    The preparation of BP single crystals by high pressure flux method

    Single crystals of BP, a III-V compound semiconductor, were obtained by the high pressure flux method. Cu3P and Ni12P5 powders were used as the flux, and mixed with BP powder. Two kinds of mixtures were prepared: (1) 1.8g (BP) + 35 G (Cu3P) and (2) 1.7 g (BP) + 25 g (Ni12P5). They were compressed into pellets, heated at 1300 C for 24 h in an induction furnace under a pressure of 1 MPa using Ar-P2 gas, and slowly cooled to room temperature. In case (1), BP single crystals grew along the (III) plane, and in case (2) they grew as an aggregate of crystallites. The cathodoluminescence spectra of the synthetic BP crystals showed peaks near 680 nm (1.82 eV) for case (1), and 500 nm (2.47 eV) for case (2).     Water is an essential nutrient and is required for growth, metabolism, lactation, and reproduction. However, water requirements for beef cattle have not been studied extensively since the 1950’s. The genetic potential for beef cattle has changed considerably since then. With the change in genetics, beef producers have observed increases in average daily gain and final body weight. The need for updated water requirements for beef cattle has also been exacerbated by global climate variability, drought conditions, and the need for efficient use of water resources. Given the increased productivity of cattle today relative to those of decades ago, increased water requirements are almost certain. Further research must be conducted to determine how those requirements have changed

    Three-Dimensional Transgenic Cell Models to Quantify Space Genotoxic Effects

    In this paper we describe a three-dimensional, multicellular tissue-equivalent model, produced in NASA-designed, rotating wall bioreactors using mammalian cells engineered for genomic containment of mUltiple copies of defined target genes for genotoxic assessment. The Rat 2(lambda) fibroblasts (Stratagene, Inc.) were genetically engineered to contain high-density target genes for mutagenesis. Stable three-dimensional, multicellular spheroids were formed when human mammary epithelial cells and Rat 2(lambda) fibroblasts were cocultured on Cytodex 3 Beads in a rotating wall bioreactor. The utility of this spheroidal model for genotoxic assessment was indicated by a linear dose response curve and by results of gene sequence analysis of mutant clones from 400micron diameter spheroids following low-dose, high-energy, neon radiation exposur

    Comparison of Genotoxic Damage in Monolayer Cell Cultures and Three-Dimensional Tissue-Like Cell Assemblies

    Assessing the biological risks associated with exposure to the high-energy charged particles encountered in space is essential for the success of long-term space exploration. Although prokaryotic and eukaryotic cell models developed in our laboratory and others have advanced our understanding of many aspects of genotoxicity, in vitro models are needed to assess the risk to humans from space radiation insults. Such models must be representative of the cellular interactions present in tissues and capable of quantifying I genotoxic damage. Toward this overall goal, the objectives of this study were to examine the effect of the localized microenvironment of cells, cultured as either 2-dimensional (2D) monolayers or 3-dimensional (3D) aggregates, on the rate and type of genotoxic damage resulting from exposure to iron charged particles, a significant portion of space radiation. We used rodent transgenic cell lines containing 50-70 copies of a LacI transgene to provide the enhanced sensitivity required to quantify mutational frequency and type in the 1,100-bp LacI target as well as assessment of DNA,damage to the entire 45-kbp construct. Cultured cells were exposed to high-ener~ir on charged particles at Brookhaven National Laboratory s Alternating Gradient Synchrotron facility for a total dose of 0, 0.1, 0.25,0.5, 1.0, or 2.0 Gy and allowed to recover for 0, 1, or 7 days, after which mutational type and frequency were evaluated. The mutational frequency was found to be higher in 3D samples than in 2D samples at all radiation doses. Mutational frequency also was higher at 7 days after irradiation than immediately after exposure. DNA sequencing of the mutant targets revealed that deletional mutations contributed an increasingly high percentage (up to 27%) of all mutations in cells as the dose was increased from 0.5 to 2 Gy. Several mutants also showed large and complex deletions in multiple locations within the Lac1 target. However, no differences in mutational type were found between the 2D and the 3D samples. These 3D tissue-like model systems can reduce the uncertainty involved in extrapolating risk between in vitro cellular and in vivo models

    How possible is the development of an operational psychometric method to assess the presence of the 5-HTTLPR s allele? Equivocal preliminary findings

    <p>Abstract</p> <p>Objective</p> <p>The s allele of the 5-hydroxytryptamine transporter-linked promoter region (5-HTTLPR) polymorphism of the serotonin transporter gene has been found to be associated with neuroticism-related traits, affective temperaments and response to selective serotonin reuptake inhibitor (SSRI) treatment. The aim of the current study was to develop a psychometric tool that could at least partially substitute for laboratory testing and could predict the presence of the s allele.</p> <p>Methods</p> <p>The study included 138 women of Caucasian origin, mean 32.20 ± 1.02 years old. All subjects completed the Hungarian standardised version of the Temperament Evaluation of the Memphis, Pisa, Paris, and San Diego Autoquestionnaire (TEMPS-A) instrument and were genotyped for 5-HTTLPR using PCR. The statistical analysis included the calculation of the Index of Discrimination (D), Discriminant Function Analysis, creation of scales on the basis of the above and then item analysis and calculation of sensitivity and specificity.</p> <p>Results</p> <p>Four indices were eventually developed, but their psychometric properties were relatively poor and their joint application did not improve the outcome.</p> <p>Conclusions</p> <p>We could not create a scale that predicts the 5-HTTLPR genotype with sufficient sensitivity and specificity, therefore we could not substitute a psychometric scale for laboratory genetic testing in predicting genotype, and also possibly affective disorder characterisation and treatment.</p

    Estimating the Effects of Weather, Dry Matter Intake, and Body Weight on Daily Water Intake in Weaned Calves

    Objective The purpose of this study was to study the effect of weather, dry matter intake, and body weight on the water requirements of weaned calves and estimating the requirements in a model.Study Description Weaned steers (n=48) were selected to study the effects of the weather, body weight, and dry matter intake on water intake in the winter (n=24) and summer (n=24) months. Calves were provided with ad libitum access to feed and water at the SDSU Cow-Calf Education and Research Facility (CCERF) and measured utilizing an automated feed and water system (Insentec, The Hague, Netherlands). Temperature, humidity, precipitation, wind speed, solar radiation, and air pressure were recorded at a Mesonet automated weather station in Brookings, SD (located 2.4 miles from the SDSU CCERF). Effects of climate data, body weight, and daily dry matter intake on daily water intake will be analyzed utilizing a mixed-effects model

    Sarcoidosis presenting with glazy mucoid sputum and dyspnea:a case report

    BACKGROUND: Patients with pulmonary sarcoidosis commonly present with a dry cough; a productive cough suggests a complicating airway infection or an alternative diagnosis such as tuberculosis or bronchiectasis. CASE PRESENTATION: A 36-year-old European (Frisian) woman recently diagnosed with pulmonary sarcoidosis presented with debilitating exertional dyspnea and cough productive of glazy mucoid sputum. Several different attempts including video-assisted thoracoscopic biopsies failed to reach a second or alternative diagnosis including an infectious, autoimmune or collagen-vascular condition. She responded to steroids but with poor tolerance to this treatment, which could not be tapered. After she was started on anti-tumor necrosis factor alpha (TNF-α) therapy with infliximab, 200 mg at three-monthly intervals, she has been fine for well over a decade. CONCLUSIONS: In this patient with sarcoidosis who had a productive cough accompanied by fever, an extensive workup and prolonged follow-up, an alternative or second diagnosis could be ruled out; we therefore conclude that this highly unusual presentation is part of the clinical spectrum of sarcoidosis

    IMPLEMENTASI KOMUNIKASI ORGANISASI BERBASIS KEARIFAN LOKAL DALAM PELAYANAN PUBLIK

    Abstrak. Penyelenggaraan komunikasi organisasi yang mengadopsi kearifan lokal dalam pelayanan publik masih sangat minim. Kearifan lokal menjadi salah satu bahasan yang menarik jika diadopsi dalam proses interaksi komunikasi organisasi. Meskipun dalam implementasinya memang bukan sesuatu yang mudah dilakukan. Dalam praktiknya hal tersebut dapat meningkatkan kinerja organisasi baik dalam memaksimalkan budaya organisasi maupun dalam meningkatkan pelayanan publik. Tujuan penelitian ini untuk menganalisis implementasi komunikasi organisasi berbasis kearifan lokal dalam pelayanan publik pada Dinas Penanaman Modal dan Pelayanan Terpadu Satu Pintu (DPMPTSP) Kabupaten Banggai Laut. Penelitian ini menggunakan pendekatan kualitatif dengan pendekatan etnografi komunikasi. Penelitian ini menggunakan data primer dan data sekunder melalui observasi, wawancara mendalam, dan dokumentasi. Hasil penelitian ini menunjukkan bahwa (1) empat bentuk kearifan lokal Banggai, yaitu moloyos, molios, maamis, dan monondok yang dimplementasikan dalam budaya organisasi DPMPTSP Kabupaten Banggai Laut merupakan turunan dari falsafah tuu-tuu, (2) dalam implementasinya, komunikasi organisasi berbasis kearifan lokal membentuk budaya organisasi yang lebih baik dan memaksimalkan kerja pelayanan publik sebagaimana hal itu dirasakan langsung oleh masyarakat yang menggunakan jasa layanan pada DPMPTSP Kabupaten Banggai Laut, dan (3) penyebaran pesan yang sering dilakukan adalah penyebaran secara serentak. Adapun arah arus informasi yang lebih banyak digunakan adalah komunikasi ke bawah dan komunikasi ke atas. Kemudian terdapat pula komunikasi horizontal dan komunikasi lintas saluran. In public service, the implementation of organizational communication that adopts local wisdom is still very limited. Although, local wisdom becomes one of the most interesting topics if it is adopted in the process of organizational communication. The implementation of local wisdom values is not something that is easy to do. However, It can actually enhance organizational performance by maximizing organizational culture and improving the public service process. This study investigates the implementation of organizational communication based on local wisdom in public service at the Investment and One Stop Integrated Services (DPMPTSP) of Banggai Laut Regency. This study uses qualitative research methods through an ethnographic communication approach by collecting data through observation, interviews, and documentation studies. The results of this study demonstrate that (1) the organizational culture of DPMPTSP of Banggai Laut Regency, which adopted four forms of Banggai local wisdom, including moloyos, molios, maamis, and monondok, is derived from the philosophy of Tuu-tuu. (2) Implementation of Organizational communication based on local wisdom can create a better organizational culture and simulate the work of public services as it is perceived by the clients of DPMPTSP Banggai Laut Regency. (3) The dissemination of messages that is often carried out is simultaneous dissemination. The direction of information flow that is more widely used is downward communication rather than upward communication. Then, there is also horizontal communication and diagonal communication.
