    New Approaches to Degenerate Interconversions of Organic Structures: 1. Levels of Structural Degeneracy and Computer-aided Search for Degenerate Rearrangements*

    The general methodology of systematic search for degenerate interconversions must be based on a certain hierarchy of degeneracy. The levels and sublevels of the proposed hierarchy are represented by topological and structural types and subtypes of degeneracy, which correspond to specific equations and to graphs identifying them. Graph operations that enable an efficient search for arbitrary types of organic reactions are formulated, and problems specific to generation of degenerate transformations are considered. In conclusion, results of computer-aided search (using the ARGENT–1 software) for new heteroderivatives and structural analogs of the Cope reaction are discussed. Many of the rearrangements thus found (especially [3,3]-shifts in cations and anions, which have not been studied to date) can be subjects of experimental investigations

    Membrane‐acting biomimetic peptoids against visceral leishmaniasis

    Visceral leishmaniasis (VL) is among the most neglected tropical diseases in the world. Drug cell permeability is essential for killing the intracellular residing parasites responsible for VL, making cell-permeating peptides a logical choice to address VL. Unfortunately, the limited biological stability of peptides restricts their usage. Sequence-specific oligo-N-substituted glycines (‘peptoids’) are a class of peptide mimics that offers an excellent alternative to peptides in terms of ease of synthesis and good biostability. We tested peptoids against the parasite Leishmania donovani in both forms, that is, intracellular amastigotes and promastigotes. N-alkyl hydrophobic chain addition (lipidation) and bromination of oligopeptoids yielded compounds with good antileishmanial activity against both forms, showing the promise of these antiparasitic peptoids as potential drug candidates to treat VL

    Pharmacological blockade of cannabinoid type II receptors and mesenchymal stem cell transplantation in a model of peripheral neuropathic pain

    Objective. To evaluate the anti-nociceptive and reparative effects of adipose-derived mesenchymal stem cells (ADMSCs) under the pharmacological blockade of cannabinoid CB2 receptors in a model of peripheral neuropathic pain.Material and methods. In 40 male Wistar rats, modeling of peripheral neuropathy (NP) was performed by excising a sciatic nerve. On day 7 of the study, ADMSCs (1 × 106 cells/kg) were transplanted into the area of sciatic nerve injury without additional influences or after administration of the CB2 receptor antagonist AM630, as well as after incubation with AM630. Within 90  days, nociceptive sensitivity was studied, as well as a detailed analysis of gait using CatWalk XT (Noldus, Netherlands). On day 21 and day 90, histostructure of the distal segment of the sciatic nerve was assessed.Results. Pharmacological blockade of CB2 receptors both on the ADMSCs and in the soft tissues surrounding the site of sciatic nerve injury led to a decrease in withdrawal threshold and withdrawal latency from day 28 of the study compared with the group of rats with NP and transplantation of ADMSCs only. Local injection of AM630 before transplantation of ADMSCs contributed to the development of  NP-induced gait disturbances and increase of the number of damaged nerve fibers in the distal segment of sciatic nerve. Transplantation of ADMSCs pretreated with  AM630 did not significantly affect the rate of recovery of gait parameters, and decreased the number of damaged nerve fibers by day 90 of study.Conclusion. Blockade of CB2 receptors, both on the membranes of MSCs and in the area of damage to the peripheral nerve, has a negative effect on the development of the anti-nociceptive and reparative effects of MSCs

    The article discusses the heavy and special weapons of the Russian army during the Caucasian war across the territory of Black Sea coastline. By heavy equipment is meant all kinds of artillery systems which were used in the fortresses at that time. The particular importance is given to the special weapons, which include the minefields (mines and fougasses). It is proved that the mines on the galvanic elements were used not only in the navy but also for the needs of the ground forces. The authors came to the conclusion that the technical superiority of the Russian army became one of the main reasons for defeat of the highlanders in the war
