    Potential Response of Soil-Borne Fungal Pathogens Affecting Crops to a Scenario of Climate Change in Europe

    A study was carried out on the potential response of soil-borne pathogens causing crop yield losses under a climate change scenario in Europe. A controlled chamber set of experiments was carried out to quantify pathogen response to temperature using pure colonies of three soil-borne fungi, representative of low (Fusarium nivale), medium-high (Athelia rolfsii) and high (Macrophomina phaseolina) temperature requirements. A generic model to simulate fungal growth response to temperature based on these experiments was developed and linked to a soil temperature model component, and to components to simulate soil water content accounting for crop water uptake of potential hosts. Pathogens relative growth was simulated over Europe using the IPCC A1B emission scenario as realization of the Hadley-CM3 global climate model, available from the European Commission and processed for use with biophysical models. The simulations resulting from using the time span centred on 2030 were compared to the baseline, centred on the year 2000, using a sample of 30 years of daily weather. The general trend of soil-borne pathogens response to the scenario of climate change is a relative increase in growth in colder areas of Europe, as a function of their temperature requirements. Projections of F. nivale in the future indicate a relative increase of this winter pathogen of wheat in Northern European countries. A. rolfsii and M. phaseolina, two soil-borne pathogens typical of warmer agricultural areas, could find more favourable conditions in areas of the Central Europe, but they differentiated in Southern Europe where A. rolfsii resulted affected by summer soil temperatures above optimum

    Degeneration and regeneration of peripheral nerves: role of thrombin and its receptor PAR-1

    The peripheral nervous system has a striking regeneration potential and after damage extensive changes in the differentiation state both of the injured neurons and of the Schwann cells are observed. Schwann cells, in particular, undergo a large scale change in gene expression becoming able to support axonal regeneration. Nerve injury is generally associated to inflammation and activation of the coagulation cascade. Thrombin acts as a polyfunctional signalling molecule exerting its physiological function through soluble target proteins and G-protein-coupled receptors, the protease-activated receptors (PARs) [1]. Recently, we have demonstrated that the activation of the main thrombin receptor, PAR-1, in Schwann cells favours their regenerative potential determining the release of factors which promote axonal regrowth [2]. The pro-regenerative potential of thrombin seems to be exerted in a narrow range of concentrations (pM-nM range). In fact, our preliminary data indicate that high levels of thrombin in the micromolar range slow down Schwann cell proliferation and induce cell death. On the contrary, PAR-1 activating peptides mimic the pro-survival but not the pro-apoptotic effects of thrombin. Controlling thrombin concentration may preserve neuronal health during nerve injury and represent a novel target for pharmacologic therapies

    The genomic footprint of social stratification in admixing American populations.

    Cultural and socioeconomic differences stratify human societies and shape their genetic structure beyond the sole effect of geography. Despite mating being limited by sociocultural stratification, most demographic models in population genetics often assume random mating. Taking advantage of the correlation between sociocultural stratification and the proportion of genetic ancestry in admixed populations, we sought to infer the former process in the Americas. To this aim, we define a mating model where the individual proportions of the genome inherited from Native American, European, and sub-Saharan African ancestral populations constrain the mating probabilities through ancestry-related assortative mating and sex bias parameters. We simulate a wide range of admixture scenarios under this model. Then, we train a deep neural network and retrieve good performance in predicting mating parameters from genomic data. Our results show how population stratification, shaped by socially constructed racial and gender hierarchies, has constrained the admixture processes in the Americas since the European colonization and the subsequent Atlantic slave trade

    Involvement of pro-inflammatory cytokines and growth factors in the pathogenesis of Dupuytren's contracture: a novel target for a possible future therapeutic strategy?

    Dupuytren's contracture (DC) is a benign fibro-proliferative disease of the hand causing fibrotic nodules and fascial cords which determine debilitating contracture and deformities of fingers and hands. The present study was designed to characterize pro-inflammatory cytokines and growth factors involved in the pathogenesis, progression and recurrence of this disease, in order to find novel targets for alternative therapies and strategies in controlling DC. The expression of pro-inflammatory cytokines and of growth factors was detected by immunohistochemistry in fibrotic nodules and normal palmar fascia resected respectively from patients affected by DC and carpal tunnel syndrome (CTS; as negative controls). Reverse transcription (RT)-PCR analysis and immunofluorescence were performed to quantify the expression of transforming growth factor (TGF)-β1, interleukin (IL)-1β and vascular endothelial growth factor (VEGF) by primary cultures of myofibroblasts and fibroblasts isolated from Dupuytren's nodules. Histological analysis showed high cellularity and high proliferation rate in Dupuytren's tissue, together with the presence of myofibroblastic isotypes; immunohistochemical staining for macrophages was completely negative. In addition, a strong expression of TGF-β1, IL-1β and VEGF was evident in the extracellular matrix and in the cytoplasm of fibroblasts and myofibroblasts in Dupuytren's nodular tissues, as compared with control tissues. These results were confirmed by RT-PCR and by immunofluorescence in pathological and normal primary cell cultures. These preliminary observations suggest that TGF-β1, IL-1β and VEGF may be considered potential therapeutic targets in the treatment of Dupuytren's disease (DD)

    Effects of the mean particle size in the deflagration index estimation for cornstarch dust

    The National Fire Protection Association (NFPA) defines the dust explosions as a “credible risk”. Hence, to meet the challenge to prevent and protect from the catastrophic effects of these phenomena, it is fundamental to know what are the characteristics and the burning conditions regarding the combustible dusts that could have an effect on the explosion violence. The KSt, also known as deflagration index, is one of the relevant parameters in dust explosions, together with the maximum explosion overpressure generated in the test chamber, the minimumignition energy and so on. In particular, the deflagration index measures the relative explosion severity and it is used in the design of the dust venting protection equipment. However, one of the criticalities of such a parameter is that is strongly affected by the particle mean diameter. Hence, in the following, it will be preliminary presented the validation of a single particle spherical model able to predict the variation of the deflagration index with the increasing mean particle size knowing just one experimental KSt value

    Impairment of the autophagic flux in astrocytes intoxicated by trimethyltin

    Autophagy is a lysosomal catabolic route for protein aggregates and damaged organelles which in different stress conditions, such as starvation, generally improves cell survival. An impairment of this degradation pathway has been reported to occur in many neurodegenerative processes. Trimethyltin (TMT) is a potent neurotoxin present as an environmental contaminant causing tremors, seizures and learning impairment in intoxicated subjects. The present data show that in rat primary astrocytes autophagic vesicles (AVs) appeared after few hours of TMT treatment. The analysis of the autophagic flux in TMT-treated astrocytes was consistent with a block of the late stages of autophagy and was accompanied by a progressive accumulation of the microtubule associated protein light chain 3 (LC3) and of p62/SQSTM1. Interestingly, an increased immunoreactivity for p62/SQSTM1 was also observed in hippocampal astrocytes detected in brain slices of TMT-intoxicated rats. The time-lapse recordings of AVs in EGFP-mCherry-LC3B transfected astrocytes demonstrated a reduced mobility of autophagosomes after TMT exposure respect to control cells. The observed block of the autophagic flux cannot be overcome by known autophagy inducers such as rapamycin or 0.5mM lithium. Although ineffective when used at 0.5mM, lithium at higher concentrations (2mM) was able to protect astrocyte cultures from TMT toxicity. This effect correlated well with its ability to determine the phosphorylation/inactivation of glycogen kinase synthase-3β (GSK-3β)

    The Ph.D. Thesis "Wege zu einer neuen Ph\ue4nomenologie: Landgrebe, Fink und Pato\u10dka im Dialog\u201d (Toward a new Phenomenology: Landgrebe, Fink and Pato\u10dka in Dialogue) focuses on the reflections of the three philosophers about phenomenology through the personal correspondence between Fink and Pato\u10dka (1933-1977) and between Pato\u10dka and Landgrebe (1940-1976). Their discussions identify some critical points inside Husserl\u2019s thought \u2013 especially concerning the phenomenological reduction \u2013 and underline the need for phenomenology to move forward to continue his own history. From the comparison between the elements that come to light from the correspondences and the main writings of the 1930s of these philosophers, are emerging the three ways to proceed on the path of Edmund Husserl. Ludwig Landgrebe develops a new method \u2013 \u201cMethode der Leitf\ue4den\u201c \u2013 which enlightens the concept of experience (Erlebnis) as a keystone of the man\u2019s self-understanding; Eugen Fink with his Phenomenology of Phenomenology radicalizes the doubt of phenomenological principles which leads to a world\u2019s constitutive investigation; finally Jan Pato\u10dka theorizes an Asubjective Phenomenology which through the schematization of intentional process reaches the phenomenological understanding of existence, anticipating a historical development of the ontology of subjectivity. The three different attempts of revision have in common a non-dogmatic recover of metaphysics which allows an improvement of phenomenological philosophy without betraying its peculiar methodology to investigate the world\u2019s phenomenon
