecules and reduce oxidative stress in aged rat kidney, the effects of betaine on the central nervous system are not well known. In this study, we investigated the effects of betaine on lipopolysaccharide (LPS)-induced memory impairment and on mRNA expression levels of proinflammatory molecules, glial markers, and GABA transporter 2 (GAT2), a betaine/GABA transporter.</p> <p>Methods</p> <p>Mice were continuously treated with betaine for 13 days starting 1 day before they were injected with LPS, or received subacute or acute administration of betaine shortly before or after LPS injection. Then, their memory function was evaluated using Y-maze and novel object recognition tests 7 and 10-12 days after LPS injection (30 μg/mouse, i.c.v.), respectively. In addition, mRNA expression levels in hippocampus were measured by real-time RT-PCR at different time points.</p> <p>Results</p> <p>Repeated administration of betaine (0.163 mmol/kg, s.c.) prevented LPS-induced memory impairment. GAT2 mRNA levels were significantly increased in hippocampus 24 hr after LPS injection, and administration of betaine blocked this increase. However, betaine did not affect LPS-induced increases in levels of mRNA related to inflammatory responses. Both subacute administration (1 hr before, and 1 and 24 hr after LPS injection) and acute administration (1 hr after LPS injection) of betaine also prevented LPS-induced memory impairment in the Y-maze test.</p> <p>Conclusions</p> <p>These data suggest that betaine has protective effects against LPS-induced memory impairment and that prevention of LPS-induced changes in GAT2 mRNA expression is crucial to this ameliorating effect.</p
