PDGFA-associated protein 1 protects mature B lymphocytes from stress-induced cell death and promotes antibody gene diversification
The establishment of protective humoral immunity is dependent on the ability of mature B cells to undergo antibody gene diversification while adjusting to the physiological stressors induced by activation with the antigen. Mature B cells diversify their antibody genes by class switch recombination (CSR) and somatic hypermutation (SHM), which are both dependent on efficient induction of activation-induced cytidine deaminase (AID). Here, we identified PDGFA-associated protein 1 (Pdap1) as an essential regulator of cellular homeostasis in mature B cells. Pdap1 deficiency leads to sustained expression of the integrated stress response (ISR) effector activating transcription factor 4 (Atf4) and induction of the ISR transcriptional program, increased cell death, and defective AID expression. As a consequence, loss of Pdap1 reduces germinal center B cell formation and impairs CSR and SHM. Thus, Pdap1 protects mature B cells against chronic ISR activation and ensures efficient antibody diversification by promoting their survival and optimal function

Mangafodipir Protects against Hepatic Ischemia-Reperfusion Injury in Mice
Mangafodipir is a contrast agent used in magnetic resonance imaging that concentrates in the liver and displays pleiotropic antioxidant properties. Since reactive oxygen species are involved in ischemia-reperfusion damages, we hypothesized that the use of mangafodipir could prevent liver lesions in a mouse model of hepatic ischemia reperfusion injury. Mangafodipir (MnDPDP) was compared to ischemic preconditioning and intermittent inflow occlusion for the prevention of hepatic ischemia-reperfusion injury in the mouse.Mice were subjected to 70% hepatic ischemia (continuous ischemia) for 90 min. Thirty minutes before the ischemic period, either mangafodipir (10 mg/kg) or saline was injected intraperitoneally. Those experimental groups were compared with one group of mice preconditioned by 10 minutes' ischemia followed by 15 minutes' reperfusion, and one group with intermittent inflow occlusion. Hepatic ischemia-reperfusion injury was evaluated by measurement of serum levels of aspartate aminotransferase (ASAT) activity, histologic analysis of the livers, and determination of hepatocyte apoptosis (cytochrome c release, caspase 3 activity). The effect of mangafodipir on the survival rate of mice was studied in a model of total hepatic ischemia.<0.01), and by higher rates of survival in treated than in untreated animals (P<0.001). The level of protection by mangafodipir was similar to that observed following intermittent inflow occlusion and higher than after ischemic preconditioning.Mangafodipir is a potential new preventive treatment for hepatic ischemia-reperfusion injury
Probiotic Sonicates Selectively Induce Mucosal Immune Cells Apoptosis through Ceramide Generation via Neutral Sphingomyelinase
This is an open-access article distributed under the terms of the Creative Commons Attribution License.-- et al.[Background]: Probiotics appear to be beneficial in inflammatory bowel disease, but their mechanism of action is incompletely understood. We investigated whether probiotic-derived sphingomyelinase mediates this beneficial effect.
[Methodology/Principal Findings]: Neutral sphingomyelinase (NSMase) activity was measured in sonicates of the probiotic L. brevis (LB) and S. thermophilus (ST) and the non-probiotic E. coli (EC) and E. faecalis (EF). Lamina propria mononuclear cells (LPMC) were obtained from patients with Crohn's disease (CD) and Ulcerative Colitis (UC), and peripheral blood mononuclear cells (PBMC) from healthy volunteers, analysing LPMC and PBMC apoptosis susceptibility, reactive oxygen species (ROS) generation and JNK activation. In some experiments, sonicates were preincubated with GSH or GW4869, a specific NSMase inhibitor. NSMase activity of LB and ST was 10-fold that of EC and EF sonicates. LB and ST sonicates induced significantly more apoptosis of CD and UC than control LPMC, whereas EC and EF sonicates failed to induce apoptosis. Pre-stimulation with anti-CD3/CD28 induced a significant and time-dependent increase in LB-induced apoptosis of LPMC and PBMC. Exposure to LB sonicates resulted in JNK activation and ROS production by LPMC. NSMase activity of LB sonicates was completely abrogated by GW4869, causing a dose-dependent reduction of LB-induced apoptosis. LB and ST selectively induced immune cell apoptosis, an effect dependent on the degree of cell activation and mediated by bacterial NSMase.

Defective metabolic programming impairs early neuronal morphogenesis in neural cultures and an organoid model of Leigh syndrome
Leigh syndrome (LS) is a severe manifestation of mitochondrial disease in children and is currently incurable. The lack of effective models hampers our understanding of the mechanisms underlying the neuronal pathology of LS. Using patient-derived induced pluripotent stem cells and CRISPR/Cas9 engineering, we developed a human model of LS caused by mutations in the complex IV assembly gene SURF1. Single-cell RNA-sequencing and multi-omics analysis revealed compromised neuronal morphogenesis in mutant neural cultures and brain organoids. The defects emerged at the level of neural progenitor cells (NPCs), which retained a glycolytic proliferative state that failed to instruct neuronal morphogenesis. LS NPCs carrying mutations in the complex I gene NDUFS4 recapitulated morphogenesis defects. SURF1 gene augmentation and PGC1A induction via bezafibrate treatment supported the metabolic programming of LSÂ NPCs, leading to restored neuronal morphogenesis. Our findings provide mechanistic insights and suggest potential interventional strategies for a rare mitochondrial disease
Factors influencing postfire dynamics of Sardinian and Dartford Warblers in Mediterranean shrublands
We studied the post-fire dynamics of two Mediterranean warblers (the Sardinian (Sylvia melanocephala) and the Dartford Warbler (Sylvia undata)) to determine whether these were influenced by local habitat recovery or by external population trends unrelated to the occurrence of fire . We used the point-count method to survey warblers from the first to the sixth breeding seasons after fire in burnt and unburnt control zones of the Iberian Peninsula. Both species increased similarly in the burnt zone until the fourth year after fire, but the Dartford Warbler increased faster than the Sardinian Warbler from then on. Six years after fire, the abundance of the Dartford Warbler was even higher in the burnt zone than in the control, where the population of this species remained constant during the study period . In contrast, a parallel increase in the populations of Sardinian Warbler in both zones was observed, thus suggesting that processes other than fire were profoundly involved in the dynamics of this species in the burnt zone. Our results indicate that the post-fire dynamics of these two species greatly differ, the Dartford Warbler being highly influenced by local habitat changes in the burnt zone and the Sardinian Warbler by population trend on a larger scale than the disturbed habitat
Efectos de la contaminación atmosférica procedente de las emisiones de la central térmica de CERCS (Catalunya, Noreste España) sobre la biodiversidad de artrópodos
The abundance and diversity of arthropods collected in Cercs, where there is a coal-fired power plant, and a control area in St. Jaume de Frontanyà were compared. The two zones studied are similar in relief, vegetation and climatology. For one year, from March 1995 to February 1996, trapped animals were collected, and taxonomic determination was performed. Results showed that number of collected arthropods were 1.7 fold higher in the polluted area than in St. Jaume de Frontanya with a special presence of the orders of C.oleoptera, Orthoptera, Julida. Opilionida and Isopoda, while Colembola was the most represented order in the non-polluted zone. Nevertheless, the abundance was significantly higher in Cercs only in summer, and in St. Jaume de Frontanyà in winter. According to their diet statistical differences between arthropod predators seasonal distribution in both zones were reported. Using Geocoding and Topic Extraction to Make Sense of Comments on Social Network Pages of Local Government Agencies

Text mining, crowdsourcing and data visualization are some approaches that allow the information and knowledge extraction from texts in comment formats, exchanged between citizens in social networks. This movement can be indirectly used as a bias for popular participation, gaining prominence in the construction of smart cities. The objective of this work is to present a method that geocodes citizens’ comments made on posts in Social Network Pages of Local Government Agencies, and extracts the most frequent topics present in these comments. In order to validate our method, we implemented a web system that implements the steps of the proposed method, and conducted a case study. The tool, and consequently the steps of the presented method, was evaluated by four software developers, which indicated that the tool was easy to use, new knowledge could be extracted from it, and some interesting improvements were pointed out by them
