    Microbiological qualification of air, water and dialysate in a haemodialysis centre; a new focus on Legionella spp.

    Background and Objectives: The microbiological monitoring of the water used for haemodialysis is important especially for Legionella and non-fermentative bacteria since patients with end stage renal disease (ESRD) are suffering from deteriorated function of immune system. Materials and Methods: A total 50 water and dialysate samples were weekly collected over a period of 10 weeks from 5 sites. Total and faecal coliforms were determined by utilizing the most probable number (MPN) method. For isolation of Legionella, water samples were inoculated on a BCYE medium. DNA extraction was performed and was used to amplify 16S rRNA gene of Legionella species. Airborne bacteria were sampled using a single stage Andersen air sampler. Results: Out of total 50 water samples, 24 samples had bacterial contamination. The highest rate of Legionella contamination was observed in the storage tank (67 cfu/ml). Legionella was not isolated from the dialysate effluent samples. The highest rate of total bacterial count was related to the dialysate effluent and the maximum total count of coliforms was related to the reverse osmosis. The isolated bacteria were Gram-negative bacilli (mostly Pseudomonas isolates), Gram-positive cocci (mostly Micrococcus spp.) and Gram-positive bacilli (mostly Bacillus spp.). Six samples were contaminated with coliforms. No faecal coliform was isolated from the samples. Conclusion: These results indicated that dialysis machine is an important source of contaminations such as Staphylococcus, Pseudomonas and Legionella. Therefore an efficient prevention program is needed to eliminate bacterial contamination of dialysis water system. Moreover, in haemodialysis centres, periodic surveillance programs for microbiological qualification can lead to a better planning for disinfection of haemodialysis water systems. © 2016, Tehran University of Medical Science.     Sapropterin Treatment Prevents Congenital Heart Defects Induced by Pregestational Diabetes Mellitus in Mice.

    Background Tetrahydrobiopterin is a cofactor of endothelial NO synthase ( eNOS ), which is critical to embryonic heart development. We aimed to study the effects of sapropterin (Kuvan), an orally active synthetic form of tetrahydrobiopterin on eNOS uncoupling and congenital heart defects ( CHD s) induced by pregestational diabetes mellitus in mice. Methods and Results Adult female mice were induced to pregestational diabetes mellitus by streptozotocin and bred with normal male mice to produce offspring. Pregnant mice were treated with sapropterin or vehicle during gestation. CHD s were identified by histological analysis. Cell proliferation, eNOS dimerization, and reactive oxygen species production were assessed in the fetal heart. Pregestational diabetes mellitus results in a spectrum of CHD s in their offspring. Oral treatment with sapropterin in the diabetic dams significantly decreased the incidence of CHD s from 59% to 27%, and major abnormalities, such as atrioventricular septal defect and double-outlet right ventricle, were absent in the sapropterin-treated group. Lineage tracing reveals that pregestational diabetes mellitus results in decreased commitment of second heart field progenitors to the outflow tract, endocardial cushions, and ventricular myocardium of the fetal heart. Notably, decreased cell proliferation and cardiac transcription factor expression induced by maternal diabetes mellitus were normalized with sapropterin treatment. Furthermore, sapropterin administration in the diabetic dams increased eNOS dimerization and lowered reactive oxygen species levels in the fetal heart. Conclusions Sapropterin treatment in the diabetic mothers improves eNOS coupling, increases cell proliferation, and prevents the development of CHD s in the offspring. Thus, sapropterin may have therapeutic potential in preventing CHD s in pregestational diabetes mellitus

    Protocol-dependence of middle cerebral artery dilation to modest hypercapnia.

    There is a need for improved understanding of how different cerebrovascular reactivity (CVR) protocols affect vascular cross-sectional area (CSA) to reduce error in CVR calculations when measures of vascular CSA are not feasible. In human participants, we delivered ∼±4 mm Hg end-tidal partial pressure of CO2 (PETCO2) relative to baseline through controlled delivery, and measured changes in middle cerebral artery (MCA) CSA (7 Tesla magnetic resonance imaging (MRI)), blood velocity (transcranial Doppler and Phase contrast MRI), and calculated CVR based on a 3-minute steady-state (+4 mm Hg PETCO2) and a ramp (-3 to +4 mm Hg of PETCO2). We observed that (1) the MCA did not dilate during the ramp protocol (slope for CSA across time P \u3e 0.05; R2 = 0.006), but did dilate by ∼7% during steady-state hypercapnia (P \u3c 0.05); and (2) MCA blood velocity CVR was not different between ramp and steady-state hypercapnia protocols (ramp: 3.8 ± 1.7 vs. steady-state: 4.0 ± 1.6 cm/s/mm Hg), although calculated MCA blood flow CVR was ∼40% greater during steady-state hypercapnia than during ramp (P \u3c 0.05) with the discrepancy due to MCA CSA changes during steady-state hypercapnia. We propose that a ramp model, across a delta of -3 to +4 mm Hg PETCO2, may provide an alternative approach to collecting CVR measures in young adults with transcranial Doppler when CSA measures are not feasible.     Survey on identification and abundance of fishes in the Chitgar Lake

    This study focused on population structure, biodiversity, and abundance of the Chitgar Lake fishes in October 2014. Based on the lake location fishes samples were collected at 34 stations in water body. In this study identified 8 family belong to 18 species comprised of Cyprinidae (11 species), Cichlidae (one species), Loricariidae (one species), Pangasiidae (one species), Serrasalmidae (one species), Poeciliidea (one species), Salmonidae (one species), and Scaridae (one species) were Identified. Among the fishes just one native fish from Capoeta bohsei species observed during the study. The dominant lake fishes were invasive species Hemiculter leucisculus, Alburnus hohenackeri, Carassius auratus, Carassius gibelio and Pseudorasbora parva. The highest abundance of fishes were H. leucisculus 62 % and A. hohenackeri 35 % by Seine net. Furthermore, those were dominant abundance (69 and 12 % respectively) in Gill net method. Finally in Cast net method A. hohenackeri 38 %, Pseudorasbora parva 21 % and H. leucisculus 17 % were dominated in the shore region. The lowest fishes abundance were P. hypophthalmus and Parrotfish. Overall more than 90 % of the fishes population were invasive and alien species. Thus, the present of invasive fishes and predator fishes luck in the Chitgar lake might be reason for ecosystem ruin, algae bloom, feed competitive, fish disease and trophy level rises

    Study of Todebin dam reservoir in Zanjan province in order to aquaculture possibility

    The Todebin dam reservoir has been constructed in central part of Abhar in the Zanjan province which has been studied with aquaculture purpose. This study was conducted on biotic and abiotic factors. The plankton, benthos and fishes were identified and the parasitology and bacteriology studies were performed on the lake fishes. Then the fisheries potential of the lake was calculated in respect of final fish farming amount and the fish releasing. The results showed the Bacillariophyta was dominated ِduring the study. the highest abundance of phytoplankton was between 2.1 and 12.5 million cells.l^-1 in the upper layer of lake as compared with the lower layer. The genus Cyclotella and Dinobryon belong to phytoplankton and Keratella and Polyathera belong to zooplankton were dominanted abundance. The mean biomass of benthos was varied from 5.5 to 29.5 g.m -2 and the Chironomidae had the most abundance. The ichtiology survey showed the presence of four cultured species and the Goldfish. The hydro-chemical results indicated no restriction for aquaculture activity. The oxygen average was more than 8 mg/l. The average of phosphate, nitrogen and hardness were measured 0.09±0.04, 2±0.8 and 237±94.5 mg/l., respectively. For the most hydrochemical factores, the values were significantly different between upper and bottom layers of the lake. The oxygen production and Chl-a amount were measured about 0.73 mg/l. and 7/03 µg/l., respectively. According to climatic restrictions, the cold fish culture is preferred than to culture of warm fishes. While the natural capacity of the lake was about 5 tones to warm fish culture, the fish production of trout was estimated about 40 tones.     Caspian trout (Salmo trutta caspius ) is one of the migratory fish species which lives and feeds in the Caspian Sea .This species distributed in south and western of the Caspian Sea . In order to rehabilitation of the Caspian trout in western area, the operation of the project started from the beginning of the fishing season of the 20112012. Specimens of the Caspian trout obtained from beach seine cooperatives and have been transferred to the fish farm in Asalem region .Totally, 44 specimens of the Caspian trout has been caught in two month period .Most of the fishes was from Kargan roud river . The mean length and weight of the brood fishes were 67.7 cm and 3122.7 g respectively .As at present Shilat uses just the broods of the Tonekaboon region for restocking of this species ,we recommend to use the broods of the western region separately for rehabilitation of the stocks of this region
